    Genomes from Verteba cave suggest diversity within the Trypillians in Ukraine

    The transition to agriculture occurred relatively late in Eastern Europe, leading researchers to debate whether it was a gradual, interactive process or a colonisation event. In the forest and forest-steppe regions of Ukraine, farming appeared during the fifth millennium BCE, associated with the Cucuteni-Trypillia cultural complex (CTCC, ~ 5000-3000 BCE). Across Europe, the Neolithisation process was highly variable across space and over time. Here, we investigate the population dynamics of early agriculturalists from the eastern forest-steppe region based on the analyses of 20 ancient genomes from the site of Verteba Cave (3935-825 cal BCE). Results reveal that the CTCC individuals' ancestry is related to both western hunter-gatherers and Near Eastern farmers, has no local ancestry associated with Ukrainian Neolithic hunter-gatherers and has steppe ancestry. An Early Bronze Age individual has an ancestry profile related to the Yamnaya expansions but with 20% of ancestry related to the other Trypillian individuals, which suggests admixture between the Trypillians and the incoming populations carrying steppe-related ancestry. A Late Bronze Age individual dated to 980-825 cal BCE has a genetic profile indicating affinity to Beaker-related populations, detected close to 1000 years after the end of the Bell Beaker phenomenon during the third millennium BCE

    Background: Juvenile Arthritis Disease Activity Score (JADAS) is a 4 variable composite disease activity (DA) score for JIA (including active 10, 27 or 71 joint count (AJC), physician global (PGA), parent/child global (PGE) and ESR). The validity of JADAS for all ILAR subtypes in the routine clinical setting is unknown. We investigated the construct validity of JADAS in the clinical setting in all subtypes of JIA through application to a prospective inception cohort of UK children presenting with new onset inflammatory arthritis. Methods: JADAS 10, 27 and 71 were determined for all children in the Childhood Arthritis Prospective Study (CAPS) with complete data available at baseline. Correlation of JADAS 10, 27 and 71 with single DA markers was determined for all subtypes. All correlations were calculated using Spearman's rank statistic. Results: 262/1238 visits had sufficient data for calculation of JADAS (1028 (83%) AJC, 744 (60%) PGA, 843 (68%) PGE and 459 (37%) ESR). Median age at disease onset was 6.0 years (IQR 2.6-10.4) and 64% were female. Correlation between JADAS 10, 27 and 71 approached 1 for all subtypes. Median JADAS 71 was 5.3 (IQR 2.2-10.1) with a significant difference between median JADAS scores between subtypes (p < 0.01). Correlation of JADAS 71 with each single marker of DA was moderate to high in the total cohort (see Table 1). Overall, correlation with AJC, PGA and PGE was moderate to high and correlation with ESR, limited JC, parental pain and CHAQ was low to moderate in the individual subtypes. Correlation coefficients in the extended oligoarticular, rheumatoid factor negative and enthesitis related subtypes were interpreted with caution in view of low numbers. Conclusions: This study adds to the body of evidence supporting the construct validity of JADAS. JADAS correlates with other measures of DA in all ILAR subtypes in the routine clinical setting. Given the high frequency of missing ESR data, it would be useful to assess the validity of JADAS without inclusion of the ESR.     Incidence and Characterisation of Methicillin-Resistant Staphylococcus aureus (MRSA) from Nasal Colonisation in Participants Attending a Cattle Veterinary Conference in the UK

    We sought to determine the prevalence of nasal colonisation with methicillin-resistant Staphylococcus aureus among cattle veterinarians in the UK. There was particular interest in examining the frequency of colonisation with MRSA harbouring mecC, as strains with this mecA homologue were originally identified in bovine milk and may represent a zoonotic risk to those in contact with dairy livestock. Three hundred and seven delegates at the British Cattle Veterinarian Association (BCVA) Congress 2011 in Southport, UK were screening for nasal colonisation with MRSA. Isolates were characterised by whole genome sequencing and antimicrobial susceptibility testing. Eight out of three hundred and seven delegates (2.6%) were positive for nasal colonisation with MRSA. All strains were positive for mecA and none possessed mecC. The time since a delegate’s last visit to a farm was significantly shorter in the MRSA-positive group than in MRSA-negative counterparts. BCVA delegates have an increased risk of MRSA colonisation compared to the general population but their frequency of colonisation is lower than that reported from other types of veterinarian conference, and from that seen in human healthcare workers. The results indicate that recent visitation to a farm is a risk factor for MRSA colonisation and that mecC-MRSA are rare among BCVA delegates (<1% based on sample size). Contact with livestock, including dairy cattle, may still be a risk factor for human colonisation with mecC-MRSA but occurs at a rate below the lower limit of detection available in this study

    Affinity for self antigen selects T<sub>reg</sub> cells with distinct functional properties

    The manner in which regulatory T cells (Treg cells) control lymphocyte homeostasis is not fully understood. We identified two Treg cell populations with differing degrees of self-reactivity and distinct regulatory functions. We found that GITR(hi)PD-1(hi)CD25(hi) (Triple(hi)) Treg cells were highly self-reactive and controlled lympho-proliferation in peripheral lymph nodes. GITR(lo)PD-1(lo)CD25(lo) (Triple(lo)) Treg cells were less self-reactive and limited the development of colitis by promoting the conversion of CD4(+) Tconv cells into induced Treg cells (iTreg cells). Although Foxp3-deficient (Scurfy) mice lacked Treg cells, they contained Triple(hi)-like and Triple(lo)-like CD4(+) T cells zsuper< T cells infiltrated the skin, whereas Scurfy Triple(lo)CD4(+) T cells induced colitis and wasting disease. These findings indicate that the affinity of the T cell antigen receptor for self antigen drives the differentiation of Treg cells into distinct subsets with non-overlapping regulatory activities

    Birds in burials: The role of avifauna in Eneolithic Tripolye mortuary rituals

    Bird remains are rare at Tripolye sites; therefore, researchers hypothesize that they were not an important economic resource for the Tripolye. The use of ornithographic iconography, vessels, and figurines suggests avifauna were important symbolically in Tripolye ideology. Here, we investigate the role of birds in a Tripolye burial context to assess their symbolic and/or economic significance in mortuary behaviours. We analysed bird remains from the Verteba Cave cemetery, located in western Ukraine and occupied by the Tripolye during phase BII through CII. Bird remains (n=178) comprise approximately 2% of the faunal sample excavated among human burials from the cave. In contrast to species represented symbolically in Tripolye art, water and marsh birds are majorly underrepresented. The bird assemblage is dominated by grouse ('Tetrao spp.'), and common quail ('Cortunix cortunix'), but also includes birds of prey, corvids, and many songbirds. The large number of gamebirds leads us to conclude that the avifauna from Verteba Cave are likely the remains from funeral feasts or food grave offerings. It also supports the idea that birds were a seasonal and local economic resource. The presence of birds of prey, corvids, and male black grouse also suggests that feathers may have been a desired resource

    The Biological Implications of the Transition to Agriculture in Ukraine: A Study of Enamel Hypoplasias

    The Tripolye were the first archaeological culture in Ukraine to cultivate domesticated cereals, practice animal husbandry, and establish large settlements with high population densities. This cultural adaptation was much different than that of mobile hunter-fisher-gatherers of the Ukrainian Mesolithic/Neolithic, and likely resulted in different outcomes for human health. This study compares the rates of enamel hypoplasias in a Tripolye skeletal population with that of Mesolithic/ Neolithic hunter-fisher-gatherers. A recently excavated sample of dentitions representing a minimum of 35 individuals from Verteba Cave was examined macroscopically for hypoplasias and was compared statistically to published rates for hunter-fisher-gatherers. The Tripolye from Verteba Cave were found to have at least one enamel hypoplasia on 18.18% of teeth, while the hunter-fisher-gatherers have hypoplastic lesions on 1.88% of teeth. When examined at the individual level, 48.57% of the Tripolye were found to have at least one hypoplasia, as compared to 12.77% of the hunter-fisher-gatherer individuals. The results indicate that the agropastoral Tripolye experienced significantly more systemic stress than the hunter-fisher-gatherers. The higher stress likely relates to dietary and behavioral variables associated with the Tripolye's agropastoral economy, including heavy reliance on cereals as weaning foods and sanitary problems linked to sedentism
