    Empirical Study in Teaching First-Year Database Students

    This paper describes the authors’ experience in team teaching two occurrences of a first-level undergraduate ‘Database Applications’ module. An analysis of the two occurrences revealed a difference in attendance, marks, seminar participation and attitude. This paper attempts to identify some reasons for this difference as well as reviewing some of the teaching activities that were used in this module. It analyzes the success of these activities and the perception and feedback from each group of students, and some possible recommended changes to the module

    CAPTCHA Accessibility Study of Online Forums

    The rise of online forums has benefited disabled users, who take advantage of better communications and more inclusion into society. However, even with accessibility laws that are supposed to provide disabled people the same equal access as non-disabled users, sites have erected technical barriers, such as CAPTCHAs, that prevent users from taking full advantage of site capability. This study analyzes 150 online forums to determine if sites use CAPTCHAs, and what types are used. Each variety presents accessibility problems to disabled users and the results of the research show that most sites use text-based CAPTCHAs, but rarely provide alternatives that would help users with visual disabilities. The research presents alternatives that site designers may wish to consider in order to allow more disabled users to access their sites

    Oat phyA containing transgenic tobacco: an examination of oat phyA mRNA stability and the role of oat phyA in germination of tobacco seed

    The regulation of oat phytochrome A (phyA) mRNA stability and the photoregulation of germination of tobacco seeds containing the oat phyA gene was examined using oat phyA containing transgenic tobacco. Oat phyA mRNA has been previously shown to be a relatively unstable mRNA. The intent of this research was to identify elements within the phyA mRNA responsible for rendering the message unstable. The approach taken was to use tobacco transformed with either the full-length or truncated versions of the oat phyA gene and examine the half-lives of the resulting oat phyA mRNAs in transgenic tobacco. RNA blot analysis of oat phyA in both oats and in tobacco transformed with the oat phyA gene showed a pattern of oat phyA hybridizable fragments smaller than the full-length mRNA band. These fragments were thought to be degradation products indicative of a short-lived message. The half-lives of the truncated oat phyA mRNAs were found to be between 90 minutes and four hours. The full-length oat phyA mRNA had a half-life of about four hours in transgenic tobacco. Determination of the half-life of both truncated and full-length oat phyA mRNAs in transgenic tobacco suggests that the amount of oat phyA hybridizable fragments does not correlate with mRNA half-life. Germination of seeds from wild-type tobacco plants requires light. Approximately 20% of the seeds from transgenic tobacco plants containing the oat phyA gene and grown under fluorescent lighting were found to germinate in the dark. Seeds from transgenic tobacco grown under fluorescent and incandescent lighting did not germinate in darkness. A higher percentage of seeds from both wild-type and transgenic plants were found to germinate when imbibed in the presence of nitrate. About 50% of the seeds from transgenic plants grown in the presence of incandescent lighting and imbibed in the presence of nitrate germinated in darkness. The higher level of phytochrome in the transgenic seedlings promoted germination and was sensitive to the light conditions under which the parent plants were grown. These findings suggest that phyA plays a role in promoting germination in tobacco seeds

    The concept of cohesion, which is normally associated with software design, is commonly used to measure the degree to which elements of a module are related. Systems constructed adhering to the principle of cohesion are expected to be more maintainable. It is proposed in this research that it may be more advantageous to apply the principle of cohesion at an earlier phase of the software development life cycle, thus placing more responsibility on the analyst who has a better understanding of the business. This paper proposes the Cohesion-Based Requirements Set (CBRS) model for improved information system maintainability. Using the CBRS technique, one may be able to positively affect the overall maintainability of the resulting system by applying a synthesis or expansion approach when gathering requirements rather than using an approach based on analysis or reduction

    Injury Surveillance During Competitive Functional Fitness Racing Events

High-intensity functional training (HIFT) is an exercise training modality that has grown considerably in popularity over the last decade. More recently, competitive functional fitness racing events have emerged from HIFT, and aim to test athletes’ proficiency across a variety of movements, skills, and energy systems. Whilst the injury risk associated with HIFT has been shown to be low and comparable to other forms of recreational fitness activities, the injury risk associated with competitive functional fitness racing events is currently unknown. A prospective cohort design was used to record medical-attention injuries during two competitive functional fitness racing events, involving 1085 competitors. A total of 26 injuries were recorded over the two competitions, resulting in an injury incidence rate of 36 per 1000 competition hours (90% confidence limits [CL]: 26-50) and injury prevalence of 2.4% (90% CL: 1.6-3.2%). The shoulder (n=4) and hand (n=4) were the most commonly injured body locations. The incidence rate in male athletes was likelyhigher than female athletes (Rate Ratio [RR]: 1.87, 90% CL: 0.95-3.69). The injury incidence rate associated with competitive functional fitness racing events is higher than for HIFT training activities, though the injury prevalence is relatively low in comparison to other sporting activities. Further research is required to understand the burden of these injuries, and identify appropriate injury prevention strategies for this emerging sport

    Positioning Dispositions in Initial Teacher Education: An Action Research Approach

    In spite of debate, ambiguity, and tension around teacher dispositions, in the past over two decades, the place of dispositions in initial teacher education (ITE) has been widely supported among policy makers and researchers. Specifically, debate on whether dispositions are teachable has largely given way to action to foster dispositions. Adopting a two-cycle participatory action research design, this study explored ways to teach the first-year teacher candidates’ dispositions in an early childhood ITE programme in New Zealand. The intervention included eight focus dispositions and corresponding strategies to teach each focus disposition. Data collection methods included student self-assessment surveys, individual and focus group interviews with students and teaching staff, team meetings, and a variety of pedagogical documentation. Ethnographic content analysis generated three themes: legitimacy of the intervention, experiential orientation of the intervention, and effect of the intervention. The study exemplifies how dispositions intervention can be incorporated in ITE programmes

    Scrutinising the Final Judging Role in Assessment of Practicum in Early Childhood Initial Teacher Education in New Zealand

    Practicum is the cornerstone of teacher education. Triadic assessment of practicum is an established model in early childhood initial teacher education in New Zealand. Based on a phenomenographic study on assessment of practicum, this paper presents a new perspective on triadic assessment. Interview data are collected from 35 participants (20 associate teachers, 5 visiting lecturers, 10 student teachers) to obtain a specimen of collective conceptions of key stakeholders about triadic assessment. The analysis reveals three sets of logically related categories of conceptions (outcome spaces) that denote the tensions around the final judging role in assessment of practicum, in particular, visiting lecturers’ scheduled observation. Based on the findings from this study, we contemplate some significant modifications to the triadic assessment model which calls in question visiting lecturers’ final judging role in assessment of practicum

Although HGSOC is the most prevalent subtype, representing 70-80% of cases, a 2013 landmark study by Domcke et al. found that the most frequently used OC cell lines are not molecularly representative of this subtype. This raises the question, if not HGSOC, from which subtype do these cell lines derive? Indeed, non-HGSOC subtypes often respond poorly to chemotherapy; therefore, representative models are imperative for developing new targeted therapeutics.MethodsNon-negative matrix factorisation (NMF) was applied to transcriptomic data from 44 OC cell lines in the Cancer Cell Line Encyclopedia, assessing the quality of clustering into 2-10 groups. Epithelial OC subtypes were assigned to cell lines optimally clustered into five transcriptionally distinct classes, confirmed by integration with subtype-specific mutations. A transcriptional subtype classifier was then developed by trialling three machine learning algorithms using subtype-specific metagenes defined by NMF. The ability of classifiers to predict subtype was tested using RNA sequencing of a living biobank of patient-derived OC models.ResultsApplication of NMF optimally clustered the 44 cell lines into five transcriptionally distinct groups. Close inspection of orthogonal datasets revealed this five-cluster delineation corresponds to the five major OC subtypes. This NMF-based classification validates the Domcke et al. analysis, in identifying lines most representative of HGSOC, and additionally identifies models representing the four other subtypes. However, NMF of the cell lines into two clusters did not align with the dualistic model of OC and suggests this classification is an oversimplification. Subtype designation of patient-derived models by a random forest transcriptional classifier aligned with prior diagnosis in 76% of unambiguous cases. In cases where there was disagreement, this often indicated potential alternative diagnosis, supported by a review of histological, molecular and clinical features.ConclusionsThis robust classification informs the selection of the most appropriate models for all five histotypes. Following further refinement on larger training cohorts, the transcriptional classification may represent a useful tool to support the classification of new model systems of OC subtypes

    Residential Moving and Preventable Hospitalizations

    OBJECTIVES: To investigate the association between moving home in the first year of life and subsequent emergency admissions for potentially preventable hospitalizations. METHODS: We undertook a cohort analysis of linked anonymized data on 237 842 children in the Welsh Electronic Cohort for Children. We included children born in Wales between April 1, 1999 and December 31, 2008. The exposure was the number of residential moves from birth up to 1 year. The main outcome was emergency admissions for potentially preventable hospitalizations (PPH) between the age of 1 and 5 years. RESULTS: After adjustment for confounders, we identified that moving home frequently in the first year of life was associated with an increased risk of emergency PPH between the ages of 1 and 5 when compared with not moving. We found significant differences associated with ≥2 moves for the following: ear, nose, and throat infections (incidence risk ratio [IRR], 1.44; 95% confidence interval [CI], 1.29–1.61); convulsions/epilepsy (IRR, 1.58; 95% CI, 1.23–2.04); injuries (IRR, 1.33; 95% CI, 1.18–1.51); dehydration/gastroenteritis (IRR, 1.51; 95% CI, 1.21–1.88); asthma (IRR, 1.61; 95% CI, 1.19–2.16); influenza/pneumonia (IRR, 1.15; 95% CI, 1.00–1.32); and dental conditions (IRR, 1.30; 95% CI, 1.03–1.64) for ≥1 moves. CONCLUSIONS: Children who move home in the first year of life are at substantially increased risk of emergency admissions for PPH in early childhood. Additional research that focuses on enhancing health and social support services for highly mobile families, educating parents about safety risks, and improving housing quality is warranted

    Pathway and network-based analysis of genome-wide association studies in multiple sclerosis

    Genome-wide association studies (GWAS) testing several hundred thousand SNPs have been performed in multiple sclerosis (MS) and other complex diseases. Typically, the number of markers in which the evidence for association exceeds the genome-wide significance threshold is very small, and markers that do not exceed this threshold are generally neglected. Classical statistical analysis of these datasets in MS revealed genes with known immunological functions. However, many of the markers showing modest association may represent false negatives. We hypothesize that certain combinations of genes flagged by these markers can be identified if they belong to a common biological pathway. Here we conduct a pathway-oriented analysis of two GWAS in MS that takes into account all SNPs with nominal evidence of association (P < 0.05). Gene-wise P-values were superimposed on a human protein interaction network and searches were conducted to identify sub-networks containing a higher proportion of genes associated with MS than expected by chance. These sub-networks, and others generated at random as a control, were categorized for membership of biological pathways. GWAS from eight other diseases were analyzed to assess the specificity of the pathways identified. In the MS datasets, we identified sub-networks of genes from several immunological pathways including cell adhesion, communication and signaling. Remarkably, neural pathways, namely axon-guidance and synaptic potentiation, were also over-represented in MS.
