    Rheology and Settling Dynamics of a Particle Filled Aqueous Polymer Gel for Hydraulic Fracturing Fluid Systems

    Outstanding Undergraduate Award for Research Excellence 2013Outstanding Undergraduate Award for Research Excellence 2015Hydraulic fracturing, more commonly known as fracking, is a hydrocarbon capturing method that uses a high pressure fluid to fracture layers of shale, which then releases the hydrocarbons which flow to the surface for capture. In order to keep the fractures open, special sands called proppants are carried in with the fracking fluid. This functions by keeping the fracture open, while allowing natural gas to flow through the fracture. Hydroxypropyl Guar (HPG) is a high molecular weight, water soluble polymer used in the food industry as a viscosifier of water based foods, and in the petroleum industry as an additive in hydraulic fracturing fluids. The fracking fluid is further modified via addition of a cross linker, which causes the fluid to become a gel, making it extremely viscous. Once the proppant is suspended in the fracture, a breaker is added to the solution which allows the fluid to flow again, while retaining the structure of the fracture. Unfortunately, some of the current fluid components used in industry are hazardous and present health risks upon unanticipated exposure. This research aims to maintain, or surpass, current fluid standards by developing potential substitutes for the presently used components. To do this, a full factorial experimental design has been conducted with cross linker concentration and chopped nylon fiber concentration as exploratory variables. It is observed that the addition of the chopped fibers significantly decreases the settling velocity of suspended proppant particles, while not significantly affecting the fluid viscosity, thus allowing for a decrease in the toxic cross linker concentration for a given required settling velocity.     Structure and Function Studies of De Novo Peptides Containing Novel Amino Acids.

    A series of amphipathic peptides rich in alpha,alpha-disubstituted amino acids (alphaalphaAA\u27s) was synthesized and studied for antimicrobial activity and helix preferences in various solvent systems. In addition, a constrained lactam dipeptide amino acid was synthesized and incorporated into a peptide to examine beta-sheet dimer formation. Several short, amphipathic, 310- and alpha-helical peptides containing 80% alphaalphaAA\u27s were synthesized using various combinations of 1-aminocyclohexane-1-carboxylic acid, alpha-aminoisobutyric acid, and 4-aminopiperidine-4-carboxylic acid. The peptides showed direct antimicrobial activity against E. coli and S. aureus. In addition, several of the peptides showed in vivo and in vitro activity against Brucella abortus and Mycobacterium chelonei. Studies were done with a strain of B. abortus expressing a green fluorescent protein to show the peptides selectively destroyed infected macrophages. The series of amphipathic 310- and alpha-helical peptides were also studied, using circular dichroism spectroscopy, for their helix preference in organic and aqueous-organic solvent systems. The alpha-helical peptides showed the expected spectra in all solvents used. One designed 3 10-helical peptide exhibited a transition from a 310-helix to an alpha-helix as the water content was increased. The other designed 310-helical peptide exhibited a spectrum indicative of an alpha-helix in all solvents tested, presumably due to steric interactions of the amino acid side-chains. A constrained lactam dipeptide amino acid, 3-(R)-N \u27-[(9H-fluoren-9-ylmethoxy)carbonyl]-amino-1-[1-(S)-methylethyledianoic acid]-2-piperidinone, was synthesized from D-glutamic acid and incorporated into a peptide. The starting material was esterified with allyl alcohol and the alpha-nitrogen was doubly protected with Boc protecting groups. Selective DIBAL reduction of the side-chain ester yielded the semi-aldehyde. Reductive amination with valine followed by removal of the remaining allyl ester and intramolecular amide bond formation yielded the fully protected constrained lactam dipeptide amino acid. Treatment with TFA and introduction of the Fmoc protecting group resulted in the amino acid ready for peptide coupling. A peptide using the constrained amino acid was synthesized and studied for beta-sheet dimer formation

Fermée peu après son ouverture pour raisons de sécurité, son entrée a été aménagée en 2016. Le réseau se développe sur deux niveaux, à 630 et 610 m d’altitude, le niveau 630 comprenant des travaux montants et le niveau 610 des travaux descendants. Entre les deux, un puits sur filon comprenant quelques courtes recherches a été boisé courant 2016. La campagne 2017 a permis de vider un puits ..

    Sainte-Marie-aux-Mines – Sainte-Barbe

    La campagne 2017 sur le carreau Sainte-Barbe a été celle de l’ouverture d’une zone de fouille de 100 m2 sur la plate-forme principale de la halde. Cette zone a été définie d’après la prospection géophysique (2015) et les sondages (2016). Le décapage à la pelle mécanique a permis d’ôter l’important recouvrement de matériaux de colluvionnement (1,00 à 1,90 m). Le nettoyage de la zone a permis de mettre au jour un bâtiment quadrangulaire d’environ 9 m par 5 m. Les niveaux de sol ont été atteints..
