    Potencial enzimático e toxigênico de fungos isolados de grãos de café

    The presence of some species of filamentous fungi in coffee beans may indicate reduced quality and risks of mycotoxins. Moreover, other species may be bioprotective of the bean integrity, indicators of environmental changes and informative. The objective of this study was to evaluate the enzymatic activity and toxigenic potential of filamentous fungi isolated from 12 samples of coffee beans (11 samples of Coffea arabica and 1 sample of Coffea canephora). 182 fungi were isolated and identified belonging to two genera: Aspergillus and Penicillium. Of the 138 fungi from the genus Aspergillus belonging to the Section Nigri and Section Circumdati tested, 28.3 % were producers of ochratoxin A, particularly for the species A. ochraceus and A. ostianus. Of the 14 isolates of Aspergillus flavus tested, 78.6 % were producers of aflatoxin B1 and B2. Aspergillus versicolor , Cladosporium cladosporioides , Penicillium roqueforti and Penicillium sp . showed an enzymatic index greater than 2 (IE> 2) for polygalacturonase and Penicillium funiculosum, Penicillium and Aspergillus sclerotiorum aurantiogriseum showed pectate lyase activity above 2 ( IE> 2). The isolates of Penicillium brevicompactum showed potential pectinolitica for the two enzymes tested. These results demonstrate that coffee beans can be an important source of fungi with biotechnological potential and potentially toxigenic fungi have limited enzyme capacity of degrading pectin -rich substrates such as mucilage and produce mycotoxins.A presença de algumas espécies de fungos filamentosos em grãos de café pode indicar redução da qualidade e riscos de micotoxinas. Por outro lado, outras espécies podem ser bioprotetoras da integridade dos grãos, indicadores de alterações ambientais e indicativo. Objetivou-se, neste estudo, avaliar a atividade enzimática e o potencial toxigênico de fungos filamentosos isolados de 12 amostras de grãos de café (11 amostras de Coffea arabica e uma amostra de Coffea canephora).Foram isolados e identificados 182 fungos pertencentes a dois gêneros: Aspergillus e Penicillium. Dos 138 fungos do gênero Aspergillus pertencentes à Seção Circumdati e Seção Nigri testados, 28,3% foram produtores de ocratoxina A, com destaque para as espécies A. ochraceus e A. ostianus. Dos 14 isolados de A. flavus testados, 78,6% foram produtores de aflatoxina B1 eB2. A. versicolor, Cladosporium cladosporioides, P. roqueforti e Penicillium sp. apresentaram índice enzimático superior a 2(IE > 2) para poligalacturonase e P. funiculosum, P. aurantiogriseum e Aspergillus sclerotiorum apresentaram atividade pectato liase acima de 2 (IE > 2). Os isolados de P. brevicompactum apresentaram potencial pectinolítico para as duas enzimas testadas.Esses resultados demonstram que os grãos de café podem ser importante fonte de fungos com potencial biotecnológico e que os fungos potencialmente toxigênicos apresentam limitada capacidade enzimática de degradar substratos ricos em pectina como a mucilagem e produzir micotoxinas

    Immunophenotypic characterization of lymphocytic infiltration in canine melanocytic tumors / Caracterização imunofenotípica da infiltração linfocítica em tumores melanocíticos caninos

    Melanocytic tumors correspond to approximately for 4 to 7% of tumors in dogs and up to 7% of malignant tumours. Melanomas generally exhibit aggressive biological behavior. In humans, abundant tumor-infiltrating lymphocytes (TIL) can be considered a good prognostic factor. Research on TIL in canine melanomas is scarce, and to date, there are no studies to verify its association with established prognostic factors. Our study aimed to evaluate the lymphocyte population in canine melanocytic tumors using immunohistochemical markers, and to relate this to pre-established clinicopathological prognostic variables and cell proliferation index (Ki-67). An exploratory cross-sectional study was conducted with 21 cases in 19 dogs, of which 71.5% were melanotic melanomas, 19% were melanocytomas and 9.5% were amelanotic melanomas. Ki-67 expression was elevated in amelanotic melanomas, indicating aggressive tumor behavior. We detected lymphocytes T and B through CD3 and CD20 markers, respectively, and found that 86.7% of the melanotic melanomas were positive for CD3, whereas 73.3% were negative for CD20. TIL showed a strong association with malignant tumors, as well as a correlation with other pre-established prognostic factors, such as necrosis, ulceration, and nuclear atypia. TIL need to be further investigated to verify its inclusion as a prognostic factor for canine melanomas

    Mapping QTL for protein and oil content in soybean

    O objetivo deste trabalho foi detectar e mapear locos de caracteres quantitativos (QTL) que afetam os conteúdos de proteína e óleo em soja (Glycine max L. Merr.). Plantas F2, derivadas do cruzamento entre a linhagem CS3032PTA276 e a variedade UFVS2012, foram cultivadas em casa de vegetação e forneceram as folhas para extração e análise de DNA. Quarenta e oito marcadores microssatélites (SSR) polimórficos foram avaliados na população F2. A avaliação dos fenótipos foi realizada em 207 famílias das progênies F2:3, em um delineamento em blocos ao acaso, com três repetições, conduzido em Viçosa, MG, em 2006. Foram detectados quatro QTL associados ao conteúdo de proteína, nos grupos de ligação D1a, G, A1, e I, e três QTL associados ao conteúdo de óleo, nos grupos A1, I e O. A variação fenotípica explicada pelos QTL variou de 6,24 a 18,94% e 17,26 a 25,93%, respectivamente, para os conteúdos de proteína e óleo. Foram detectados novos QTL associados aos conteúdos de proteína e óleo, além dos previamente relatados em outros estudos. Regiões distintas das atualmente conhecidas podem estar envolvidas no controle genético do teor de proteína e óleo na soja.The objective of this study was to detect and map quantitative trait loci (QTL) affecting soybean (Glycine max L. Merr.) protein and oil contents. F2 plants, derived from the cross between the CS3032PTA276 line and the variety UFVS2012, were grown in a greenhouse and provided the leaves for DNA extraction and analysis. Forty-eight polymorphic microsatelite markers (SSR) were evaluated in the F2 population. Evaluation of the phenotype was performed in 207 families from F2:3 progenies, in a complete block design with three replicates, carried out in Viçosa, MG, Brazil, in 2006. Four QTL associated with protein content, in linkage groups D1a, G, A1, and I, and three QTL for oil content in groups A1, I and O were identified. Phenotypic variation for protein and oil explained by QTL ranged from 6.24 to 18.94% and 17.26 to 25.93%, respectively. New QTL associated with protein and oil contents were detected, besides those previously reported in other studies.     Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Summary Background Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. Methods We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung’s disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. Findings We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung’s disease) from 264 hospitals (89 in high-income countries, 166 in middleincome countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in lowincome countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. Interpretation Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between lowincome, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030

    Development of orally disintegrating films for release of bioactive compounds

    É crescente o interesse dos consumidores por produtos naturais com apelo funcional. Por isso tem se estudado novos veículos de liberação de compostos bioativos, como por exemplo, os filmes de desintegração oral. A própolis é uma fonte natural rica em compostos ativos, como por exemplo, os compostos fenólicos, que lhe confere atividade antimicrobiana e antioxidante. O colágeno hidrolisado pode apresentar diversos benefícios quando ingerido, como aumento da densidade óssea e representa uma boa fonte de peptídeos. Este trabalho teve como objetivo desenvolver e caracterizar os filmes de desintegração oral à base de gelatina aditivados com colágeno hidrolisado (CH) e extrato etanólico de própolis (EEP). Os filmes foram produzidos pela técnica de casting mantendo constante a massa de gelatina(mG) colágeno hidrolisado(mCH) em 2 g de mG+CH (g/100g de solução filmogênica), e a concentração de sorbitol em 30g (g/100g de mG+CH). Foram avaliadas diferentes concentrações de CH (0, 10, 20 e 30 g/100 g de mG+CH) e de EEP (0, 100 e 200 g /100 g de mG+CH). Os filmes foram caracterizados em relação à cor e opacidade, matéria total solúvel, umidade, propriedades mecânicas (tensão na ruptura e elongação), microscopia eletrônica de varredura, mucoadesividade, ângulo de contato, tempo de desintegração, grau de intumescimento, concentração de compostos fenólicos, liberação in vitro, cinética de liberação, calorimetria diferencial de varredura, espectroscopia de infravermelho com transforma de Fourier (FTIR), atividade antimicrobiana e estabilidade dos compostos fenólicos em função do tempo armazenamento. O aumento da concentração de CH provocou aumento significativo da solubilidade. Nas propriedades mecânicas foi observado aumento da elongação para os filmes com adição de colágeno em relação ao filme controle e redução significativa da tensão na ruptura com o aumento da concentração de CH. Não foi observado variação da microestrutura com a adição de colágeno e os filmes com maior concentração de CH foram mais mucoadesivos. O aumento da concentração de CH provocou redução do tempo de desintegração, apresentando uma liberação mais rápida dos compostos fenólicos. Em relação ao aumento da concentração de EEP foram observados alterações nos parâmetros de cor e opacidade devido a cor característica do extrato. Observou-se redução da solubilidade e aumento na tensão de ruptura com o aumento de EEP. Para filmes aditivados com EEP foi observada uma microestrutura menos compacta e desordenada e aumento da mucoadesividade. O aumento da concentração de EEP provocou aumento do tempo de desintegração, com redução do grau de intumescimento, e uma liberação mais lenta dos compostos fenólicos. O processo de produção dos filmes não provocou degradação dos compostos fenólicos e os filmes aditivados apresentaram atividade antimicrobiana contra Staphylococcus aureus. Os filmes de desintegração oral mantiveram a concentração de compostos fenólicos após 84 dias de armazenamento. Desta forma, os filmes de desintegração oral aditivados com extrato etanólico de própolis e colágeno hidrolisado representam uma boa alternativa como veículo de compostos fenólicos na cavidade oral.There is an increase in the interest of consumers for natural products with functional appeal. Therefore, it has been studied new vehicles for release of bioactive compounds, such as orally disintegrating films. Propolis is a rich natural source of active compounds, such as phenolic compounds, responsible for antimicrobial and antioxidant activity. Hydrolyzed collagen may have several benefits when ingested, such as increased bone density and it is a good source of peptides. This study aimed the development and characterization of orally disintegrating films gelatin-based additives with hydrolyzed collagen (CH) and ethanol extract of propolis (EEP). The films were produced by casting, keeping mass constant of gelatin (mG) and hydrolyzed collagen (mCH) at 2 g of mG+CH/100 g filmogenic solution, and the concentration of sorbitol at 30 g/100 g mG+CH. It was evaluating different concentrations of CH (0, 10, 20 and 30 g/100 g of mG+CH) and EEP (0, 100 and 200 g/100 g of mG+CH). The films were characterized regarding color and opacity, total soluble matter, moisture, mechanical properties (tensile strength and elongation), scanning electron microscopy, mucoadhesion, contact angle, disintegration time, swelling degree, concentration of phenolic compounds, in vitro release, release kinetics, differential scanning calorimetry, Fourier transform infrared spectroscopy (FTIR), antimicrobial activity and stability of phenolic compounds were evaluated during the storage time. Increasing in concentration of CH caused a significant increase of solubility. For mechanical properties was observed increased at elongation for films with collagen in relation to control film and significant reduction of tensile strength with increasing of CH. It was not observed difference in microstructure because of CH addition and the films with higher concentrations of CH showed higher mucoadhesive. The increased of CH concentration promoted a reduction in disintegration time, showed a faster release of phenolic compounds. The increased of EEP concentration influenced color parameters and opacity due to color of the extract. It was observed a reduction of the solubility and an increase in tensile strength with the increase of EEP. The addition of EEP promoted a microstructure less compact and more disordered structure and increased mucoadhesion. Increased in EEP concentration increased disintegration time, reduced swelling degree, and caused a slower release of phenolic compounds. The production process did not cause degradation of phenolic compounds and additives films exhibited antimicrobial activity against Staphylococcus aureus. The orally disintegrating films remained the concentration of phenolic compounds after 84 days of storage. Thus, the orally disintegrating film additives with ethanol extract of propolis and hydrolyzed collagen represent a good alternative as a vehicle of phenolic compounds in the oral cavity

    Incorporation of natural active compounds into polymer matrices for oral release using the printing technique

    A tecnologia de impressão para incorporação do princípio ativo tem se mostrado promissora para o desenvolvimento de filmes para liberação oral. Estudos mostram que esta técnica foi eficiente na incorporação de fármacos sintéticos, entretanto pode ser interessante avaliar a possibilidade do uso desta técnica para incorporação de extratos naturais, devido as diversas atividades farmacológicas que eles podem apresentar. Dentre as fontes naturais, a romã (Punica granatum L.) tem se destacado pelo seu potencial antioxidante, anti-inflamatório e antimicrobiano. Este trabalho teve como objetivo avaliar as diferentes partes da Punica granatum L. como fonte de compostos ativos e analisar a possibilidade do uso da técnica de impressão para incorporação destes compostos em substratos para liberação oral. A extração dos compostos ativos foi avaliada para as diferentes partes da romã (casca, folha, mesocarpo e semente) em relação ao rendimento de sólidos solúveis totais, concentração de compostos fenólicos e flavonoides. O melhor extrato foi concentrado em rotaevaporador e avaliado em relação aos parâmetros importantes para uma solução de impressão (tensão superficial e viscosidade). Para utilização como substrato foram avaliados filmes à base de carboximetilcelulose (CMC), hidroxipropilmetilcelulose (HPMC) e blenda (CMC:HPMC - 1:1) produzidos pela técnica de tape-casting e papel comestível comercial. O extrato foi impresso utilizando uma impressora jato de tinta (L375, EPSON). Os substratos foram impressos com diferentes números de camada (1 à 4 camadas de impressão) e caracterizados em relação ao conteúdo de água, cor, pH de superfície, microscopia eletrônica de varredura e de força atômica, tempo de desintegração, concentração de compostos fenólicos, liberação in vitro, cinética de liberação, espectroscopia de infravermelho, difração de raio-X e estabilidade dos compostos fenólicos. A melhor extração foi obtida do mesocarpo, utilizando-se a temperatura de 70 ºC, tempo de 30 min e solução hidroalcoólica com 70% de álcool. Observou-se que os extratos obtidos do mesorcarpo (sem e com concentração) apresentaram intervalos de valores de tensão superficial e viscosidade dentro dos valores observados na literatura para impressora, entretato o extrato obtido do mesocarpo com remoção de 40% de solvente apresentou maior concentração de compostos fenólicos e por isso foi utilizado como solução de impressão. Analisando os substratos, observou-se que os filmes obtidos por tape-casting apresentaram maior tensão na ruptura, elongação e módulo elástico que o papel comestível comercial. Adicionalmente, os extratos apresentaram boa afinidade com os substratos testados. Os filmes à base de HPMC e blenda de CMC:HPMC não foram utilizados para impressão por curvarem rapidamente após serem retirados da placa, visto que essa deformação impediu que passassem pela cabeça de impressão sem serem danificados. Em relação aos filmes impressos, observou-se que o aumento do número de camadas aumentou a concentração de compostos fenólicos, possibilitando a produção sob demanda, de acordo com a necessidade individual. Não houve variação do pH de superfície e o tempo de desintegração do papel comestível comercial foi superior aos filmes à base de carboximetilcelulose. A liberação dos compostos foi influenciada pelo substrato utilizado e o lado de exposição (lado sem impressão e lado impresso). Os compostos fenólicos foram estáveis nos filmes no intervalo de 196 dias. Desta forma, pode-se concluir que a técnica de impressão pode ser utilizada para incorporação dos compostos fenólicos extraídos do mesocarpo da romã.Printing technology for incorporation of the active compounds has shown promise for the development of oral release films. Studies show that this technique was efficient in the incorporation of synthetic drugs, however it may be interesting to evaluate the possibility of using this technique to incorporate natural extracts, due to the various pharmacological activities that they can present. Among the natural sources, pomegranate (Punica granatum L.) has been highlighted by its antioxidant, anti-inflammatory and antimicrobial potential. This work aimed to evaluate the different parts of Punica granatum L. as a source of active compounds and to analyze the possibility of using the printing technique to incorporate these compounds into substrates for oral release. The extraction of the active compounds was evaluated for different parts of the pomegranate (bark, leaf, mesocarp and seed) in relation to the yield of total soluble solids, concentration of phenolic compounds and flavonoids. The best extract was concentrated in a rotavaporator and evaluated in relation to the parameters important for a printing solution (surface tension and viscosity). Films based on carboxymethylcellulose (CMC), hydroxypropylmethylcellulose (HPMC) and blend (CMC: HPMC - 1: 1) produced by the tape-casting technique and commercial edible paper were evaluated for use as substrate. The extract was printed using a piezoelectric inkjet printer (L375, EPSON). The substrates were printed with different numbers of layers (1 to 4 printing layers) and characterized in relation to water content, color, surface pH, scanning and atomic force electron microscopy, disintegration time, phenolic compound concentration, release in vitro, release kinetics, infrared spectroscopy, X-ray diffraction and stability of phenolic compounds. The best extract was obtaind from the mesocarp, using the extraction temperature of 70ºC, time of 30 min and hydroalcoholic solution with 70% ethyl alcohol. It was observed that the extracts obtained from the mesorcarp (without concentration and with different percentages of concentration) presented ranges of values of surface tension and viscosity within the values observed in the literature for printer, and the extract with removal of 40 % of solvent present hither concentration of phenolic compounds and therefore was used as print solution. By analyzing the substrates, it was observed that the films obtained by tape-casting had a higher tension in the rupture, elongation and elastic modulus than commercial edible paper. In addition, the extracts showed good affinity with the substrates tested. HPMC-based films and blends (HPMC:CMC) were not used for printing by bending rapidly after being removed from the plate as this deformation prevented them from passing through the print head without being damaged. In relation to the printed films, it was observed that the increase in the number of layers also increased the concentration of phenolic compounds, allowing the production on demand, according to the individual needs. There was no variation of the surface pH and the disintegration time of the commercial edible paper was higher than the carboxymethylcellulose based films. The release of the compounds was influenced by the substrate used and the exposure side (unprinted side and printed side). The phenolic compounds were stable in the films within the 196 day. In this way, it can be concluded that the printing technique can be used to incorporate the phenolic compounds extracted from the mesocarp of the pomegranate

Os filmes foram avaliados em relação aos fenóis totais, propriedade mecânica, solubilidade, ângulo de contato, tempo de desintegração e microestrutura. Os filmes foram produzidos por "casting" com concentrações de 2 g de massa proteica (gelatina e colágeno hidrolisado), 30 g de sorbitol/100 g de massa proteica e 100 g de extrato fenólico de própolis/100 g de massa proteica. Colágeno hidrolisado foi incorporado nas concentrações de 0, 10, 20 e 30 g/100 g de massa proteica. Verificou-se que o aumento da concentração de colágeno hidrolisado provocou redução na tensão de ruptura e aumento da elongação, entretanto todos os filmes apresentaram comportamento plástico. Observou-se também aumento da solubilidade à 25 ºC e redução do ângulo de contato, bem como do tempo de desintegração.
