    Submersed Aquatic Vegetation in Chesapeake Bay: Sentinel Species in a Changing World

    Chesapeake Bay has undergone profound changes since European settlement. Increases in human and livestock populations, associated changes in land use, increases in nutrient loadings, shoreline armoring, and depletion of fish stocks have altered the important habitats within the Bay. Submersed aquatic vegetation (SAV) is a critical foundational habitat and provides numerous benefits and services to society. In Chesapeake Bay, SAV species are also indicators of environmental change because of their sensitivity to water quality and shoreline development. As such, SAV has been deeply integrated into regional regulations and annual assessments of management outcomes, restoration efforts, the scientific literature, and popular media coverage. Even so, SAV in Chesapeake Bay faces many historical and emerging challenges. The future of Chesapeake Bay is indicated by and contingent on the success of SAV. Its persistence will require continued action, coupled with new practices, to promote a healthy and sustainable ecosystem

    Establishing the baseline level of repetitive element expression in the human cortex

    Background: Although nearly half of the human genome is comprised of repetitive sequences, the expression profile of these elements remains largely uncharacterized. Recently developed high throughput sequencing technologies provide us with a powerful new set of tools to study repeat elements. Hence, we performed whole transcriptome sequencing to investigate the expression of repetitive elements in human frontal cortex using postmortem tissue obtained from the Stanley Medical Research Institute. Results: We found a significant amount of reads from the human frontal cortex originate from repeat elements. We also noticed that Alu elements were expressed at levels higher than expected by random or background transcription. In contrast, L1 elements were expressed at lower than expected amounts. Conclusions: Repetitive elements are expressed abundantly in the human brain. This expression pattern appears to be element specific and can not be explained by random or background transcription. These results demonstrate that our knowledge about repetitive elements is far from complete. Further characterization is required to determine the mechanism, the control, and the effects of repeat element expression

    GPI 2.0: Performance Evaluation of the Wavefront Sensor's EMCCD

    The Gemini Planet Imager (GPI) is a high contrast imaging instrument that aims to detect and characterize extrasolar planets. GPI is being upgraded to GPI 2.0, with several subsystems receiving a re-design to improve the instrument's contrast. To enable observations on fainter targets and increase stability on brighter ones, one of the upgrades is to the adaptive optics system. The current Shack-Hartmann wavefront sensor (WFS) is being replaced by a pyramid WFS with an low-noise electron multiplying CCD (EMCCD). EMCCDs are detectors capable of counting single photon events at high speed and high sensitivity. In this work, we characterize the performance of the HN\"u 240 EMCCD from N\"uv\"u Cameras, which was custom-built for GPI 2.0. The HN\"u 240 EMCCD's characteristics make it well suited for extreme AO: it has low dark current (<< 0.01 e-/pix/fr), low readout noise (0.1 e-/pix/fr at a gain of 5000), high quantum efficiency ( 90% at wavelengths from 600-800 nm; 70% from 800-900 nm), and fast readout (up to 3000 fps full frame). Here we present test results on the EMCCD's noise contributors, such as the readout noise, pixel-to-pixel variability and CCD bias. We also tested the linearity and EM gain calibration of the detector. All camera tests were conducted before its integration into the GPI 2.0 PWFS system.Comment: 16 pages, 14 figures.     Enhanced Food Anticipatory Activity Associated with Enhanced Activation of Extrahypothalamic Neural Pathways in Serotonin2C Receptor Null Mutant Mice

    The ability to entrain circadian rhythms to food availability is important for survival. Food-entrained circadian rhythms are characterized by increased locomotor activity in anticipation of food availability (food anticipatory activity). However, the molecular components and neural circuitry underlying the regulation of food anticipatory activity remain unclear. Here we show that serotonin2C receptor (5-HT2CR) null mutant mice subjected to a daytime restricted feeding schedule exhibit enhanced food anticipatory activity compared to wild-type littermates, without phenotypic differences in the impact of restricted feeding on food consumption, body weight loss, or blood glucose levels. Moreover, we show that the enhanced food anticipatory activity in 5-HT2CR null mutant mice develops independent of external light cues and persists during two days of total food deprivation, indicating that food anticipatory activity in 5-HT2CR null mutant mice reflects the locomotor output of a food-entrainable oscillator. Whereas restricted feeding induces c-fos expression to a similar extent in hypothalamic nuclei of wild-type and null mutant animals, it produces enhanced expression in the nucleus accumbens and other extrahypothalamic regions of null mutant mice relative to wild-type subjects. These data suggest that 5-HT2CRs gate food anticipatory activity through mechanisms involving extrahypothalamic neural pathways

    Determinants of circumcision and willingness to be circumcised by Rwandan men, 2010

    <p>Abstract</p> <p>Background</p> <p>Male Circumcision (MC) has been recommended as one of the preventive measures against sexual HIV transmission by the World Health Organization (WHO). Rwanda has adopted MC as recommended but the country is a non-traditionally circumcising society. The objective was to explore knowledge and perception of Rwandan men on Male Circumcision (MC) and to determine the factors associated with the willingness to be circumcised and to circumcise their sons.</p> <p>Methods</p> <p>This cross sectional study was conducted in 29 districts of Rwanda between January and March 2010. Data were collected using a structured questionnaire among men aged 15-59 years. The rate of MC was measured and its perception from respondents, and then the factors associated with the willingness to go for MC were analysed using multiple logistic regressions.</p> <p>Results</p> <p>A total of 1098 men were interviewed. Among respondents 17% (95% CI 14-19%) reported being circumcised. About three-quarter (72%) could define MC, but 37% of adolescent could not. Half of the participants were willing to get circumcised and 79% of men would accept circumcision for their sons. The main motivators for MC were its benefits in HIV/STI prevention (69%) and improving hygiene (49%). Being too old was the main reason (32%) reported by men reluctant to undergo MC and younger men were afraid of pain in particular those less than 19 years old (42%). The willingness to circumcise was significantly associated with younger age, living in the Eastern Province, marital status, and the knowledge of the preventive role of circumcision.</p> <p>Conclusions</p> <p>Adolescents and young adults were more willing to be circumcised. It is critical to ensure the availability of pain free services in order to satisfy the increasing demand for the scale up of MC in Rwanda.</p

    Essential Role of Chromatin Remodeling Protein Bptf in Early Mouse Embryos and Embryonic Stem Cells

    We have characterized the biological functions of the chromatin remodeling protein Bptf (Bromodomain PHD-finger Transcription Factor), the largest subunit of NURF (Nucleosome Remodeling Factor) in a mammal. Bptf mutants manifest growth defects at the post-implantation stage and are reabsorbed by E8.5. Histological analyses of lineage markers show that Bptf−/− embryos implant but fail to establish a functional distal visceral endoderm. Microarray analysis at early stages of differentiation has identified Bptf-dependent gene targets including homeobox transcriptions factors and genes essential for the development of ectoderm, mesoderm, and both definitive and visceral endoderm. Differentiation of Bptf−/− embryonic stem cell lines into embryoid bodies revealed its requirement for development of mesoderm, endoderm, and ectoderm tissue lineages, and uncovered many genes whose activation or repression are Bptf-dependent. We also provide functional and physical links between the Bptf-containing NURF complex and the Smad transcription factors. These results suggest that Bptf may co-regulate some gene targets of this pathway, which is essential for establishment of the visceral endoderm. We conclude that Bptf likely regulates genes and signaling pathways essential for the development of key tissues of the early mouse embryo

    A global experiment on motivating social distancing during the COVID-19 pandemic

    Finding communication strategies that effectively motivate social distancing continues to be a global public health priority during the COVID-19 pandemic. This cross-country, preregistered experiment (n = 25,718 from 89 countries) tested hypotheses concerning generalizable positive and negative outcomes of social distancing messages that promoted personal agency and reflective choices (i.e., an autonomy-supportive message) or were restrictive and shaming (i.e., a controlling message) compared with no message at all. Results partially supported experimental hypotheses in that the controlling message increased controlled motivation (a poorly internalized form of motivation relying on shame, guilt, and fear of social consequences) relative to no message. On the other hand, the autonomy-supportive message lowered feelings of defiance compared with the controlling message, but the controlling message did not differ from receiving no message at all. Unexpectedly, messages did not influence autonomous motivation (a highly internalized form of motivation relying on one’s core values) or behavioral intentions. Results supported hypothesized associations between people’s existing autonomous and controlled motivations and self-reported behavioral intentions to engage in social distancing. Controlled motivation was associated with more defiance and less long-term behavioral intention to engage in social distancing, whereas autonomous motivation was associated with less defiance and more short- and long-term intentions to social distance. Overall, this work highlights the potential harm of using shaming and pressuring language in public health communication, with implications for the current and future global health challenges
