    Thermo-mechanical sensitivity calibration of nanotorsional magnetometers

    We report on the fabrication of sensitive nanotorsional resonators, which can be utilized as magnetometers for investigating the magnetization dynamics in small magnetic elements.     Can Baryonic Features Produce the Observed 100 Mpc Clustering?

    We assess the possibility that baryonic acoustic oscillations in adiabatic models may explain the observations of excess power in large-scale structure on 100h^-1 Mpc scales. The observed location restricts models to two extreme areas of parameter space. In either case, the baryon fraction must be large (Omega_b/Omega_0 > 0.3) to yield significant features. The first region requires Omega_0 < 0.2h to match the location, implying large blue tilts (n>1.4) to satisfy cluster abundance constraints. The power spectrum also continues to rise toward larger scales in these models. The second region requires Omega_0 near 1, implying Omega_b well out of the range of big bang nucleosynthesis constraints; moreover, the peak is noticeably wider than the observations suggest. Testable features of both solutions are that they require moderate reionization and thereby generate potentially observable (about 1 uK) large-angle polarization, as well as sub-arc-minute temperature fluctuations. In short, baryonic features in adiabatic models may explain the observed excess only if currently favored determinations of cosmological parameters are in substantial error or if present surveys do not represent a fair sample of 100h^-1 Mpc structures.Comment: LaTeX, 7 pages, 5 Postscript figures, submitted to ApJ Letter

    Associations between purine metabolites and clinical symptoms in schizophrenia

    Background: The antioxidant defense system, which is known to be dysregulated in schizophrenia, is closely linked to the dynamics of purine pathway. Thus, alterations in the homeostatic balance in the purine pathway may be involved in the pathophysiology of schizophrenia. Methodology/Principal Findings: Breakdown products in purine pathway were measured using high-pressure liquid chromatography coupled with a coulometric multi-electrode array system for 25 first-episode neuroleptic-naïve patients with schizophrenia at baseline and at 4-weeks following initiation of treatment with antipsychotic medication. Associations between these metabolites and clinical and neurological symptoms were examined at both time points. The ratio of uric acid and guanine measured at baseline predicted clinical improvement following four weeks of treatment with antipsychotic medication. Baseline levels of purine metabolites also predicted clinical and neurological symtpoms recorded at baseline; level of guanosine was associated with degree of clinical thought disturbance, and the ratio of xanthosine to guanosine at baseline predicted degree of impairment in the repetition and sequencing of actions. Conclusions/Significance: Findings suggest an association between optimal levels of purine byproducts and dynamics in clinical symptoms and adjustment, as well as in the integrity of sensory and motor processing. Taken together, alterations in purine catabolism may have clinical relevance in schizophrenia pathology

    NF90 regulates inducible IL-2 gene expression in T cells

    Activation of T cells induces the production of T cell growth and survival factor interleukin (IL) 2. Regulatory T cells intrinsically fail to induce IL-2 expression upon activation and can suppress IL-2 production in conventional T cells. Thus, the control of IL-2 expression is critically important to T cell immune responses, yet the mechanisms remain incompletely understood. Nuclear factor (NF) 90 is a zinc-finger DNA- and double-stranded RNA-binding protein subunit that binds specifically to the antigen receptor response element (ARRE)/NF of activated T cells target sequence in the IL-2 proximal promoter. Inducible binding of NF90 to the IL-2 promoter in vivo is shown by chromatin immunoprecipitation. NF90 gene-targeted mice exhibit perinatal lethality. Compared with newborn NF90+/+ mice, newborn NF90−/− mice demonstrate severe impairment of IL-2 expression. Compared with wild-type cells, T cells deficient in NF90 are impaired in ARRE and IL-2 transcriptional activation and IL-2 mRNA stabilization. Fetal liver cells from NF90 gene-targeted mice were transplanted into irradiated adult recombination activating gene (RAG)–2−/− and IL-2Rγ−/− mice deficient in T cells, B cells, and natural killer cells. NF90+/+- and NF90−/−-RAG chimeric mice showed grossly normal repopulation of the thymus and spleen, but only NF90−/− T cells were severely impaired in IL-2 gene expression. Compared with littermates, NF90−/− RAG chimeric mice exhibited profound T cell lymphocytopenia in the peripheral circulation. Thus, NF90 regulates inducible IL-2 transcription, mRNA stability, and gene expression in T cells and represents a novel therapeutic target for the modulation of T cell immune responses

    Altered brain energetics induces mitochondrial fission arrest in Alzheimer's Disease.

    Altered brain metabolism is associated with progression of Alzheimer's Disease (AD). Mitochondria respond to bioenergetic changes by continuous fission and fusion. To account for three dimensional architecture of the brain tissue and organelles, we applied 3-dimensional electron microscopy (3D EM) reconstruction to visualize mitochondrial structure in the brain tissue from patients and mouse models of AD. We identified a previously unknown mitochondrial fission arrest phenotype that results in elongated interconnected organelles, "mitochondria-on-a-string" (MOAS). Our data suggest that MOAS formation may occur at the final stages of fission process and was not associated with altered translocation of activated dynamin related protein 1 (Drp1) to mitochondria but with reduced GTPase activity. Since MOAS formation was also observed in the brain tissue of wild-type mice in response to hypoxia or during chronological aging, fission arrest may represent fundamental compensatory adaptation to bioenergetic stress providing protection against mitophagy that may preserve residual mitochondrial function. The discovery of novel mitochondrial phenotype that occurs in the brain tissue in response to energetic stress accurately detected only using 3D EM reconstruction argues for a major role of mitochondrial dynamics in regulating neuronal survival

    Multiple Isoforms of ANRIL in Melanoma Cells: Structural Complexity Suggests Variations in Processing

    The long non-coding RNA ANRIL, antisense to the CDKN2B locus, is transcribed from a gene that encompasses multiple disease-associated polymorphisms. Despite the identification of multiple isoforms of ANRIL, expression of certain transcripts has been found to be tissue-specific and the characterisation of ANRIL transcripts remains incomplete. Several functions have been associated with ANRIL. In our judgement, studies on ANRIL functionality are premature pending a more complete appreciation of the profusion of isoforms. We found differential expression of ANRIL exons, which indicates that multiple isoforms exist in melanoma cells. In addition to linear isoforms, we identified circular forms of ANRIL (circANRIL). Further characterisation of circANRIL in two patient-derived metastatic melanoma cell lines (NZM7 and NZM37) revealed the existence of a rich assortment of circular isoforms. Moreover, in the two melanoma cell lines investigated, the complements of circANRIL isoforms were almost completely different. Novel exons were also discovered. We also found the family of linear ANRIL was enriched in the nucleus, whilst the circular isoforms were enriched in the cytoplasm and they differed markedly in stability. With respect to the variable processing of circANRIL species, bioinformatic analysis indicated that intronic Arthrobacter luteus (Alu) restriction endonuclease inverted repeats and exon skipping were not involved in selection of back-spliced exon junctions. Based on our findings, we hypothesise that “ANRIL” has wholly distinct dual sets of functions in melanoma. This reveals the dynamic nature of the locus and constitutes a basis for investigating the functions of ANRIL in melanoma.Peer reviewe

    Field-scale mapping of evaporative stress indicators of crop yield: An application over Mead, NE, USA

    The Evaporative Stress Index (ESI) quantifies temporal anomalies in a normalized evapotranspiration (ET) metric describing the ratio of actual-to-reference ET (fRET) as derived from satellite remote sensing. At regional scales (3–10 km pixel resolution), the ESI has demonstrated the capacity to capture developing crop stress and impacts on regional yield variability in water-limited agricultural regions. However, its performance in some regions where the vegetation cycle is intensively managed appears to be degraded due to spatial and temporal limitations in the standard ESI products. In this study, we investigated potential improvements to ESI by generating maps of ET, fRET, and fRET anomalies at high spatiotemporal resolution (30-m pixels, daily time steps) using a multi-sensor data fusion method, enabling separation of landcover types with different phenologies and resilience to drought. The study was conducted for the period 2010–2014 covering a region around Mead, Nebraska that includes both rainfed and irrigated crops. Correlations between ESI and measurements of maize yield were investigated at both the field and county level to assess the potential of ESI as a yield forecasting tool. To examine the role of crop phenology in yield-ESI correlations, annual input fRET time series were aligned by both calendar day and by biophysically relevant dates (e.g. days since planting or emergence). At the resolution of the operational U.S. ESI product (4 km), adjusting fRET alignment to a regionally reported emergence date prior to anomaly computation improves r2 correlations with county-level yield estimates from 0.28 to 0.80. At 30-m resolution, where pure maize pixels can be isolated from other crops and landcover types, county-level yield correlations improved from 0.47 to 0.93 when aligning fRET by emergence date rather than calendar date. Peak correlations occurred 68 days after emergence, corresponding to the silking stage for maize when grain development is particularly sensitive to soil moisture deficiencies. The results of this study demonstrate the utility of remotely sensed ET in conveying spatially and temporally explicit water stress information to yield prediction and crop simulation models
