Crime and Drugs : An Economic Approach
We present a model which ties together rational drug consumption, taxation, crime and other drug-related externalities. Drug control policy is addressed using an optimal tax framework. Consumption, possession and production of a drug may be prohibited, legalized or decriminalized. In all regimes illicit production of a drug may take place and drug-related crime occurs. We show that illicit drug production, the price elasticity of demand for a drug, the addictive nature of a drug, the effectiveness of drug enforcement strategies, and income distribution all influence optimal (second best) policy. Prohibition is contrasted with decriminalization and legalization, and where legalization yields a higher welfare than prohibition we show that this can be associated with greater drug-related crime and more drug addiction. Infectious bronchitis virus nonstructural protein 4 alone induces membrane pairing
Positive-strand RNA viruses, such as coronaviruses, induce cellular membrane rearrangements during replication to form replication organelles allowing for efficient viral RNA synthesis. Infectious bronchitis virus (IBV), a pathogenic avian of significant importance to the global poultry industry, has been shown to induce the formation of double membrane vesicles (DMVs), zippered endoplasmic reticulum (zER) and tethered vesicles, known as spherules. These membrane rearrangements are virally induced; however, it remains unclear which viral proteins are responsible. In this study, membrane rearrangements induced when expressing viral non-structural proteins (nsps) from two different strains of IBV were compared. Three non-structural transmembrane proteins, nsp3, nsp4, and nsp6, were expressed in cells singularly or in combination and the effects on cellular membranes investigated using electron microscopy and electron tomography. In contrast to previously studied coronaviruses, IBV nsp4 alone is necessary and sufficient to induce membrane pairing; however, expression of the transmembrane proteins together was not sufficient to fully recapitulate DMVs. This indicates that although nsp4 is able to singularly induce membrane pairing, further viral or host factors are required in order to fully assemble IBV replicative structures. This study highlights further differences in the mechanism of membrane rearrangements between members of the coronavirus family

A tad too high: Sensitivity to UV-B radiation may limit invasion potential of American bullfrogs (Lithobates catesbeianus) in the Pacific Northwest invasion range
Biological invasion potential can be strongly influenced by abiotic factors such as temperature, water availability, and solar radiation. Invasive species that possess phenotypically plastic traits can mediate impacts from these stressors, but may be unable to recognize and respond to dangerous levels in a novel environment. Understanding potential constraints on appropriate trait responses induced by abiotic stressors can aid in the management and control of important invaders. Our study explored tolerance and plastic trait response to UV-B radiation in an invasive anuran, the American bullfrog (Lithobates catesbeianus Shaw, 1802). We experimentally quantified larval mortality rates and color change responses across two larval size classes. In a second experiment, we investigated the potential for a correlated color change and behavioral (refuge use) response in the small size class. We predicted that individuals would respond to stressful and potentially harmful UV-B exposure rates with darkening of body coloration, and when refuge was available, a correlated defense strategy utilizing both color change and refuge. We found an increase in larval mortality across both size classes at UV-B exposure rates typical to both low and high elevation aquatic breeding sites (10-12 μW/cm² and 20-24 μW/cm² , respectively). Only bullfrog larvae in the small size class exhibited a darkening in body color when exposed to high UV-B treatments. Although this smaller size class did exhibit color plasticity, individuals did not correlate changes in body coloration with changes in refuge use. These results suggest ontogenetic differences (estimated by size class) in plastic color response to UV-B stress as well as constraints on behavioral use of refuge. These findings are important in understanding differences in bullfrog occupancy of breeding habitats across an elevational gradient, particularly in Oregon’s Cascade Mountain Range, where bullfrog distributions are currently limited at elevations above 1000 m.Keywords: Amphibian, Ultraviolet radiation, Color,

Genomic regions associated with muscularity in beef cattle differ in five contrasting cattle breeds

Linear type traits, which reflect the muscular characteristics of an animal, could provide insight into how, in some cases, morphologically very different animals can yield the same carcass weight. Such variability may contribute to differences in the overall value of the carcass since primal cuts vary greatly in price; such variability may also hinder successful genome-based association studies. Therefore, the objective of our study was to identify genomic regions that are associated with five muscularity linear type traits and to determine if these significant regions are common across five different breeds. Analyses were carried out using linear mixed models on imputed whole-genome sequence data in each of the five breeds, separately. Then, the results of the within-breed analyses were used to conduct an across-breed meta-analysis per trait.
Results
We identified many quantitative trait loci (QTL) that are located across the whole genome and associated with each trait in each breed. The only commonality among the breeds and traits was a large-effect pleiotropic QTL on BTA2 that contained the MSTN gene, which was associated with all traits in the Charolais and Limousin breeds. Other plausible candidate genes were identified for muscularity traits including PDE1A, PPP1R1C and multiple collagen and HOXD genes. In addition, associated (gene ontology) GO terms and KEGG pathways tended to differ between breeds and between traits especially in the numerically smaller populations of Angus, Hereford, and Simmental breeds. Most of the SNPs that were associated with any of the traits were intergenic or intronic SNPs located within regulatory regions of the genome.
Conclusions
The commonality between the Charolais and Limousin breeds indicates that the genetic architecture of the muscularity traits may be similar in these breeds due to their similar origins. Conversely, there were vast differences in the QTL associated with muscularity in Angus, Hereford, and Simmental. Knowledge of these differences in genetic architecture between breeds is useful to develop accurate genomic prediction equations that can operate effectively across breeds. Overall, the associated QTL differed according to trait, which suggests that breeding for a morphologically different (e.g. longer and wider versus shorter and smaller) more efficient animal may become possible in the future
Genomic Regions Associated With Skeletal Type Traits in Beef and Dairy Cattle Are Common to Regions Associated With Carcass Traits, Feed Intake and Calving Difficulty
Linear type traits describing the skeletal characteristics of an animal are moderately to strongly genetically correlated with a range of other performance traits in cattle including feed intake, reproduction traits and carcass merit; thus, type traits could also provide useful insights into the morphological differences among animals underpinning phenotypic differences in these complex traits. The objective of the present study was to identify genomic regions associated with five subjectively scored skeletal linear traits, to determine if these associated regions are common in multiple beef and dairy breeds, and also to determine if these regions overlap with those proposed elsewhere to be associated with correlated performance traits. Analyses were carried out using linear mixed models on imputed whole genome sequence data separately in 1,444 Angus, 1,129 Hereford, 6,433 Charolais, 8,745 Limousin, 1,698 Simmental, and 4,494 Holstein-Friesian cattle, all scored for the linear type traits. There was, on average, 18 months difference in age at assessment of the beef versus the dairy animals. While the majority of the identified quantitative trait loci (QTL), and thus genes, were both trait-specific and breed-specific, a large-effect pleiotropic QTL on BTA6 containing the NCAPG and LCORL genes was associated with all skeletal traits in the Limousin population and with wither height in the Angus. Other than that, little overlap existed in detected QTLs for the skeletal type traits in the other breeds. Only two QTLs overlapped the beef and dairy breeds; both QTLs were located on BTA5 and were associated with height in both the Angus and the Holstein-Friesian, despite the difference in age at assessment. Several detected QTLs in the present study overlapped with QTLs documented elsewhere that are associated with carcass traits, feed intake, and calving difficulty. While most breeding programs select for the macro-traits like carcass weight, carcass conformation, and feed intake, the higher degree of granularity with selection on the individual linear type traits in a multi-trait index underpinning the macro-level goal traits, presents an opportunity to help resolve genetic antagonisms among morphological traits in the pursuit of the animal with optimum performance metrics.</p
Remote Infrared Imaging of the Space Shuttle During Hypersonic Flight: HYTHIRM Mission Operations and Coordination
The Hypersonic Thermodynamic Infrared Measurements (HYTHIRM) project has been responsible for obtaining spatially resolved, scientifically calibrated in-flight thermal imagery of the Space Shuttle Orbiter during reentry. Starting with STS-119 in March of 2009 and continuing through to the majority of final flights of the Space Shuttle, the HYTHIRM team has to date deployed during seven Shuttle missions with a mix of airborne and ground based imaging platforms. Each deployment of the HYTHIRM team has resulted in obtaining imagery suitable for processing and comparison with computational models and wind tunnel data at Mach numbers ranging from over 18 to under Mach 5. This paper will discuss the detailed mission planning and coordination with the NASA Johnson Space Center Mission Control Center that the HYTHIRM team undergoes to prepare for and execute each mission
Three-dimensional photolithographic micropatterning: a novel tool to probe the complexities of cell migration
In order to independently study the numerous variables that influence cell movement, it will be
necessary to employ novel tools and materials that allow for exquisite control of the cellular
microenvironment. In this work, we have applied advanced 3D micropatterning technology, known as
two-photon laser scanning lithography (TP-LSL), to poly(ethylene glycol) (PEG) hydrogels modified with
bioactive peptides in order to fabricate precisely designed microenvironments to guide and
quantitatively investigate cell migration. Specifically, TP-LSL was used to fabricate cell adhesive PEGRGDS
micropatterns on the surface of non-degradable PEG-based hydrogels (2D) and in the interior of
proteolytically degradable PEG-based hydrogels (3D). HT1080 cell migration was guided down these
adhesive micropatterns in both 2D and 3D, as observed via time-lapse microscopy. Differences in cell
speed, cell persistence, and cell shape were observed based on variation of adhesive ligand, hydrogel
composition, and patterned area for both 2D and 3D migration. Results indicated that HT1080s migrate
faster and with lower persistence on 2D surfaces, while HT1080s migrating in 3D were smaller and
more elongated. Further, cell migration was shown to have a biphasic dependence on PEG-RGDS
concentration and cells moving within PEG-RGDS micropatterns were seen to move faster and with
more persistence over time. Importantly, the work presented here begins to elucidate the multiple
complex factors involved in cell migration, with typical confounding factors being independently
controlled. The development of this unique platform will allow researchers to probe how cells behave
within increasingly complex 3D microenvironments that begin to mimic specifically chosen aspects of
the in vivo landscape
Kepler-16: A Transiting Circumbinary Planet
We report the detection of a planet whose orbit surrounds a pair of low-mass
stars. Data from the Kepler spacecraft reveal transits of the planet across
both stars, in addition to the mutual eclipses of the stars, giving precise
constraints on the absolute dimensions of all three bodies. The planet is
comparable to Saturn in mass and size, and is on a nearly circular 229-day
orbit around its two parent stars. The eclipsing stars are 20% and 69% as
massive as the sun, and have an eccentric 41-day orbit. The motions of all
three bodies are confined to within 0.5 degree of a single plane, suggesting

Planetary Candidates Observed by Kepler. VIII. A Fully Automated Catalog With Measured Completeness and Reliability Based on Data Release 25
We present the Kepler Object of Interest (KOI) catalog of transiting
exoplanets based on searching four years of Kepler time series photometry (Data
Release 25, Q1-Q17). The catalog contains 8054 KOIs of which 4034 are planet
candidates with periods between 0.25 and 632 days. Of these candidates, 219 are
new and include two in multi-planet systems (KOI-82.06 and KOI-2926.05), and
ten high-reliability, terrestrial-size, habitable zone candidates. This catalog
was created using a tool called the Robovetter which automatically vets the
DR25 Threshold Crossing Events (TCEs, Twicken et al. 2016). The Robovetter also
vetted simulated data sets and measured how well it was able to separate TCEs
caused by noise from those caused by low signal-to-noise transits. We discusses
the Robovetter and the metrics it uses to sort TCEs. For orbital periods less
than 100 days the Robovetter completeness (the fraction of simulated transits
that are determined to be planet candidates) across all observed stars is
greater than 85%. For the same period range, the catalog reliability (the
fraction of candidates that are not due to instrumental or stellar noise) is
greater than 98%. However, for low signal-to-noise candidates between 200 and
500 days around FGK dwarf stars, the Robovetter is 76.7% complete and the
catalog is 50.5% reliable. The KOI catalog, the transit fits and all of the
simulated data used to characterize this catalog are available at the NASA
