    Effects of the Chronic Use of Acetaminophen on Pregnant Rats

    Purpose: to evaluate the effects of acetaminophen on the pregnancy of female albino rats. Methods: forty pregnant rats were separated into four groups. All the animals received daily by gavage 1 ml of acetaminophen solution from the first day (day zero) until the 20th day of pregnancy: group I - only distilled water (control); groups II, III and IV, respectively, 125, 500 and 1,500 mg/kg body weight of acetaminophen dissolved in distilled water. The animals were weighed on days 0, 7, 15 and 20 of pregnancy. Results: our results showed that the rats that received the medication presented a reduction in weight when compared to the control group. The incidence of reabsorption of the embryos was 2.0, 3.5 and 7.0 times higher than in the control, in groups II, III and IV, respectively. Groups GII and GIV showed a clear reduction in the weight of the concepts. In GIV there was a 50% reduction in weight increase of fetuses and placentas when compared to the control, and 15.7% of external malformations were also found. Conclusions: the continuous use of acetaminophen should be avoided at doses higher than 70 mg/kg per during pregnancy.Objetivos: estudar a ação do acetaminofeno administrado durante toda a prenhez da rata albina. Métodos: 40 ratas albinas prenhes foram distribuídas ao acaso em quatro grupos numericamente iguais: GI - recebeu diariamente 1 ml de água destilada por gavagem (controle); GII, GIII e GIV foram tratadas, respectivamente, com 125, 500 e 1.500 mg/kg de peso corporal de acetaminofeno, dose única, diária, por gavagem, sempre diluído em 1 ml de água destilada, desde o dia zero até o 20º dia de prenhez. O ganho de peso das matrizes foi avaliado nos dias 0, 7, 15 e 20 de prenhez, sendo que no 20º dia todos os animais foram sacrificados. Resultados: nossos achados mostraram sempre menor peso entre as matrizes que receberam o medicamento, quando comparadas com o controle. A incidência de reabsorção dos embriões foi da ordem de 2,0, 3,5 e 7,0 vezes maior que no controle, considerando-se, respectivamente, os grupos GII, GIII e GIV. Os grupos GIII e GIV tiveram redução do peso dos conceptos. No GIV, houve redução de 50% no crescimento de fetos e placentas com relação ao controle; aqui, também foram encontradas 15,7% de malformações externas.     Effects of the topical administration of copaiba oil ointment (Copaifera langsdorffii) in skin flaps viability of rats

    PURPOSE: To evaluate the effects of copaiba oil ointment (Copaifera langsdorffii) on dorsal skin flaps in rats. METHODS: Adult male rats (n=30) were distributed into three groups of ten animals each, as follows: GC - control; GCA - absolute control and GT - treated with copaiba ointment. The rats were subjected to dorsal cutaneous skin flap surgery and the animals from the GC and GT received post-operative treatment for eight consecutive days. The animals from the GCA group did not receive treatment while the animals from the GC group received daily topical treatment of ointment without the active ingredient and the animals from the GT group were daily treated with 10% copaiba oil ointment. At the end of each experimental period the lesions were evaluated according to the percentage of necrotic area. Then, fragments from cranial, median and caudal parts were fixed in Boüin's solution and processed for paraffin embedding. The morphology of histological sections (5µm) was evaluated and the number of leucocytes, fibroblasts and blood vessels was also analyzed. The data obtained were submitted to ANOVA test complemented by Tukey-Kramer test (pGC and GT), while the morphology showed larger granulation tissue with bulky fibroblasts and collagen fibers more arranged in the GT group. The morphometry showed a significant higher number of blood vessels in the median and caudal parts (GT>GCA and GC), leucocytes in the cranial part (GT>GC>GCA), and also fibroblasts in the median (GT and GC> GCA) and caudal parts (GT>GC and GCA) (p<0.05).     The purpose of the present study was to evaluate the effects of acetylsalicylic acid (ASA) on the pregnancy of female albino rats. We used 60 pregnant female rats which were divided into six groups of ten cache. All the animals received daily by gavage, from the 5th (day zero) until the 20th day of pregnancy, 1 ml of the following: Group I - only distilled water (control); Group II - 0.2% aqueous solution of carboxymethylcellulose (vehicle); Groups III, IV, V and VI - 1, 10, 100 and 400 mg/kg body weight respectively, of ASA diluted in 0.2% carboxymethylcellulose solution. The animals were weighed on days 0, 7, 14 and 20 of pregnancy. Our results showed that the animals treated with 100 mg of ASA presented a reduction in the number of live newborns. The animals treated with 400 mg/kg/day presented not only a reduction in the number of live newborns but also decrease in maternal, newborn and placental weight.O objetivo deste trabalho foi avaliar os efeitos do ácido acetilsalicílico (AAS) sobre a prenhez da rata albina. Para tanto, utilizamos 60 ratas prenhes divididas ao acaso em seis grupos numericamente iguais. Todas receberam diariamente por gavagem o volume total de 1 ml, do 5º até o 20º dia de prenhez, com as seguintes características: Grupo I - somente água destilada (controle); Grupo II - solução aquosa de carboximetilcelulose 0,2% (veículo); Grupos III, IV, V e VI - respectivamente, 1, 10, 100 e 400 mg/kg de peso de AAS dissolvido em solução de carboximetilcelulose a 0,2%. Os pesos maternos foram anotados nos dias zero, 7º, 14º e 20º de prenhez. No 20º dia as matrizes foram sacrificadas.     Fluoxetine effects on periodontogenesis: histomorphometrical and immunohistochemical analyses in rats

    Reports have indicated that serotonin plays an important role in cell migration and differentiation during the organogenesis of several tissues, including the oral types. Administration of selective serotonin reuptake inhibitor (SSRI) drugs during pregnancy could affect the delivery of serotonin to embryonic tissues altering its development. Objective This study aimed to assess the effects of fluoxetine, a selective serotonin reuptake inhibitor, on the formation of the periodontal ligament during pregnancy and lactation in rat pups. Material and Methods Twelve pregnant rats of Wistar lineage were divided into four study groups. In the control group, 0.9% sodium chloride solution was administered orally, throughout the entire period of the 21 days of pregnancy (CG group) and in the CGL group, it was administrated during pregnancy and lactation (from day 1 of pregnancy to the 21st day after birth). Fluoxetine was administered orally at the dose of 20 mg/kg in a group treated during pregnancy only (FG group), and during pregnancy and lactation (FGL group). Histometrical, histochemical and immunohistochemical analysis of the maxillary first molar periodontium region of the 24 rat pups was made under light microscopy, and periodontal ligament collagen was qualitatively evaluated under a polarizing light microscope. Results The quantity of fibroblasts (p=0.006), osteoblasts (p=0.027) and cementoblasts (p=0.001) was reduced in pups from the rats that received fluoxetine during pregnancy and lactation. No alterations were seen in the collagen fibers. Conclusion These findings suggest that periodontal tissue may be sensitive to fluoxetine, and its interference in reducing periodontal cells depends on exposure time during lactation

    Efeito da ivermectina sobre a ciclicidade, gestação e desenvolvimento neonatal em ratos

    This work evaluates the estrous cycle, gestation and lactation of rats treated with ivermectin. Control (GI) rats received distilled water; and the remaining were treated during 45 days with ivermectin (GII=0.5; GIII=1.0; GIV=2.0; GV=4.0; GVI=8.0 and GVII=10.0 mg/kg) by the oral route, the drug being administered every three day, in a total of 15 administrations. Colpocytological examinations were then performed during 15 consecutive days. Four animals from each group were euthanized, and samples of ovaries and uteri were taken for histological evaluations. The remaining animals were mated and treated throughout the gestation period and further until the 15th postpartum day. Colpocytologic examinations revealed that the animals of groups II, III, IV, V, VI and VII presented a higher incidence of the estrus phase, in comparison with the control group (group I). Histopathologic studies showed that the groups treated with ivermectin presented an increased concentration of hyperplasic endometrial glands, whereas the morphology of the ovary was not altered. The treatment did not affect the gestation lenght, the number of newborns nor did it caused congenital mortality or newborns malformations. During lactation, there was a significant slowering of the body weight gain of the litter. It is concluded that the treatment of female rats with ivermectin increases the incidence of the estrus phase. In addition, a definite deleterious effect is exerted on nursing animals as revealed by the reduced body weight gain of the litter.O objetivo do presente trabalho foi avaliar o ciclo estral, gestação e lactação de ratas tratadas com ivermectina. 82 ratas albinas foram divididas em sete grupos. Os animais receberam água destilada (GI) ou diferentes doses de ivermectina, por via oral (GII=0,5; GIII=1,0; GIV=2,0; GV=4,0; GVI=8,0 e GVII=10,0 mg/kg). Os animais foram tratados por 45 dias, com ivermectina a cada três dias, totalizando 15 aplicações. Após esse período foi realizado exame colpocitológico durante 15 dias consecutivos. Ao final, quatro animais de cada grupo foram sacrificados e ovários e úteros retirados e processados para avaliação histológica. Os animais restantes foram acasalados e tratados com ivermectina, nas doses correspondentes a cada grupo, no 1º, 4º, 7º, 10º, 13º e 16º dias de gestação. Ao nascimento, os neonatos foram contados, analisados quanto à existência de defeitos congênitos, mortalidade e pesados até o 15º dia de lactação. Durante a lactação, as matrizes receberam novamente ivermectina no 1º, 4º, 7º, 10º e 13º dias. Nossos resultados mostraram que os animais dos grupos II, III, IV, V, VI e VII apresentaram maior incidência de estro em relação a GI. Quanto à histopatologia, os grupos tratados com a ivermectina apresentaram maior concentração de glândulas endometriais hiperplásicas. O tratamento não afetou tempo de gestação, número de neonatos, mortalidade ou defeitos congênitos. Na lactação observamos perda de peso na prole das matrizes tratadas com ivermectina. Pode-se concluir que a ivermectina, aumenta a incidência de estro e não deve ser indicada para uso em animais lactantes

    INTOXICAÇÃO POR DICLORVÓS E CIPERMETRINA EM BOVINOS EM MATO GROSSO – RELATO DE CASO

    Relata-se um surto de intoxicação por inseticida organofosforado e piretroide em vacas leiteiras no município de Cuiabá, Estado de Mato Grosso. Morreram onze vacas, de um total de 25 animais. Entre as principais manifestações clínicas observaram-se apatia, anorexia, sialorreia, diarreia de coloração escurecida, por vezes com estrias de sangue, tremores musculares, incoordenação e dificuldade respiratória. As lesões observadas à necropsia consistiram em edema pulmonar, enfisema subpleural e hemorragias de intensidade e formas diversas em vários órgãos. Não foram observadas alterações microscópicas significativas em nenhum dos animais. A pesquisa toxicológica em conteúdos de rúmen, retículo, omaso e abomaso resultaram negativas. Amostras de fígados e rins resultaram positivas para o grupo de inseticidas organofosforados e piretroides. PALAVRAS-CHAVES: Inseticidas, organofosforados, piretroides, vacas

    Neoangiogênese de retalhos cutâneos em ratos tratados com óleo de copaíba

    The aim of this work was to evaluate morphometric aspects of subdermic skin flap neoangiogenesis in rats treated with a 10% copaiba (Copaifera langsdorffii) oil ointment. The experiment design was a completely randomized, with three pharmacological resources. Ten replicates were used for each resource, and each animal was considered one experimental unit. Thirty Wistar rats were submitted to subdermic dorsal skin flap elevation and distributed into three groups: absolute control group, in which the animals did not receive any treatment; control group, in which the animals received topical treatment only with vehicle (glycerin and petroleum jelly), and treated group, in which the animals were treated daily with a 10% copaiba oil ointment. The rats were treated and observed for eight days after surgery. On the eighth day, the skin flap underwent a macroscopic analysis and fragments were collected from its cranial, medial and caudal portions for histopathological analysis. The morphometric analysis showed significant difference for proliferation of newly formed vessels in the middle and caudal portions of the skin flap in the treated group. Copaiba oil showed efficiency in the increase of neoangiogenesis of subdermic skin flaps in rats.O objetivo deste trabalho foi avaliar morfometricamente a neoangiogênese de retalhos cutâneos subdérmicos em ratos tratados com óleo de copaíba (Copaifera langsdorffii) em pomada a 10%. O delineamento experimental foi inteiramente casualizado, composto por três recursos farmacológicos. Foram utilizadas dez repetições para cada recurso, e cada animal foi considerado uma unidade experimental. Trinta ratos Wistar foram submetidos à elevação do retalho cutâneo dorsal subdérmico e distribuídos em três grupos: grupo controle absoluto, no qual os animais não receberam nenhum tratamento; grupo controle, no qual os animais receberam tratamento tópico diário com pomada com apenas veículo (glicerina e vaselina); e grupo tratado, no qual os animais foram tratados diariamente com óleo de copaíba em pomada a 10%. Os ratos foram tratados e observados por oito dias após o ato operatório. No oitavo dia de pós-operatório, realizou-se a análise macroscópica do retalho e foram coletados fragmentos das porções cranial, média e caudal do retalho cutâneo para análise histopatológica. A análise morfométrica mostrou diferença significativa para o número de novos vasos sanguíneos nas partes média e caudal do retalho cutâneo no grupo tratado. O óleo de copaíba mostra-se eficiente no aumento da neoangiogênese em retalhos cutâneos subdérmicos de ratos

    Natural and Experimental Poisoning of Cattle by Enterolobium contortisiliquum Pods (Fabaceae Mimosoideae) in Central-Western Brazil

    Enterolobium contortisiliquum pods are commonly identified as being the cause of abortions and photosensitivity in cattle. This paper describes the clinical and pathological aspects of a natural outbreak of hepatogenous photosensitization by the pods of E. contortisiliquum in Brazil and the results of experimental poisoning in three bovines. The history of natural poisoning was obtained at the site of the outbreak. Clinical examinations were carried out and hepatic enzymes were analyzed. A post-mortem examination was carried out at the outbreak location on a cow that had died. Several samples were collected from the animal's internal organs to carry out histopathological examinations. E. contortisiliquum pods were given to cows in order to reproduce the disease. The natural poisonings were characterized by photosensitization and aborted young. Enterolobium photosensitization was not experimentally reproduced, but the animals fell sick and recovered after 52 days

    Serological survey of toxoplasmosis and leptospirosis in neotropical wild mammals from Aracaju Zoo, Sergipe, Brazil

    Os zoológicos modernos são instituições destinadas à manutenção da fauna selvagem com o objetivo de promover a conservação, pesquisa científica, lazer, recreação e educação ambiental. A ampla variedade de espécies selvagens, vivendo em condições diferentes do seu habitat natural, representa um ambiente propício à disseminação de doenças, muitas delas zoonóticas. Devido à escassez de dados e à relevância dos mamíferos selvagens neste contexto epidemiológico, tanto na toxoplasmose, quanto na leptospirose, foi efetuado o inquérito sorológico para toxoplasmose e leptospirose em mamíferos selvagens neotropicais do Zoológico de Aracaju, Sergipe, Brasil. Para tanto foram colhidas amostras sanguíneas de 32 animais, adultos, de ambos os sexos incluindo: 14 macacos-prego (Cebus libidinosus), quatro macacos-prego-do-peito-amarelo (Cebus xanthosternus), três onças-suçuaranas (Puma concolor), uma onça-pintada (Pantheraonca), uma raposa (Cerdocyon thous), seis guaxinins (Procyon cancrivorus), dois quatis (Nasua nasua) e um papa-mel (Eira barbara). Para a pesquisa de anticorpos anti-Toxoplasma gondii foi utilizado o Teste de Aglutinação Modificada (MAT ³"1:25) e para pesquisa de anticorpos anti-Leptospira spp. foi utilizado o teste de Soroaglutinação Microscópica (ponto de corte ³1:100) com uma coleção de antígenos vivos que incluiu 24 variantes sorológicas de leptospiras patogênicas e duas leptospiras saprófitas. Dentre os 32 mamíferos, 17 (53,1%) apresentaram anticorpos anti-T. gondii e quatro (12,5%) foram positivos para anticorpos anti-Leptospira spp. De acordo com o sexo, 60% (9/15) dos machos e 47,1% (8/17) das fêmeas foram soropositivos para T. gondii e 26,7% (4/15) dos machos apresentaram anticorpos anti-Leptospira spp. Dos mamíferos que apresentaram anticorpos anti-T. gondii, 47% (8/17) nasceram no zoológico, 41,2% (7/17) foram oriundos de outras instituições e dois (11,8%) foram ) provenientes da natureza. Em relação aos quatro mamíferos soropositivos para Leptospira spp., três (75%) foram procedentes da natureza e um (25%) nasceu no zoológico. Este foi o primeiro inquérito sorológico de anticorpos anti-Leptospira spp. em primatas e carnívoros neotropicais em um zoológico do Nordeste do Brasil e descreveu pela primeira vez a ocorrência de anticorpos anti-T. gondii e anti-Leptospira spp. com sorovar mais provável Copenhageni no primata ameaçado de extinção macaco-prego-de-peito-amarelo (C. xanthosternus) em Aracaju, SEModern zoological gardens are institutions to wildlife maintenance, aiming its conservation, the performance of scientific research and leisure, recreation and environ-mental education activities. The variety of wild species living in conditions different from the one found in its natural habitats represents a propitious environment for diseases spread, specially the zoonotic ones. Due to data shortage and wild mammals' epidemio-logical relevance both for toxoplasmosis as for leptospirosis, this study aimed to determine the serological survey of toxoplasmosis and leptospirosis in Neotropical wild mammals, from Zoo of Aracaju, Sergipe, Brazil. Blood samples were collected from 32 wild mammals, adults and from both sexes: 14 wild bearded capuchins (Cebus libidinosus), four golden-bellied capuchins (Cebus xanthosternus), three pumas (Puma concolor), one jaguar (Pantheraonca), one crab-eating fox (Cerdocyon thous), six crab-eating raccoons (Procyon cancrivorus), two South American coatis (Nasua nasua), and one tayra (Eira barbara). Sera were tested to Toxoplasma gondii antibodies by the modified agglutination test (MAT e"1:25), and by the microscopic seroaglutination test (cut-off e"1:100) using 24 serovar of pathogenic leptospiras and two serovar of saprophyte leptospiras. Antibodies to T. gondii were found in 17 of 32 (53.1%) wild mammals, and antibodies to Leptospira spp. were found in 4 of 32 (12.5%) wild mammals. In relation to gender, 9 of 15 (60%) males, and 8 of 17 (47.1%) females had antibodies to Toxoplasma gondii, and 4 of 15 (26.7%) males had antibodies to Leptospira spp. Considering the origin of the seropositive animals to T. gondii, 8 of 17 (47%) were born in the zoo, 7 of 17 (41.2%) were from other zoos, and 2 (11.8%) were from the wild. Three of 4 (75%) mammals seropositive to Leptospira spp. were born in zoo, and 1 of 4 (25%) was from the wild. This is the first serological survey of Leptospira spp. in Neotropical and carnivores from Northeast zoo of Brazil, and the first time that antibodies to T. gondii and Leptospira spp. (serovar Copenhageni) were observed in the threaten specie of primate golden-bellied capuchins (Cebus xanthosternus) in Aracaju, S
