    Architecture and host interface of environmental chlamydiae revealed by electron cryotomography

    Chlamydiae comprise important pathogenic and symbiotic bacteria that alternate between morphologically and physiologically different life stages during their developmental cycle. Using electron cryotomography, we characterize the ultrastructure of the developmental stages of three environmental chlamydiae: Parachlamydia acanthamoebae, Protochlamydia amoebophila and Simkania negevensis. We show that chemical fixation and dehydration alter the cell shape of Parachlamydia and that the crescent body is not a developmental stage, but an artefact of conventional electron microscopy. We further reveal type III secretion systems of environmental chlamydiae at macromolecular resolution and find support for a chlamydial needle-tip protein. Imaging bacteria inside their host cells by cryotomography for the first time, we observe marked differences in inclusion morphology and development as well as host organelle recruitment between the three chlamydial organisms, with Simkania inclusions being tightly enveloped by the host endoplasmic reticulum. The study demonstrates the power of electron cryotomography to reveal structural details of bacteria–host interactions that are not accessible using traditional methods

    HD 101088, An Accreting 14 AU Binary in Lower Centaurus Crux With Very Little Circumstellar Dust

    We present high resolution (R=55,000) optical spectra obtained with MIKE on the 6.5 m Magellan Clay Telescope as well as Spitzer MIPS photometry and IRS low resolution (R~60) spectroscopy of the close (14 AU separation) binary, HD 101088, a member of the ~12 Myr old southern region of the Lower Centaurus Crux (LCC) subgroup of the Scorpius-Centaurus OB association. We find that the primary and/or secondary is accreting from a tenuous circumprimary and/or circumsecondary disk despite the apparent lack of a massive circumbinary disk. We estimate a lower limit to the accretion rate of > 1x10^-9 solar masses per year, which our multiple observation epochs show varies over a timescale of months. The upper limit on the 70 micron flux allows us to place an upper limit on the mass of dust grains smaller than several microns present in a circumbinary disk of 0.16 moon masses. We conclude that the classification of disks into either protoplanetary or debris disks based on fractional infrared luminosity alone may be misleading.Comment: 8 pages, 2 figures, ApJ accepte

    Radiometric Stability of the SABER Instrument

    The SABER instrument on the National Aeronautics and Space Administration Thermosphere‐Ionosphere‐Mesosphere Energetics and Dynamics satellite continues to provide a long‐term record of Earth\u27s stratosphere, mesosphere, and lower thermosphere. The SABER data are being used to examine long‐term changes and trends in temperature, water vapor, and carbon dioxide. A tacit, central assumption of these analyses is that the SABER instrument radiometric calibration is not changing with time; that is, the instrument is stable. SABER stratospheric temperatures and those derived from Global Positioning System Radio Occultation measurements are compared to examine SABER\u27s stability. Global Positioning System Radio Occultation measurements are inherently stable due to the accuracy and traceability of the measured phase delay rate to the Système Internationale definition of the second. Differences in global annual mean SABER and COSMIC lower stratospheric temperatures show little significant change with time in the 11 years spanning 2007–2017. From this analysis we infer that SABER temperatures are stable to better than 0.1 to 0.2 K per decade
