Multiplicity of Meaning in the Last Moments of Hamlet

This essay examines ambiguities in Hamlet and argues that wordplay represents the nature of Hamlet's mind, making audiences keenly aware of his inner dissatisfaction and come to expect some unambiguous resolution at the end of the tragedy

Polymicrobial oral biofilm models: simplifying the complex

    Get PDF
Over the past century, numerous studies have used oral biofilm models to investigate growth kinetics, biofilm formation, structure and composition, antimicrobial susceptibility and host–pathogen interactions. In vivo animal models provide useful models of some oral diseases; however, these are expensive and carry vast ethical implications. Oral biofilms grown or maintained in vitro offer a useful platform for certain studies and have the advantages of being inexpensive to establish and easy to reproduce and manipulate. In addition, a wide range of variables can be monitored and adjusted to mimic the dynamic environmental changes at different sites in the oral cavity, such as pH, temperature, salivary and gingival crevicular fluid flow rates, or microbial composition. This review provides a detailed insight for early-career oral science researchers into how the biofilm models used in oral research have progressed and improved over the years, their advantages and disadvantages, and how such systems have contributed to our current understanding of oral disease pathogenesis and aetiology

Saltatory Relaxation of the Cosmological Constant

We modify and extend an earlier proposal by Brown and Teitelboim to relax the effective cosmological term by nucleation of branes coupled to a three-index gauge potential. Microscopic considerations from string/M theory suggest two major innovations in the framework. First, the dependence of brane properties on the compactification of extra dimensions may generate a very small quantized unit for jumps in the effective cosmological term. Second, internal degrees of freedom for multiply coincident branes may enhance tunneling rates by exponentially large density of states factors. These new features essentially alter the relaxation dynamics. By requiring stability on the scale of the lifetime of the universe, rather than absolute stability, we derive a non-trivial relation between the supersymmetry breaking scale and the value of the cosmological term. It is plausibly, though not certainly, satisfied in Nature.Comment: 25 pages, revised and expanded discussio

The Effects of Nicotine in the Neonatal Quinpirole Rodent Model of Psychosis: Neural Plasticity Mechanisms and Nicotinic Receptor Changes

Neonatal quinpirole (NQ) treatment to rats increases dopamine D2 receptor sensitivity persistent throughout the animal’s lifetime. In Experiment 1, we analyzed the role of α7 and α4β2 nicotinic receptors (nAChRs) in nicotine behavioral sensitization and on the brain-derived neurotrophic factor (BDNF) response to nicotine in NQ- and neonatally saline (NS)-treated rats. In Experiment 2, we analyzed changes in α7 and α4β2 nAChR density in the nucleus accumbens (NAcc) and dorsal striatum in NQ and NS animals sensitized to nicotine. Male and female Sprague-Dawley rats were neonatally treated with quinpirole (1 mg/kg) or saline from postnatal days (P)1–21. Animals were given ip injections of either saline or nicotine (0.5 mg/kg free base) every second day from P33 to P49 and tested on behavioral sensitization. Before each injection, animals were ip administered the α7 nAChR antagonist methyllycaconitine (MLA; 2 or 4 mg/kg) or the α4β2 nAChR antagonist dihydro beta erythroidine (DhβE; 1 or 3 mg/kg). Results revealed NQ enhanced nicotine sensitization that was blocked by DhβE. MLA blocked the enhanced nicotine sensitization in NQ animals, but did not block nicotine sensitization. NQ enhanced the NAcc BDNF response to nicotine which was blocked by both antagonists. In Experiment 2, NQ enhanced nicotine sensitization and enhanced α4β2, but not 7, nAChR upregulation in the NAcc. These results suggest a relationship between accumbal BDNF and α4β2 nAChRs and their role in the behavioral response to nicotine in the NQ model which has relevance to schizophrenia, a behavioral disorder with high rates of tobacco smoking
