    Habitat and bycatch effects on population parameters of inshore lizardfish (Synodus foetens) in the north central Gulf of Mexico

    We examined the effect of habitat and shrimp trawl bycatch on the density, size, growth, and mortality of inshore lizardfish (Synodus foetens), a nonexploited species that is among the most widespread and abundant benthic fishes in the north central Gulf of Mexico. Results of quarterly trawl sampling conducted from spring 2004 through spring 2005 revealed that inshore lizardfish are most abundant on sand habitat, but larger fish are more common on shell rubble habitat. There was no significant difference in fish density between habitats exposed to shrimp trawling on the open shelf versus those habitats within a permitted artificial reef zone that served as a de facto no-trawl area; this finding indicates that either inshore lizardfish experienced minimal effects from trawling or, more likely, that fish moved between trawled and nontrawled habitats. Exploitation ratio (bycatch mortality/total morality) estimates derived from catch curve analysis ranged from 0.43 inside the artificial reef zone to 0.55 outside the reef zone, thus indicating that inshore lizardfish are subject to significant fishing mortality in the north central Gulf of Mexico despite the lack of a directed fishery for the species. We infer from this result that effects of shrimp trawl bycatch may be significant at the population level for nonexploited species and that a broader ecosystem-scale examination of bycatch effects is warranted

For a set of linearly independent states unambiguous discrimination is possible if we allow for the possibility of an inconclusive result.     Organizational boundaries of medical practice: the case of physician ownership of ancillary services

    Physician ownership of in-office ancillary services (IOASs) has come under increasing scrutiny. Advocates of argue that IOASs allow physicians to supervise the quality and coordination of care. Critics have argued that IOASs create financial incentives for physicians to increase ancillary service volume. In this paper we develop a conceptual framework to evaluate the tradeoffs associated with physician ownership of IOASs. There is some evidence supporting the existence of scope and transaction economies in IOASs. Improvement in flow and continuity of care are likely to generate scope economies and improvements in quality monitoring and reductions in consumer transaction costs are likely to generate transaction economies. Other factors include the capture of upstream and downstream profits, but these incentives are likely to be small compared to scope and transaction economies. Policy debates on the merits of IOASs should include an explicit assessment of these tradeoffs.     Measurement disturbance tradeoffs in three-qubit unsupervised quantum classification

    We consider measurement disturbance tradeoffs in quantum machine learning protocols which seek to learn about quantum data. We study the simplest example of a binary classification task, in the unsupervised regime. Specifically, we investigate how a classification of two qubits, that can each be in one of two unknown states, affects our ability to perform a subsequent classification on three qubits when a third is added. Surprisingly, we find a range of strategies in which a non-trivial first classification does not affect the success rate of the second classification. There is, however, a non-trivial measurement disturbance tradeoff between the success rate of the first and second classifications, and we fully characterise this tradeoff analytically

    Safe and just operating spaces for regional social-ecological systems

    Humanity faces a major global challenge in achieving wellbeing for all, while simultaneously ensuring that the biophysical processes and ecosystem services that underpin wellbeing are exploited within scientifically informed boundaries of sustainability. We propose a framework for defining the safe and just operating space for humanity that integrates social wellbeing into the original planetary boundaries concept (Rockström et al., 2009a,b) for application at regional scales. We argue that such a framework can: (1) increase the policy impact of the boundaries concept as most governance takes place at the regional rather than planetary scale; (2) contribute to the understanding and dissemination of complexity thinking throughout governance and policy-making; (3) act as a powerful metaphor and communication tool for regional equity and sustainability. We demonstrate the approach in two rural Chinese localities where we define the safe and just operating space that lies between an environmental ceiling and a social foundation from analysis of time series drawn from monitored and palaeoecological data, and from social survey statistics respectively. Agricultural intensification has led to poverty reduction, though not eradicated it, but at the expense of environmental degradation. Currently, the environmental ceiling is exceeded for degraded water quality at both localities even though the least well-met social standards are for available piped water and sanitation. The conjunction of these social needs and environmental constraints around the issue of water access and quality illustrates the broader value of the safe and just operating space approach for sustainable development

    TgPRELID, a Mitochondrial Protein Linked to Multidrug Resistance in the Parasite Toxoplasma gondii

    New drugs to control infection with the protozoan parasite Toxoplasma gondii are needed as current treatments exert toxic side effects on patients. Approaches to develop novel compounds for drug development include screening of compound libraries and targeted inhibition of essential cellular pathways. We identified two distinct compounds that display inhibitory activity against the parasite's replicative stage: F3215-0002, which we previously identified during a compound library screen, and I-BET151, an inhibitor of bromodomains, the "reader" module of acetylated lysines. In independent studies, we sought to determine the targets of these two compounds using forward genetics, generating resistant mutants and identifying the determinants of resistance with comparative genome sequencing. Despite the dissimilarity of the two compounds, we recovered resistant mutants with nonsynonymous mutations in the same domain of the same gene, TGGT1_254250, which we found encodes a protein that localizes to the parasite mitochondrion (designated TgPRELID after the name of said domain). We found that mutants selected with one compound were cross resistant to the other compound, suggesting a common mechanism of resistance. To further support our hypothesis that TgPRELID mutations facilitate resistance to both I-BET151 and F3215-0002, CRISPR (clustered regularly interspaced short palindromic repeat)/CAS9-mediated mutation of TgPRELID directly led to increased F3215-0002 resistance. Finally, all resistance mutations clustered in the same subdomain of TgPRELID. These findings suggest that TgPRELID may encode a multidrug resistance factor or that I-BET151 and F3215-0002 have the same target(s) despite their distinct chemical structures. IMPORTANCE We report the discovery of TgPRELID, a previously uncharacterized mitochondrial protein linked to multidrug resistance in the parasite Toxoplasma gondii. Drug resistance remains a major problem in the battle against parasitic infection, and understanding how TgPRELID mutations augment resistance to multiple, distinct compounds will reveal needed insights into the development of new therapies for toxoplasmosis and other related parasitic diseases

    Consumer Purchase Likelihood of Landscape Management Pest Scouting Program

    The US landscape industry consists of 632,000 businesses with >1 million persons employed in 2022. The most common service that landscape service providers (LSPs) perform is pest management. Over the past 25 years, LSPs have been challenged to adopt more holistic approaches to pest management via the use of nonchemical and less toxic chemical controls. Integrated pest management (IPM), specifically scouting, may be a useful approach for LSPs to manage pests more sustainably and market new services, such as biological control releases. Scant literature is available on LSP scouting practices or consumer acceptance of scouting services. The goal of this study was to determine if IPM-aware consumers were more likely to purchase a scouting program offered by an LSP. An online survey was distributed across the United States through a third-party panel service. The final sample included 928 usable responses. Data were analyzed using a binary logistic regression model. Fifty-seven percent of respondents reported having some knowledge or were very knowledgeable of IPM. Respondents 65 years of age and older were 13.1% points less likely to purchase a scouting service. Education level did not influence purchase likelihood. Consumer knowledge of IPM had a positive influence on the purchase likelihood, respondents with “some knowledge” (5.6%) and “very knowledgeable” (8.6%) were more likely to buy IPM services. Further, if the consumer was open to purchasing the scouting program, it is plausible that they might be more willing to allow an LSP to use a combination of chemical and nonchemical methods to manage pests

    Mining alleles for tar spot complex resistance from CIMMYT's maize Germplasm Bank

    The tar spot complex (TSC) is a devastating disease of maize (Zea mays L.), occurring in 17 countries throughout Central, South, and North America and the Caribbean, and can cause grain yield losses of up to 80%. As yield losses from the disease continue to intensify in Central America, Phyllachora maydis, one of the causal pathogens of TSC, was first detected in the United States in 2015, and in 2020 in Ontario, Canada. Both the distribution and yield losses due to TSC are increasing, and there is a critical need to identify the genetic resources for TSC resistance. The Seeds of Discovery Initiative at CIMMYT has sought to combine next-generation sequencing technologies and phenotypic characterization to identify valuable alleles held in the CIMMYT Germplasm Bank for use in germplasm improvement programs. Individual landrace accessions of the “Breeders' Core Collection” were crossed to CIMMYT hybrids to form 918 unique accessions topcrosses (F1 families) which were evaluated during 2011 and 2012 for TSC disease reaction. A total of 16 associated SNP variants were identified for TSC foliar leaf damage resistance and increased grain yield. These variants were confirmed by evaluating the TSC reaction of previously untested selections of the larger F1 testcross population (4,471 accessions) based on the presence of identified favorable SNPs. We demonstrated the usefulness of mining for donor alleles in Germplasm Bank accessions for newly emerging diseases using genomic variation in landraces

    Study protocol for the implementation and evaluation of the Self-harm Assessment and Management for General Hospitals programme in Ireland (SAMAGH)

    Background: Previous self-harm is one of the strongest predictors of future self-harm and suicide. Increased risk of repeated self-harm and suicide exists amongst patients presenting to hospital with high-risk self-harm and major self-harm repeaters. However, so far evidence-based training in the management of self-harm for mental health professionals is limited. Within this context, we aim to develop, implement and evaluate a training programme, SAMAGH, Self-harm Assessment and Management Programme for General Hospitals in Ireland. SAMAGH aims to (a) reduce hospital-based self-harm repetition rates and (b) increase rates of mental health assessments being conducted with self-harm patients. We also aim to evaluate the training on self-harm knowledge, attitudes, and skills related outcomes of healthcare professionals involved in the training. Methods/design: The study will be conducted in three phases. First, the SAMAGH Training Programme has been developed, which comprises two parts: 1) E-learning Programme and 2) Simulation Training. Second, SAMAGH will be delivered to healthcare professionals from general hospitals in Ireland. Third, an outcome and process evaluation will be conducted using a pre-post design. The outcome evaluation will be conducted using aggregated data from the National Self-Harm Registry Ireland (NSHRI) on self-harm repetition rates from all 27 public hospitals in Ireland. Aggregated data based on the 3-year average (2016, 2017, 2018) self-harm repetition rates prior to the implementation of the SAMAGH will be used as baseline data, and NSHRI data from 6 and 12 months after the implementation of SAMAGH will be used as follow-up. For the process evaluation, questionnaires and focus groups will be administered and conducted with healthcare professionals who completed the training. Discussion: This study will contribute to the evidence base regarding the effectiveness of an evidence informed training programme that aims to reduce repeated hospital self-harm presentations and to improve compliance with self-harm assessment and management. This study is also expected to contribute to self-harm and suicide training with the possibility of being translated to other settings. Its feasibility will be evaluated through a process evaluation
