    TRPM8 mechanism of autonomic nerve response to cold in respiratory airway

    Breathing cold air without proper temperature exchange can induce strong respiratory autonomic responses including cough, airway constriction and mucosal secretion, and can exacerbate existing asthma conditions and even directly trigger an asthma attack. Vagal afferent fiber is thought to be involved in the cold-induced respiratory responses through autonomic nerve reflex. However, molecular mechanisms by which vagal afferent fibers are excited by cold remain unknown. Using retrograde labeling, immunostaining, calcium imaging, and electrophysiological recordings, here we show that a subpopulation of airway vagal afferent nerves express TRPM8 receptors and that activation of TRPM8 receptors by cold excites these airway autonomic nerves. Thus activation of TRPM8 receptors may provoke autonomic nerve reflex to increase airway resistance. This putative autonomic response may be associated with cold-induced exacerbation of asthma and other pulmonary disorders, making TRPM8 receptors a possible target for prevention of cold-associated respiratory disorders

    Three-dimensional topological structures and formation processes of dislocations in Au nanowire under tension loading

    Dislocation is an important microstructural configuration in most crystalline materials, but it is still difficult for experiments to detect its local atomic structures and formation processes. Molecular dynamics simulation provides a powerful tool to investigate the dislocation by means of some geometrical structural analysis methods, but most of these methods cannot accurately distinguish the nuances of local configurations. Herein, we propose a new microstructural analysis method of cluster-type-index method (CTIM). The three-dimensional (3D) topological structures and formation processes of dislocations during tensioning Au nanowire are illuminated by means of CTIM. It is found that the favourite local atomic structures in dislocations are the defective FCC clusters represented by the CTIM indexes (13, 3/1421 2/1431 2/1441 4/1541 2/1661). The vector analysis method based on the accumulation of lattice distortions cannot identified the aggregation of disordered atoms in the initial nucleation of dislocation. In the formation processes of dislocation atoms, three trajectories of T1: FCC→(13, 3/1421 2/1431 2/1441 4/1541 2/1661), T2: FCC→(12, 2/1311 1/1411 9/1421) and T3: FCC→(11, 4/1311 7/1421) are favorite. The dislocation atoms following the T1 trajectory are mainly located at the core of dislocations, while that following the T2 and T3 trajectories is at the front of dislocations. CTIM would provide an effective tool to investigate the defective structures and their interactions in nanocrystalline structures

    A Fast Infrared Radiative Transfer Model for Overlapping Clouds

    A fast infrared radiative transfer model (FIRTM2) appropriate for application to both single-layered and overlapping cloud situations is developed for simulating the outgoing infrared spectral radiance at the top of the atmosphere (TOA). In FIRTM2 a pre-computed library of cloud reflectance and transmittance values is employed to account for one or two cloud layers, whereas the background atmospheric optical thickness due to gaseous absorption can be computed from a clear-sky radiative transfer model. FIRTM2 is applicable to three atmospheric conditions: 1) clear-sky, 2) single-layered ice or water cloud, and 3) two simultaneous cloud layers in a column (e.g., ice cloud overlying water cloud). Moreover, FIRTM2 outputs the derivatives (i.e., Jacobians) of the TOA brightness temperature with respect to cloud optical thickness and effective particle size. Sensitivity analyses have been carried out to assess the performance of FIRTM2 for two spectral regions, namely the longwave (LW) band (587.3 - 1179.5/cm) and the short-to-medium wave (SMW) band (1180.1 - 2228.9/cm). The assessment is carried out in terms of brightness temperature differences (BTD) between FIRTM2 and the well-known discrete ordinates radiative transfer model (DISORT), henceforth referred to as BTD (F-D). The BTD (F-D) values for single-layered clouds are generally less than 0.8 K. For the case of two cloud layers (specifically ice cloud over water cloud), the BTD(F-D) values are also generally less than 0.8 K except for the SMW band for the case of a very high altitude (>15 km) cloud comprised of small ice particles. Note that for clear-sky atmospheres, FIRTM2 reduces to the clear-sky radiative transfer model that is incorporated into FIRTM2, and the errors in this case are essentially those of the clear-sky radiative transfer model

    Down-regulation of miR-27a might inhibit proliferation and drug resistance of gastric cancer cells

    <p>Abstract</p> <p>Aims</p> <p>Here we aimed to firstly investigate the role of miR-27a in proliferation and multidrug resistance of gastric cancer cells.</p> <p>Methods</p> <p>The role of miR-27a in gastric cancer cells was detected using MTT assay, soft agar assay, flow cytometry assay, nude mice assay, real-time PCR, western blot and reporter gene assay, etc.</p> <p>Results</p> <p>Down-regulation of miR-27a could inhibit porliferation of gastric cancer cells in vitro and in vivo. Down-regulation of miR-27a could also confer sensitivity of drugs on gastric cancer cells, and might increase accumulation and decrease releasing amount of adriamycin in gastric cancer cells. Down-regulation of miR-27a could significantly decrease the expression of P-glycoprotein and the transcriptional activity of cyclin D1, and up-regulate the expression of p21.</p> <p>Conclusions</p> <p>MiR-27a might play important roles in porliferation and drug resistance of gastric cancer. MiR-27a might be considered as a useful target for cancer therapy.</p

    A simple proof of Perelman's collapsing theorem for 3-manifolds

    We will simplify earlier proofs of Perelman's collapsing theorem for 3-manifolds given by Shioya-Yamaguchi and Morgan-Tian. Among other things, we use Perelman's critical point theory (e.g., multiple conic singularity theory and his fibration theory) for Alexandrov spaces to construct the desired local Seifert fibration structure on collapsed 3-manifolds. The verification of Perelman's collapsing theorem is the last step of Perelman's proof of Thurston's Geometrization Conjecture on the classification of 3-manifolds. Our proof of Perelman's collapsing theorem is almost self-contained, accessible to non-experts and advanced graduate students.     N-Glycosylation of ß4 Integrin Controls the Adhesion and Motility of Keratinocytes

    α6ß4 integrin is an essential component of hemidesmosomes and modulates cell migration in wound healing and cancer invasion. To elucidate the role of N-glycosylation on ß4 integrin, we investigated keratinocyte adhesion and migration through the re-expression of wild-type or N-glycosylation-defective ß4 integrin (ΔNß4) in ß4 integrin null keratinocytes. N-glycosylation of ß4 integrin was not essential for the heterodimer formation of ß4 integrin with α6 integrin and its expression on a cell surface, but N-glycosylation was required for integrin-mediated cell adhesion and migration. Concomitantly with the reduction of ß4 integrin in the membrane microdomain, the intracellular signals of Akt and ERK activation were decreased in cells expressing ΔNß4 integrin. Forced cross-linking of ß4 integrin rescued the decreased ERK activation in ΔNß4 integrin-expressing cells to a similar extent in wild-type ß4 integrin-expressing cells. Surprisingly, compared with cells expressing wild-type ß4 integrin, an alternation in N-glycan structures expressed on epidermal growth factor receptor (EGFR), and the induction of a stronger association between EGFR and ß4 integrin were observed in ΔNß4 integrin-expressing cells. These results clearly demonstrated that N-glycosylation on ß4 integrin plays an essential role in keratinocyte cellular function by allowing the appropriate complex formation on cell surfaces

    Reduced expression of N-Myc downstream-regulated gene 2 in human thyroid cancer

    <p>Abstract</p> <p>Background</p> <p><it>NDRG</it>2 (N-Myc downstream-regulated gene 2) was initially cloned in our laboratory. Previous results have shown that <it>NDRG</it>2 expressed differentially in normal and cancer tissues. Specifically, <it>NDRG</it>2 mRNA was down-regulated or undetectable in several human cancers, and over-expression of <it>NDRG</it>2 inhibited the proliferation of cancer cells. <it>NDRG</it>2 also exerts important functions in cell differentiation and tumor suppression. However, it remains unclear whether <it>NDRG</it>2 participates in carcinogenesis of the thyroid.</p> <p>Methods</p> <p>In this study, we investigated the expression profile of human <it>NDRG</it>2 in thyroid adenomas and carcinomas, by examining tissues from individuals with thyroid adenomas (n = 40) and carcinomas (n = 35), along with corresponding normal tissues. Immunohistochemistry, quantitative RT-PCR and western blot methods were utilized to determine both the protein and mRNA expression status of Ndrg2 and c-Myc.</p> <p>Results</p> <p>The immunostaining analysis revealed a decrease of Ndrg2 expression in thyroid carcinomas. When comparing adenomas or carcinomas with adjacent normal tissue from the same individual, the mRNA expression level of <it>NDRG</it>2 was significantly decreased in thyroid carcinoma tissues, while there was little difference in adenoma tissues. This differential expression was confirmed at the protein level by western blotting. However, there were no significant correlations of <it>NDRG</it>2 expression with gender, age, different histotypes of thyroid cancers or distant metastases.</p> <p>Conclusion</p> <p>Our data indicates that <it>NDRG</it>2 may participate in thyroid carcinogenesis. This finding provides novel insight into the important role of <it>NDRG2 </it>in the development of thyroid carcinomas. Future studies are needed to address whether the down-regulation of <it>NDRG</it>2 is a cause or a consequence of the progression from a normal thyroid to a carcinoma.</p

    Geochemistry of soil gas in the seismic fault zone produced by the Wenchuan Ms 8.0 earthquake, southwestern China

    The spatio-temporal variations of soil gas in the seismic fault zone produced by the 12 May 2008 Wenchuan Ms 8.0 earthquake were investigated based on the field measurements of soil gas concentrations after the main shock. Concentrations of He, H2, CO2, CH4, O2, N2, Rn, and Hg in soil gas were measured in the field at eight short profiles across the seismic rupture zone in June and December 2008 and July 2009. Soil-gas concentrations of more than 800 sampling sites were obtained. The data showed that the magnitudes of the He and H2 anomalies of three surveys declined significantly with decreasing strength of the aftershocks with time. The maximum concentrations of He and H2 (40 and 279.4 ppm, respectively) were found in three replicates at the south part of the rupture zone close to the epicenter. The spatio-temporal variations of CO2, Rn, and Hg concentrations differed obviously between the north and south parts of the fault zone. The maximum He and H2 concentrations in Jun 2008 occurred near the parts of the rupture zone where vertical displacements were larger. The anomalies of He, H2, CO2, Rn, and Hg concentrations could be related to the variation in the regional stress field and the aftershock activity
