PRAS40 suppresses atherogenesis through inhibition of mTORC1-dependent pro-inflammatory signaling in endothelial cells
Endothelial pro-inflammatory activation plays a pivotal role in atherosclerosis, and many pro-inflammatory and atherogenic signals converge upon mechanistic target of rapamycin (mTOR). Inhibitors of mTOR complex 1 (mTORC1) reduced atherosclerosis in preclinical studies, but side effects including insulin resistance and dyslipidemia limit their clinical use in this context. Therefore, we investigated PRAS40, a cell type-specific endogenous modulator of mTORC1, as alternative target. Indeed, we previously found PRAS40 gene therapy to improve metabolic profile; however, its function in endothelial cells and its role in atherosclerosis remain unknown. Here we show that PRAS40 negatively regulates endothelial mTORC1 and pro-inflammatory signaling. Knockdown of PRAS40 in endothelial cells promoted TNFα-induced mTORC1 signaling, proliferation, upregulation of inflammatory markers and monocyte recruitment. In contrast, PRAS40-overexpression blocked mTORC1 and all measures of pro-inflammatory signaling. These effects were mimicked by pharmacological mTORC1-inhibition with torin1. In an in vivo model of atherogenic remodeling, mice with induced endothelium-specific PRAS40 deficiency showed enhanced endothelial pro-inflammatory activation as well as increased neointimal hyperplasia and atherosclerotic lesion formation. These data indicate that PRAS40 suppresses atherosclerosis via inhibition of endothelial mTORC1-mediated pro-inflammatory signaling. In conjunction with its favourable effects on metabolic homeostasis, this renders PRAS40 a potential target for the treatment of atherosclerosis
Expected Workforce Requirements for the Green Transition
In dieser Arbeit wird die erwartete zusätzliche Arbeitsnachfrage untersucht, die durch das Wachstum der Produktion grüner Produkte in Österreich entstehen wird. Hier schätzen wir ab, wie viele zusätzliche Mitarbeiter benötigt werden und in welchen Wirtschaftssektoren sie beschäftigt sein werden, um die identifizierten Branchen zu fördern.
Wir stellen fest, dass Österreichs fünf vielversprechendste Möglichkeiten zur grünen Diversifizierung den größten Arbeitskräftebedarf in den Sektoren "Maschinen und Geräte" sowie "Reparatur von Maschinen und Geräten" verursachen werden. Die insgesamt geschätzte Zunahme der Arbeitsnachfrage in allen Sektoren beträgt etwa 6,30 Mitarbeiter.
Die Ergebnisse dieser Studie sind über ein Online-Dashboard zugänglich und können erforscht werden, das die die Untersuchung von maßgeschneiderten Szenarien ermöglicht. Das Dashboard ist veröffentlicht unter: https://ecto.rfte.at
Strain-Tuning of the Optical Properties of Semiconductor Nanomaterials by Integration onto Piezoelectric Actuators
The tailoring of the physical properties of semiconductor nanomaterials by
strain has been gaining increasing attention over the last years for a wide
range of applications such as electronics, optoelectronics and photonics. The
ability to introduce deliberate strain fields with controlled magnitude and in
a reversible manner is essential for fundamental studies of novel materials and
may lead to the realization of advanced multi-functional devices. A prominent
approach consists in the integration of active nanomaterials, in thin epitaxial
films or embedded within carrier nanomembranes, onto
Pb(Mg1/3Nb2/3)O3-PbTiO3-based piezoelectric actuators, which convert electrical
signals into mechanical deformation (strain). In this review, we mainly focus
on recent advances in strain-tunable properties of self-assembled InAs quantum
dots embedded in semiconductor nanomembranes and photonic structures.
Additionally, recent works on other nanomaterials like rare-earth and metal-ion
doped thin films, graphene and MoS2 or WSe2 semiconductor two-dimensional
materials are also reviewed. For the sake of completeness, a comprehensive
comparison between different procedures employed throughout the literature to
fabricate such hybrid piezoelectric-semiconductor devices is presented. Very
recently, a novel class of micro-machined piezoelectric actuators have been
demonstrated for a full control of in-plane stress fields in nanomembranes,
which enables producing energy-tunable sources of polarization-entangled
photons in arbitrary quantum dots. Future research directions and prospects are

Agent-based simulations for protecting nursing homes with prevention and vaccination strategies
Due to its high lethality amongst the elderly, the safety of nursing homes
has been of central importance during the COVID-19 pandemic. With test
procedures becoming available at scale, such as antigen or RT-LAMP tests, and
increasing availability of vaccinations, nursing homes might be able to safely
relax prohibitory measures while controlling the spread of infections (meaning
an average of one or less secondary infections per index case). Here, we
develop a detailed agent-based epidemiological model for the spread of
SARS-CoV-2 in nursing homes to identify optimal prevention strategies. The
model is microscopically calibrated to high-resolution data from nursing homes
in Austria, including detailed social contact networks and information on past
outbreaks. We find that the effectiveness of mitigation testing depends
critically on the timespan between test and test result, the detection
threshold of the viral load for the test to give a positive result, and the
screening frequencies of residents and employees. Under realistic conditions
and in absence of an effective vaccine, we find that preventive screening of
employees only might be sufficient to control outbreaks in nursing homes,
provided that turnover times and detection thresholds of the tests are low
enough. If vaccines that are moderately effective against infection and
transmission are available, control is achieved if 80% or more of the
inhabitants are vaccinated, even if no preventive testing is in place and
residents are allowed to have visitors. Since these results strongly depend on
vaccine efficacy against infection, retention of testing infrastructures,
regular voluntary screening and sequencing of virus genomes is advised to

ifo Konjunkturprognose 2009/2010: Abschwung setzt sich fort
Die Weltwirtschaft steckt in der tiefsten Rezession seit der großen Depression. Seit diesem Frühjahr dürfte sich das Kontraktionstempo jedoch abgeschwächt haben. Weltweit sind Programme zur Stützung der Konjunktur aufgelegt worden, zudem zeigt die expansive Geldpolitik der Zentralbanken allmählich Wirkung. Auch sind in zahlreichen Ländern Maßnahmen zur Stabilisierung der Finanzmärkte ergriffen worden. Schließlich werden die Realeinkommen durch die starken Rohstoffpreisrückgänge gestützt. Das vom ifo Institut erhobene Weltwirtschaftsklima ist im zweiten Quartal 2009 erstmals seit dem Herbst 2007 wieder gestiegen. Der Anstieg des Indikators resultiert jedoch ausschließlich aus den günstigeren Erwartungen für die nächsten sechs Monate; die Einschätzung der derzeitigen wirtschaftlichen Lage hat sich demgegenüber noch weiter verschlechtert und fiel auf einen neuen historischen Tiefstand. Die deutsche Wirtschaft befindet sich in der schwersten Rezession seit Bestehen der Bundesrepublik. Nach den bisher vorliegenden amtlichen Ergebnissen sank die gesamtwirtschaftliche Produktion im ersten Quartal 2009 saison- und kalenderbereinigt gegenüber dem Jahresendquartal um 3,8%, bereits im vierten Quartal 2008 war die Wirtschaftsleistung um 2,2% geschrumpft. Deutschland verzeichnet damit von allen großen europäischen Volkswirtschaften den schärfsten Wachstumseinbruch. Die gesamtwirtschaftliche Produktion dürfte nach diesem drastischen Rückgang auch im zweiten Quartal gesunken sein, jedoch mit deutlich verringertem Tempo. Für den Beginn einer allmählichen Stabilisierung spricht die aktuelle Entwicklung einer Reihe von gewichtigen Konjunkturindikatoren wie Produktion und Auftragseingang sowie das ifo Geschäftsklima.
