    Genome-Wide Association Study of Lung Adenocarcinoma in East Asia and Comparison With a European Population

    Lung adenocarcinoma is the most common type of lung cancer. Known risk variants explain only a small fraction of lung adenocarcinoma heritability. Here, we conducted a two-stage genome-wide association study of lung adenocarcinoma of East Asian ancestry (21,658 cases and 150,676 controls; 54.5% never-smokers) and identified 12 novel susceptibility variants, bringing the total number to 28 at 25 independent loci. Transcriptome-wide association analyses together with colocalization studies using a Taiwanese lung expression quantitative trait loci dataset (n = 115) identified novel candidate genes, including FADS1 at 11q12 and ELF5 at 11p13. In a multi-ancestry meta-analysis of East Asian and European studies, four loci were identified at 2p11, 4q32, 16q23, and 18q12. At the same time, most of our findings in East Asian populations showed no evidence of association in European populations. In our studies drawn from East Asian populations, a polygenic risk score based on the 25 loci had a stronger association in never-smokers vs. individuals with a history of smoking (Pinteraction = 0.0058). These findings provide new insights into the etiology of lung adenocarcinoma in individuals from East Asian populations, which could be important in developing translational applications

    Omecamtiv mecarbil in chronic heart failure with reduced ejection fraction, GALACTIC‐HF: baseline characteristics and comparison with contemporary clinical trials

    Aims: The safety and efficacy of the novel selective cardiac myosin activator, omecamtiv mecarbil, in patients with heart failure with reduced ejection fraction (HFrEF) is tested in the Global Approach to Lowering Adverse Cardiac outcomes Through Improving Contractility in Heart Failure (GALACTIC‐HF) trial. Here we describe the baseline characteristics of participants in GALACTIC‐HF and how these compare with other contemporary trials. Methods and Results: Adults with established HFrEF, New York Heart Association functional class (NYHA) ≥ II, EF ≤35%, elevated natriuretic peptides and either current hospitalization for HF or history of hospitalization/ emergency department visit for HF within a year were randomized to either placebo or omecamtiv mecarbil (pharmacokinetic‐guided dosing: 25, 37.5 or 50 mg bid). 8256 patients [male (79%), non‐white (22%), mean age 65 years] were enrolled with a mean EF 27%, ischemic etiology in 54%, NYHA II 53% and III/IV 47%, and median NT‐proBNP 1971 pg/mL. HF therapies at baseline were among the most effectively employed in contemporary HF trials. GALACTIC‐HF randomized patients representative of recent HF registries and trials with substantial numbers of patients also having characteristics understudied in previous trials including more from North America (n = 1386), enrolled as inpatients (n = 2084), systolic blood pressure < 100 mmHg (n = 1127), estimated glomerular filtration rate < 30 mL/min/1.73 m2 (n = 528), and treated with sacubitril‐valsartan at baseline (n = 1594). Conclusions: GALACTIC‐HF enrolled a well‐treated, high‐risk population from both inpatient and outpatient settings, which will provide a definitive evaluation of the efficacy and safety of this novel therapy, as well as informing its potential future implementation

    Cooperative Guidance Law for the Mother-Cabin of the Anti-UAV Cluster Mother-Son Missile

    This article investigates a novel operational pattern for intercepting UAV clusters over the range of middle and far distances using mother-son missiles. To address the guidance issue of the mother-cabin in the operational pattern, a special cooperative guidance law, which takes into account acceleration constraint and satisfies the constraints of impact time, speed, and zero line of sight angle (LOS), is proposed. Based on consistency theory and sliding mode control theory, the proposed cooperative guidance law is specifically designed for the mother-cabin of the mother-son missile. This approach offers several advantages, including a simple structure and smooth controller output, and smooth flight trajectory. Finally, numerical simulations are presented to demonstrate the effectiveness and applicability of the cooperative guidance law

    A new Brachiopod fauna from the early to middle permian of Southern Qinghai Province, Northwest China

    A new brachiopod fauna is described from the Early and Middle Permian of Zadoi and Zhidoi counties, southern Qinghai (Changdu block), northwest China. This fauna includes 13 species in nine genera with Spinomarginifera concentrica n. sp. and Transennatia waterhousei n. sp. The Early to Middle Permian brachiopod fauna from southern Qinghai is very similar to the contemporary Cathaysian faunas of South China with which the new fauna shares 70 per cent of its species. On the other hand, the Qinghai brachiopods also demonstrate a significant link with the Permian brachiopod fauna of the Sino-Mongolian-Japanese Province in northeast China, as suggested by Marginifera septentrionalis and Attenuatella. In particular, the occurrence of the bipolar brachiopod genus Attenuatella suggests that southern Qinghai may have played an important role as a biogeographic stepping stone in the marine faunal interchanges between the northern and southern hemispheres during the Early and Middle Permian.<br /

    Interleukin-34 protects against sepsis in mice by regulating CXCL1/CCL2 immune response

    Purpose: To study the protective effect of interleukin-34 (IL-34) against sepsis in mice, and the mechanism involved. Methods: Ninety healthy male mice were selected and assigned to sham, model and recombinant IL-34 protein groups. The activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were assayed. Moreover, histopathological changes in lung, liver and kidney were recorded, and levels of C-X-C Motif Chemokine Ligand 1 (CXCL1) and C-C Motif Chemokine Ligand 2 (CCL2) in each group of mice were measured. Results: Peritoneal lavage fluid and serum concentrations of AST, ALT, LDH, CXCL1 and CCL2 were significantly elevated, relative to sham mice (p &lt; 0.05). Mice survival in the drug group was markedly increased from day 1 to day 5; also, serum ALT, LDH and AST were significantly reduced, while CXCL1 and CCL2 concentrations in serum and peritoneal lavage fluid were increased, relative to model mice (p &lt; 0.05). Conclusion: IL-34 improves survival of septic mice by inducing CXCL1/CCL2 immune response, resulting in a protective effect on the airway. Thus, the CXCL1/CCL2 pathway mediated by IL-34 may be useful in the development of drugs for the treatment of sepsis
