A comparison of manual counting of rabbit reticulocytes with ADVIA 2120i analyzer counting
We compared manual counting of reticulocytes in rabbits with automatic counting using an ADVIA 2120i analyzer. Reproducibility and the influence of different anticoagulants (EDTA and Li-heparin) were also examined. Blood samples of 331 rabbits (method comparison, n = 289; reproducibility, n = 33; comparison of anticoagulants, n = 9) were tested. The reticulocyte numbers of each specimen were manually determined twice for method comparison. Passing–Bablok regressions, Bland–Altman plots, and the coefficient of variation (CV) were used to evaluate statistical significance. Good correlation (rs = 0.81) was observed between manual reticulocyte counting (groups 1–4) and the ADVIA 2120i. Quantification with the ADVIA 2120i was reproducible for relative reticulocyte numbers (EDTA, CV = 4.24%; Li-heparin, CV = 3.63%) and absolute reticulocyte numbers (EDTA, CV = 5.64%; Li-heparin, CV = 3.81%). The absolute and relative reticulocyte numbers were significantly higher in Li-heparin samples than in EDTA samples (absolute, p = 0.009; relative, p = 0.016). The ADVIA 2120i is suitable for counting reticulocytes in rabbit blood samples, but reticulocyte numbers are higher by manual counting than by ADVIA 2120i counting. Therefore, microscopic confirmation of quantifications is recommended when high numbers of reticulocytes are observed. The anticoagulant of choice is EDTA

Background: Flexible design of clinical trials may allow tailoring ongoing investigations to better suit actual circumstances such as an unexpectedly low effect size without compromising error control properties. Available statistical methods are, however, complex.Method: A two-stage flexible design is represented in form of a simple chance tree. Critical limits accounting for one interim analysis are derived using only basic rules of probability calculus.Conclusion: By means of chance trees the principles of flexible trial design may be communicated more easily, thus hopefully adding to good design and conduct of clinical trials.Hintergrund: Flexible Designs für klinische Studien erlauben eine bessere Anpassung laufender Untersuchungen an aktuelle Umstände, wie z.B. eine unerwartet kleine Effektgröße, ohne die statistische Fehler-Kontrolle zu durchbrechen. Die vorhandenen statistischen Verfahren sind jedoch komplex.Methodik: Ein zweistufiges, flexibles Design wird in Form eines einfachen Wahrscheinlichkeitsbaums repräsentiert. Kritische Grenzen, die einer Zwischenauswertung Rechnung tragen, werden mittels fundamentaler Regeln der Wahrscheinlichkeitsrechnung abgeleitet.Schlussfolgerung: Mithilfe eines Wahrscheinlichkeitsbaums können die Prinzipien flexibler Studiendesigns einfach kommuniziert werden und somit hoffentlich zu guter Planung und Durchführung von klinischen Studien beitragen
Carbene based photochemical molecular assemblies for solar driven hydrogen generation
Novel photocatalysts based on ruthenium complexes with NHC (N-heterocyclic carbene)-type bridging ligands have been prepared and structurally and photophysically characterised. The identity of the NHC-unit of the bridging ligand was established unambiguously by means of X-ray structural analysis of a heterodinuclear ruthenium–silver complex. The photophysical data indicate ultrafast intersystem crossing into an emissive and a non-emissive triplet excited state after excitation of the ruthenium centre. Exceptionally high luminescence quantum yields of up to 39% and long lifetimes of up to 2 μs are some of the triplet excited state characteristics. Preliminary studies into the visible light driven photocatalytic hydrogen formation show no induction phase and constant turnover frequencies that are independent on the concentration of the photocatalyst. In conclusion this supports the notion of a stable assembly under photocatalytic conditions
Topology and hemodynamics of the cortical cerebrovascular system
The cerebrovascular system continuously delivers oxygen and energy substrates to the brain, which is one of the organs with the highest basal energy requirement in mammals. Discontinuities in the delivery lead to fatal consequences for the brain tissue. A detailed understanding of the structure of the cerebrovascular system is important for a multitude of (patho-)physiological cerebral processes and many noninvasive functional imaging methods rely on a signal that originates from the vasculature. Furthermore, neurodegenerative diseases often involve the cerebrovascular system and could contribute to neuronal loss. In this review, we focus on the cortical vascular system. In the first part, we present the current knowledge of the vascular anatomy. This is followed by a theory of topology and its application to vascular biology. We then discuss possible interactions between cerebral blood flow and vascular topology, before summarizing the existing body of the literature on quantitative cerebrovascular topology
The Future of IT in Peace and Security

Naturally, a textbook like this can only cover a selective part of research and a certain point in time.
