    An Analysis of Electronic Data as a Type of Legal Evidence

    With the popularity and development of the Internet, E-commerce gradually spearhead the international trade, and become the main medium of trade. The application and development of E-commerce poses a severe challenge toward the traditional legal system, and bring about lots of new legal problems, especially worrisome is the evidence of E-commerce, which constitutes the major stumbling blocks in the development of international E-commerce. Simultaneously, the evidence of electronic contract remains a new problem in China’s Evidence Law. All along, views concerning the legal status of electronic contract vary greatly in theoretical circle, what’s more, its application is comparatively chaotic. Therefore, the evidence of electronic contract has become a pressing problem. In view of the sophistication of classification of electronic contract evidence and its peculiarity, the author deems it necessary to think of the evidence of the electronic contract as a brand-new independent category, so as to further facilitate the development of E-commerce in international trade

    GENERATION OF CRACKS IN HIGHWAY EMBANKMENT ON BLACK COTTON SOIL

    This research revealed the crack generation of the highway embankment from the water losing shrinkage of the wet black cotton soil (BCS), which is a type of soil with high swell-shrink potential. The road seepage meter was used to test the permeability of filling materials, which was used to replace BCS. The moisture content and embankment deflection of BCS foundation were measured after the rainy season. Based on the coupled consolidation theory for unsaturated soil, the change in additional tension stress of the embankment induced by water loss shrinkage of BCS was simulated by Abaqus. The results indicated that the rainfall seeped into the foundation through highly permeable refill materials to result in BCS expansion and decrease the embankment strength. After the rainy season, the additional tensile stress caused by water loss shrinkage of BCS induces cracking of highway embankment, and the maximum cracking depth often appears at the shoulder of highway. The deep and wide cracks are easy to appear in the low embankment constructed on a thick BCS foundation under strong evaporation

    Small RNA interference-mediated gene silencing of heparanase abolishes the invasion, metastasis and angiogenesis of gastric cancer cells

    <p>Abstract</p> <p>Background</p> <p>Heparanase facilitates the invasion and metastasis of cancer cells, and is over-expressed in many kinds of malignancies. Our studies indicated that heparanase was frequently expressed in advanced gastric cancers. The aim of this study is to determine whether silencing of heparanase expression can abolish the malignant characteristics of gastric cancer cells.</p> <p>Methods</p> <p>Three heparanase-specific small interfering RNA (siRNAs) were designed, synthesized, and transfected into cultured gastric cancer cell line SGC-7901. Heparanase expression was measured by RT-PCR, real-time quantitative PCR and Western blot. Cell proliferation was detected by MTT colorimetry and colony formation assay. The <it>in vitro </it>invasion and metastasis of cancer cells were measured by cell adhesion assay, scratch assay and matrigel invasion assay. The angiogenesis capabilities of cancer cells were measured by tube formation of endothelial cells.</p> <p>Results</p> <p>Transfection of siRNA against 1496-1514 bp of encoding regions resulted in reduced expression of heparanase, which started at 24 hrs and lasted for 120 hrs post-transfection. The siRNA-mediated silencing of heparanase suppressed the cellular proliferation of SGC-7901 cells. In addition, the <it>in vitro </it>invasion and metastasis of cancer cells were attenuated after knock-down of heparanase. Moreover, transfection of heparanase-specific siRNA attenuated the <it>in vitro </it>angiogenesis of cancer cells in a dose-dependent manner.</p> <p>Conclusions</p> <p>These results demonstrated that gene silencing of heparanase can efficiently abolish the proliferation, invasion, metastasis and angiogenesis of human gastric cancer cells <it>in vitro</it>, suggesting that heparanase-specific siRNA is of potential values as a novel therapeutic agent for human gastric cancer.</p

    MicroRNA Transcriptomic Analysis of Heterosis during Maize Seed Germination

    Heterosis has been utilized widely in the breeding of maize and other crops, and plays an important role in increasing yield, improving quality and enhancing stresses resistance, but the molecular mechanism responsible for heterosis is far from clear. To illustrate whether miRNA-dependent gene regulation is responsible for heterosis during maize germination, a deep-sequencing technique was applied to germinating embryos of a maize hybrid, Yuyu22, which is cultivated widely in China and its parental inbred lines, Yu87-1 and Zong3. The target genes of several miRNAs showing significant expression in the hybrid and parental lines were predicted and tested using real-time PCR. A total of 107 conserved maize miRNAs were co-detected in the hybrid and parental lines. Most of these miRNAs were expressed non-additively in the hybrid compared to its parental lines. These results indicated that miRNAs might participate in heterosis during maize germination and exert an influence via the decay of their target genes. Novel miRNAs were predicted follow a rigorous criterion and only the miRNAs detected in all three samples were treated as a novel maize miRNA. In total, 34 miRNAs belonged to 20 miRNA families were predicted in germinating maize seeds. Global repression of miRNAs in the hybrid, which might result in enhanced gene expression, might be one reason why the hybrid showed higher embryo germination vigor compared to its parental lines

    Identification of QTLs for Arsenic Accumulation in Maize (Zea mays L.) Using a RIL Population

    The Arsenic (As) concentration in different tissues of maize was analyzed using a set of RIL populations derived from an elite hybrid, Nongda108. The results showed that the trend of As concentration in the four measured tissues was leaves>stems>bracts>kernels. Eleven QTLs for As concentration were detected in the four tissues. Three QTLs for As concentration in leaves were mapped on chromosomes 1, 5, and 8, respectively. For As concentration in the bracts, two QTLs were identified, with 9.61% and 10.03% phenotypic variance. For As concentration in the stems, three QTLs were detected with 8.24%, 14.86%, and 15.23% phenotypic variance. Three QTLs were identified for kernels on chromosomes 3, 5, and 7, respectively, with 10.73%, 8.52%, and 9.10% phenotypic variance. Only one common chromosomal region between SSR marker bnlg1811 and umc1243 was detected for QTLs qLAV1 and qSAC1. The results implied that the As accumulation in different tissues in maize was controlled by different molecular mechanism. The study demonstrated that maize could be a useful plant for phytoremediation of As-contaminated paddy soil, and the QTLs will be useful for selecting inbred lines and hybrids with low As concentration in their kernels

    The water lily genome and the early evolution of flowering plants

    Water lilies belong to the angiosperm order Nymphaeales. Amborellales, Nymphaeales and Austrobaileyales together form the so-called ANA-grade of angiosperms, which are extant representatives of lineages that diverged the earliest from the lineage leading to the extant mesangiosperms1–3. Here we report the 409-megabase genome sequence of the blue-petal water lily (Nymphaea colorata). Our phylogenomic analyses support Amborellales and Nymphaeales as successive sister lineages to all other extant angiosperms. The N. colorata genome and 19 other water lily transcriptomes reveal a Nymphaealean whole-genome duplication event, which is shared by Nymphaeaceae and possibly Cabombaceae. Among the genes retained from this whole-genome duplication are homologues of genes that regulate flowering transition and flower development. The broad expression of homologues of floral ABCE genes in N. colorata might support a similarly broadly active ancestral ABCE model of floral organ determination in early angiosperms. Water lilies have evolved attractive floral scents and colours, which are features shared with mesangiosperms, and we identified their putative biosynthetic genes in N. colorata. The chemical compounds and biosynthetic genes behind floral scents suggest that they have evolved in parallel to those in mesangiosperms.     Stance phraseology in academic discourse: cross-disciplinary variation in authors’ presence

    This study aims to investigate the stance phrases used by agricultural science and economics scholars to establish their identity and authority in academic discourse. Adopting a corpus-driven approach, this study first retrieved 2- to 5- grams from two self-built corpora consisting of agriculture and economics research articles and then classified them functionally by following the three metafunctions in Systemic Functional Linguistics (SFL). Statistical comparisons show that agriculture researchers use cognitive, attitude, and hedging phrases more frequently, while the economics corpus contains significantly more evidentiality phrases. This paper contributes to the categorization of stance phrases by synthesizing and refining existing stance classifications and incorporating the reason-oriented dimension. The findings can enrich our understanding of disciplinary variation in academic discourse, and can inform academic writing pedagogy with respect to curriculum design and materials developmentEl objetivo de este artículo es analizar las colocaciones que emplean los investigadores de Ingeniería Agronómica y Economía para marcar su posición (stance) como autores, y, así, establecer su identidad y autoridad en el discurso académico. Desde un enfoque dirigido por corpus o corpus-driven, se han recuperado en primer lugar los n-gramas de orden 2, 3, 4 y 5 de dos corpus propios compuestos por artículos científicos de Ingeniería Agronómica y Economía y, posteriormente, se han clasificado con criterios funcionales de acuerdo con las tres metafunciones de la Lingüística Sistémico-Funcional. A través de comparaciones estadísticas, se ha evidenciado que los investigadores de Ingeniería Agronómica utilizan más colocaciones de naturaleza cognitiva, actitudinal y mitigadora con mayor frecuencia, mientras que el corpus de artículos de Economía contiene un número significativamente mayor de colocaciones de carácter evidencial. Asimismo, este artículo presenta una aportación respecto a la categorización de las colocaciones que permiten marcar la posición (stance) del autor, ya que sintetiza y mejora las clasificaciones ya existentes e incorpora una dimensión de naturaleza racional dentro del concepto de stance. Los resultados obtenidos enriquecen nuestra comprensión de la variación disciplinar en el discurso académico y pueden ser de gran utilidad para la didáctica de la escritura académica en relación con el diseño del currículum y el desarrollo de materiale

    Remote Sensing Based Detection of Crop Phenology for Agricultural Zones in China Using a New Threshold Method

    Abstract: In recent years, the use of high temporal resolution satellite data has been emerging as an important tool to study crop phenology. Most methods to detect phenological events based on satellite data use thresholds to identify key events in the lifecycle of the crop. In this study, a new method was used to define such thresholds for identifying the start and end of the growing season (SOS/EOS) for 43 different agricultural zones in China. The method used 2000–2003 NOAA Advanced Very High Resolution Radiometer (AVHRR) satellite data with a spatial resolution of eight kilometers and a temporal resolution of 15 days. Following data pre-processing, time series for the normalized difference vegetation index (NDVI or N), slope of the NDVI curve (S), and difference (D) between the NDVI value and a base NDVI value for bare land without snow were constructed. For each zone, an optimal set of threshold values for N, D, and S was determined, based on the remote sensing data and observed SOS/EOS data for 2003 at 261 agro-meteorological stations. Results were verified by comparing the accuracy of the new proposed NDS threshold method with the results of three other methods for SOS/EOS detection with remote sensing data.
