    Na(v )1.8-null mice show stimulus-dependent deficits in spinal neuronal activity

    BACKGROUND: The voltage gated sodium channel Na(v )1.8 has a highly restricted expression pattern to predominantly nociceptive peripheral sensory neurones. Behaviourally Na(v )1.8-null mice show an increased acute pain threshold to noxious mechanical pressure and also deficits in inflammatory and visceral, but not neuropathic pain. Here we have made in vivo electrophysiology recordings of dorsal horn neurones in intact anaesthetised Na(v )1.8-null mice, in response to a wide range of stimuli to further the understanding of the functional roles of Na(v )1.8 in pain transmission from the periphery to the spinal cord. RESULTS: Na(v )1.8-null mice showed marked deficits in the coding by dorsal horn neurones to mechanical, but not thermal, -evoked responses over the non-noxious and noxious range compared to littermate controls. Additionally, responses evoked to other stimulus modalities were also significantly reduced in Na(v )1.8-null mice where the reduction observed to pinch > brush. The occurrence of ongoing spontaneous neuronal activity was significantly less in mice lacking Na(v )1.8 compared to control. No difference was observed between groups in the evoked activity to electrical activity of the peripheral receptive field. CONCLUSION: This study demonstrates that deletion of the sodium channel Na(v )1.8 results in stimulus-dependent deficits in the dorsal horn neuronal coding to mechanical, but not thermal stimuli applied to the neuronal peripheral receptive field. This implies that Na(v )1.8 is either responsible for, or associated with proteins involved in mechanosensation

    The effects of more realistic forms of lead heterogeneity in soil on uptake, biomass and root response of two brassica species

    The spatial heterogeneity of soil constituents is known to have significant impacts on plant growth and plant uptake of nutrients and contaminants, yet studies have rarely used patterns of heterogeneity based on those found in the field. Heterogeneity refers to how lumpy materials are distributed in the soil, whilst homogeneity is the uniformity in the distribution of such materials. We identified patterns of lead contamination at historically polluted field sites and conducted pot trials using field–based parameters to determine the pattern of distribution of lead within the pots. We examined plant Pb uptake and growth in simulated low, medium and high heterogeneity environments as well as a control homogeneous treatment. We found a significant effect of Pb spatial heterogeneity on uptake and biomass of two Brassica species (Brassica napus and Brassica juncea), both candidate species for phytoremediation projects. Biomass was 4 to 5 fold lower in the high heterogeneity treatment and total plant Pb uptake as Pb mass in (µg) was 40 to 80% lower, compared to the homogeneous treatment. Plant lead concentration (mg/kg) increased by a factor of 2 with increasing heterogeneity. Peak uptake was observed in low and medium heterogeneity treatments of B. napus and B. juncea respectively. We also explored roots behaviour in the high heterogeneity treatment and found variation in root mass by 20 to 80% between concentric patches with significant (P < 0.05) differences between patches and species. High proportion of roots (40 to 50%) were proliferated in patches of lower Pb concentration. The tap root was a greater proportion of root in B. napus, which was absent in B. juncea. Results suggest that root morphology of this plant species might be a factor influencing the placement of roots in concentric patches and consequently the overall root response to Pb spatial heterogeneity. This is an indication that the root response could be realistic of that experienced by plants in field conditions. Generally result showed that spatial heterogeneity of Pb has a significant effect on plant growth and biomass. This study also demonstrated that the presence and extent of in situ heterogeneity of Pb in soil plays an important role in Pb uptake by plants. This work has implications for improving the phytoremediation of Pb contaminated land, phytomining, the reliability of risk assessment/models of human exposure to Pb and the quality of trace mineral content of agricultural produce

    Microstructure mixing observations and finescale parameterizations in the Beaufort Sea

Microstructure measurements from a free-falling profiler survey showed very low [O(10−10) W kg−1] turbulent kinetic energy dissipation rates ε. A finescale parameterization based on both shear and strain measurements was applied to estimate the ratio of shear to strain Rω and ε at the mooring location, and a strain-based parameterization was applied to the microstructure survey (which occurred approximately 100 km away from the mooring site) for direct comparison with microstructure results. The finescale parameterization worked well, with discrepancies ranging from a factor of 1–2.5 depending on depth. The largest discrepancies occurred at depths with high shear. Mean Rω was 17, and Rω showed high variability with values ranging from 3 to 50 over 8 days. Observed ε was slightly elevated (factor of 2–3 compared with a later survey of 11 profiles taken over 3 h) from 25 to 125 m following a wind event which occurred at the beginning of the mooring deployment, reaching a maximum of ε= 6 × 10−10 W kg−1 at 30-m depth. Velocity signals associated with near-inertial waves (NIWs) were observed at depths greater than 200 m, where the Atlantic Water mass represents a reservoir of oceanic heat. However, no evidence of elevated ε or heat fluxes was observed in association with NIWs at these depths in either the microstructure survey or the finescale parameterization estimates.This work was supported by NSF Grants PLR 14-56705 and PLR-1303791 and by NSF Graduate Research Fellowship Grant DGE-1650112

    Effects Of A Low Dose Of Fish Oil On Inflammatory Markers Of Brazilian Hiv-infected Adults On Antiretroviral Therapy: A Randomized, Parallel, Placebo-controlled Trial

    Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Background: The benefits of antiretroviral therapy for HIV-infected subjects have been limited by an increased risk of metabolic and cardiovascular diseases. The objective of this study was to assess the effects of a low dose of marine omega-3 fatty acids on inflammatory marker concentrations in HIV-infected subjects under antiretroviral therapy (ART). Methods: This was a randomized, parallel, placebo-controlled trial that investigated the effects of 3 g fish oil/day (540 mg of eicosapentaenoic acid-EPA plus 360 mg of docosahexaenoic acid-DHA) or 3 g soy oil/day (placebo) for 24 weeks in 83 male and non-pregnant female HIV-infected adults on ART. Results: There were no differences between groups for the measures at baseline. Multilevel analyses revealed no statistically significant relationship between the longitudinal changes in high sensitivity-C reactive protein (hs-CRP) (Wald Chi2 = 0.17, p = 0.918), fibrinogen (Wald Chi2 = 3.82, p = 0.148), and factor VIII (Wald Chi2 = 5.25, p = 0.073) with fish oil. No significant changes in interleukin-6 (IL6), interleukin-1 beta (IL1-beta) and tumor necrosis factor-alpha (TNF-alpha) serum concentrations were observed with fish oil supplements for 12 weeks. Conclusions: Compared to placebo, a low dose of 900 mg omega-3 fatty acids (EPA plus DHA) in fish oil capsules did not change hs-CRP, fibrinogen, factor VIII, IL6, IL1-beta and TNF-alpha serum concentrations in HIV-infected subjects on ART. Further investigations should consider the assessment of more sensitive inflammatory markers or higher doses to evaluate the effects of marine omega-3 fatty acids in this population.
