    Management Skills for the Contemporary College President: A Critical Review

    This review of the literature related to the contemporary college president attempted to describe changes to the presidential role during the past 30 years. In addition to describing changes to the role, the review explores the personal, organizational, interpersonal, and leadership skills required for leading a contemporary university. The findings particularly note that with the changing demands for accountability in higher education, presidents are being forced to spend more of their time and be more focused on issues of finance, law, and public interactions. These issues tend to collide in discussions of public policy and within the framework of state legislative proposals. Continued professional development for college presidents is recommended, especially in terms of aligning work place priorities with the institution’s mission

    This Article examines some of the legal issues that permeate the performance review process in higher education and offers some suggestions for improving faculty evaluations. As will be apparent, the courts have generally given institutions of higher education great latitude in devising and administering performance review programs. However, colleges and universities do not have carte blanche with their performance reviews and there are critical constraints on what institutions of higher education can do. These constraints are rooted in principles of fundamental fairness and due process, as well as constitutional and statutory requirements

    Structure and Functional Responsibilities of Graduate Schools: An Organizational Analysis

    The contemporary graduate ‘school’ is facing a number of significant challenges. In addition to the fundamental question of its role as a shared service provider, graduate education units are exploring ways that they can demonstrate a value-added component to the graduate school experience. These activities include offering graduate certificates about how to be a faculty member, how to best teach and mentor graduate students, and offering undergraduate courses on how to get into graduate school. The current study explored 84 graduate ‘school’ or equivalent unit organizational charts, noting major differences between those with research administration attached to them and those without such responsibilities. Findings also included the identification of small administrative staffs and growing innovation to serve graduate student needs, such as providing mental health services and food pantries

    Short tandem repeat profiling: part of an overall strategy for reducing the frequency of cell misidentification

    The role of cell authentication in biomedical science has received considerable attention, especially within the past decade. This quality control attribute is now beginning to be given the emphasis it deserves by granting agencies and by scientific journals. Short tandem repeat (STR) profiling, one of a few DNA profiling technologies now available, is being proposed for routine identification (authentication) of human cell lines, stem cells, and tissues. The advantage of this technique over methods such as isoenzyme analysis, karyotyping, human leukocyte antigen typing, etc., is that STR profiling can establish identity to the individual level, provided that the appropriate number and types of loci are evaluated. To best employ this technology, a standardized protocol and a data-driven, quality-controlled, and publically searchable database will be necessary. This public STR database (currently under development) will enable investigators to rapidly authenticate human-based cultures to the individual from whom the cells were sourced. Use of similar approaches for non-human animal cells will require developing other suitable loci sets. While implementing STR analysis on a more routine basis should significantly reduce the frequency of cell misidentification, additional technologies may be needed as part of an overall authentication paradigm. For instance, isoenzyme analysis, PCR-based DNA amplification, and sequence-based barcoding methods enable rapid confirmation of a cell line’s species of origin while screening against cross-contaminations, especially when the cells present are not recognized by the species-specific STR method. Karyotyping may also be needed as a supporting tool during establishment of an STR database. Finally, good cell culture practices must always remain a major component of any effort to reduce the frequency of cell misidentification

    Ischemic preconditioning improves maximal performance in humans

    Repeated episodes of ischemia followed by reperfusion, commonly referred to as ischemic preconditioning (IPC), represent an endogenous protective mechanism that delays cell injury. IPC also increases blood flow and improves endothelial function. We hypothesize that IPC will improve physical exercise performance and maximal oxygen consumption. The purpose of the study was to examine the effect of ischemic preconditioning in leg skeletal muscles on cycling exercise performance in healthy individuals. Fifteen healthy, well-trained subjects performed two incremental maximal exercise tests on a bicycle ergometer. Power output, oxygen consumption, ventilation, respiratory quotient, and heart rate were measured continuously. Blood pressure and blood lactate were measured before and after the test. One exercise test was performed after the application of ischemic preconditioning, using a protocol of three series of 5-min ischemia at both legs with resting periods of 5 min in between. The other maximal cycling test served as a control. Tests were conducted in counterbalanced order, at least 1 week apart, at the same time of the day. The repeated ischemic periods significantly increased maximal oxygen consumption from 56.8 to 58.4 ml/min per kg (P = 0.003). Maximal power output increased significantly from 366 to 372 W (P = 0.05). Ischemic preconditioning had no effect on ventilation, respiratory quotient, maximal heart rate, blood pressure or on blood lactate. Repeated short-term leg ischemia prior to an incremental bicycle exercise test improves maximal oxygen consumption by 3% and power output by 1.6%. This protocol, which is suggested to mimic the effects of ischemic preconditioning, may have important implications for exercise performance

    Conditional Gene Knockout in Human Cells with Inducible CRISPR/Cas9.

    The advent of the easily programmable and efficient CRISPR/Cas9 nuclease system has revolutionized genetic engineering. While conventional gene knockout experiments using CRISPR/Cas9 are very valuable, these are not well suited to study stage-specific gene function in dynamic situations such as development or disease. Here we describe a CRISPR/Cas9-based OPTimized inducible gene KnockOut method (OPTiKO) for conditional loss-of-function studies in human cells. This approach relies on an improved tetracycline-inducible system for conditional expression of single guide RNAs (sgRNAs) that drive Cas9 activity. In order to ensure homogeneous and stable expression, the necessary transgenes are expressed following rapid and efficient single-step genetic engineering of the AAVS1 genomic safe harbor. When implemented in human pluripotent stem cells (hPSCs), the approach can be then efficiently applied to virtually any hPSC-derived human cell type at various stages of development or disease
