    Polar Coordinate Farm Bot Final Project Report

    FarmBot Inc. is a local startup that is advancing precision agriculture through automation and open source technology. The original FarmBot model, FarmBot Genesis, operates over a rectangular area of 3 x 1.5 meters. It plants, waters, weeds and grows produce autonomously using CNC technology. Our project sponsor and inventor of FarmBot, Rory Aronson, tasked our team with developing and testing a polar coordinate farmbot capable of performing on par with the Cartesian farmbot in growing a circular vegetable garden in a user’s backyard. A successful project was defined by meeting specific customer requirements and engineering specifications predetermined and discussed with our sponsor. The customer requirements include: a central pivot, accurate planting, scalable farming area, off the shelf parts, and easy customization by the user. Some of the most important polar farmbot engineering specifications include a minimum farmable area of 4.5 m2 , maximum cost of $2200, rotational range of at least 360 degrees, and an average velocity of .5 meters per second

    Recent Development of Graphene-Based Cathode Materials for Dye-Sensitized Solar Cells

    Dye-sensitized solar cells (DSSCs) have attracted extensive attention for serving as potential low-cost alternatives to silicon-based solar cells. As a vital role of a typical DSSC, the counter electrode (CE) is generally employed to collect electrons via the external circuit and speed up the reduction reaction of I3- to I- in the redox electrolyte. The noble Pt is usually deposited on a conductive glass substrate as CE material due to its excellent electrical conductivity, electrocatalytic activity, and electrochemical stability. To achieve cost-efficient DSSCs, reasonable efforts have been made to explore Pt-free alternatives. Recently, the graphene-based CEs have been intensively investigated to replace the high-cost noble Pt CE. In this paper, we provided an overview of studies on the electrochemical and photovoltaic characteristics of graphene-based CEs, including graphene, graphene/Pt, graphene/carbon materials, graphene/conducting polymers, and graphene/inorganic compounds. We also summarize the design and advantages of each graphene-based material and provide the possible directions for designing new graphene-based catalysts in future research for high-performance and low-cost DSSCs

    ZIF-8-Based Quasi-Solid-State Electrolyte for Lithium Batteries.

    The quasi-solid-state electrolytes (QSSEs) with an inorganic skeleton, a solid-liquid composite material combining their respective merits, exhibit high ionic conductivity and mechanical strength. However, most quasi-solid electrolytes prepared by immobilizing ionic liquid (IL) or organic liquid electrolyte in inorganic scaffold generally have poor interface compatibility and low lithium ion migration number, which limits its application. Herein, we design and prepare a ZIF-8-based QSSE (ZIF-8 QSSE) in which the ZIF-8 has a special cage structure and interaction with the guest electrolyte to form a composite electrolyte with good ionic conductivity about 1.05 × 10-4 S cm-1 and a higher lithium-ion transference number of about 0.52. With the ZIF-8 QSSE, a protype lithium battery coupled with LiCoO2 cathode shows good electrochemical performances with an initial discharge capacity of 135 mAh g-1 at 50 mA g-1 and a remaining capacity of 119 mAh g-1 after 100 cycles, only 0.119% capacity degradation per cycle. It is worth noting that the ZIF-8-based QSSEs have good thermal stability up to 350 °C that does not show thermal runaway, which is significantly higher than that of a conventional organic liquid battery system

    Mechanistic Study of the Phytocompound, 2- β

    Bidens pilosa, a medicinal herb worldwide, is rich in bioactive polyynes. In this study, by using high resolution 2-dimensional gel electrophoresis coupled with mass spectrometry analysis, as many as 2000 protein spots could be detected and those whose expression was specifically up- or downregulated in Jurkat T cells responsive to the treatment with 2-β-D-glucopyranosyloxy-1-hydroxytrideca-5,7,9,11-tetrayne (GHTT) can be identified. GHTT treatment can upregulate thirteen proteins involved in signal transduction, detoxification, metabolism, energy pathways, and channel transport in Jurkat cells. Nine proteins, that is, thioredoxin-like proteins, BH3 interacting domain death agonist (BID protein involving apoptosis), methylcrotonoyl-CoA carboxylase beta chain, and NADH-ubiquinone oxidoreductase, were downregulated in GHTT-treated Jurkat cells. Further, bioinformatics tool, Ingenuity software, was used to predict signaling pathways based on the data obtained from the differential proteomics approach. Two matched pathways, relevant to mitochondrial dysfunction and apoptosis, in Jurkat cells were inferred from the proteomics data. Biochemical analysis further verified both pathways involving GHTT in Jurkat cells. These findings do not merely prove the feasibility of combining proteomics and bioinformatics methods to identify cellular proteins as key players in response to the phytocompound in Jurkat cells but also establish the pathways of the proteins as the potential therapeutic targets of leukemia

    The Taiwan Birth Panel Study: a prospective cohort study for environmentally- related child health

    <p>Abstract</p> <p>Background</p> <p>The Taiwan Birth Panel Study (TBPS) is a prospective follow-up study to investigate the development of child health and disease in relation to in-utero and/or early childhood environmental exposures. The rationale behind the establishment of such a cohort includes the magnitude of potential environmental exposures, the timing of exposure window, fatal and children's susceptibility to toxicants, early exposure delayed effects, and low-level or unknown neurodevelopmental toxicants.</p> <p>Methods</p> <p>A total of 486 mother-infant paired was enrolled from April 2004 to January 2005 in this study. Maternal blood before delivery, placenta and umbilical cord blood at birth, and mothers' urine after delivery were collected. The follow-up was scheduled at birth, 4, 6 months, and 1, 2, 3 and 5 years. The children's blood, urine, hair, and saliva were collected at 2 years of age and children's urine was collected at 5 years of age as well. The study has been approved by the ethical committee of National Taiwan University Hospital. All the subjects signed the inform consent on entering the study and each of the follow up.</p> <p>Results</p> <p>Through this prospective birth cohort, the main health outcomes were focused on child growth, neurodevelopment, behaviour problem and atopic diseases. We investigated the main prenatal and postnatal factors including smoking, heavy metals, perfluorinated chemicals, and non-persistent pesticides under the consideration of interaction of the environment and genes.</p> <p>Conclusions</p> <p>This cohort study bridges knowledge gaps and answers unsolved issues in the low-level, prenatal or postnatal, and multiple exposures, genetic effect modification, and the initiation and progression of "environmentally-related childhood diseases."</p

    Nuclear Export and Import of Human Hepatitis B Virus Capsid Protein and Particles

    It remains unclear what determines the subcellular localization of hepatitis B virus (HBV) core protein (HBc) and particles. To address this fundamental issue, we have identified four distinct HBc localization signals in the arginine rich domain (ARD) of HBc, using immunofluorescence confocal microscopy and fractionation/Western blot analysis. ARD consists of four tight clustering arginine-rich subdomains. ARD-I and ARD-III are associated with two co-dependent nuclear localization signals (NLS), while ARD-II and ARD-IV behave like two independent nuclear export signals (NES). This conclusion is based on five independent lines of experimental evidence: i) Using an HBV replication system in hepatoma cells, we demonstrated in a double-blind manner that only the HBc of mutant ARD-II+IV, among a total of 15 ARD mutants, can predominantly localize to the nucleus. ii) These results were confirmed using a chimera reporter system by placing mutant or wild type HBc trafficking signals in the heterologous context of SV40 large T antigen (LT). iii) By a heterokaryon or homokaryon analysis, the fusion protein of SV40 LT-HBc ARD appeared to transport from nuclei of transfected donor cells to nuclei of recipient cells, suggesting the existence of an NES in HBc ARD. This putative NES is leptomycin B resistant. iv) We demonstrated by co-immunoprecipitation that HBc ARD can physically interact with a cellular factor TAP/NXF1 (Tip-associated protein/nuclear export factor-1), which is known to be important for nuclear export of mRNA and proteins. Treatment with a TAP-specific siRNA strikingly shifted cytoplasmic HBc to nucleus, and led to a near 7-fold reduction of viral replication, and a near 10-fold reduction in HBsAg secretion. v) HBc of mutant ARD-II+IV was accumulated predominantly in the nucleus in a mouse model by hydrodynamic delivery. In addition to the revised map of NLS, our results suggest that HBc could shuttle rapidly between nucleus and cytoplasm via a novel TAP-dependent NES

    Bear bile: dilemma of traditional medicinal use and animal protection

    Bear bile has been used in Traditional Chinese Medicine (TCM) for thousands of years. Modern investigations showed that it has a wide range of pharmacological actions with little toxicological side effect and the pure compounds have been used for curing hepatic and biliary disorders for decades. However, extensive consumption of bear bile made bears endangered species. In the 1980's, bear farming was established in China to extract bear bile from living bears with "Free-dripping Fistula Technique". Bear farming is extremely inhumane and many bears died of illness such as chronic infections and liver cancer. Efforts are now given by non-governmental organizations, mass media and Chinese government to end bear farming ultimately. At the same time, systematic research has to be done to find an alternative for bear bile. In this review, we focused on the literature, laboratory and clinical results related to bear bile and its substitutes or alternative in English and Chinese databases. We examined the substitutes or alternative of bear bile from three aspects: pure compounds derived from bear bile, biles from other animals and herbs from TCM. We then discussed the strategy for stopping the trading of bear bile and issues of bear bile related to potential alternative candidates, existing problems in alternative research and work to be done in the future
