    Biochemical constituents of Gracilaria edulis cultured from spores

    Gracilaria edulis was cultivated in west coast of India off Narakkal by reproductive method using the carpospores. The culture period was from November to March with regular sampling of growing plants for the estimation of biochemical constituents, quantitative and qualitative estimation of agar. The biochemical constituents like protein and carbohydrate content increased corresponding to the age of the plant whereas the lipid content declined. Harvesting of the crop can be determined from the qualitative and the quantitative estimation of agar along with other biochemical constituents.     Design and development of FRP mobile fish vending trolley for hygienic fish marketing

    A mobile fish vending trolley has been designed and developed by Indian Council of Agricultural Research (ICAR) - All India Coordinated Research Project on Plasticulture Engineering and Technology (AICRP on PET) centre at ICAR-Central Institute of Freshwater Aquaculture (ICAR-CIFA), Bhubaneswar to aid the fisher folks for vending their fish harvests in hygienic condition. The unique selling proposition (USP) of the vending unit is its unibody design, where icebox for storing fish, 20 l capacity water storage tank for hand washing, tool box, fish cutting deck and waste collection chambers are integrated into the unit. The icebox can store 100 kg of fish in ice which can be sold in a single day by the fisher folks. The complete unit of mobile fish vending carriage is fabricated with fibre reinforced plastic (FRP), because of its high strength compared to other plastics, ease of fabrication and good insulation property of fiberglass (i.e. thermal conductivity= 0.04 w/mK at normal room temperature 25 ºC). The dimensions of the carriage unit is 4.0’x2’9”x2’6” in which half portion is dedicated for the insulated ice box and the remaining portion houses the fish cutting deck, waste collection chamber and tool box. This fish vending trolley would be a boon for the marginal fish vendors, who want to sell fish in a hygienic condition and to increase income generation potential by selling 100 kg fish in a day

    Magnetic Coupling Between Non-Magnetic Ions: Eu3+ in EuN and EuP

    We consider the electronic structure of, and magnetic exchange (spin) interactions between, nominally nonmagnetic Eu^3+ ions (4f^6, S=3, L=3, J=0) within the context of the rocksalt structure compounds EuN and EuP. Both compounds are ionic [Eu^3+; N^3- and P^3-] semimetals similar to isovalent GdN. Treating the spin polarization within the 4f shell, and then averaging consistent with the J=0 configuration, we estimate semimetallic band overlaps (Eu 5d with pnictide 2p or 3p) of ~0.1 eV (EuN) and ~1.0 eV (EuP) that increase (become more metallic) with pressure. The calculated bulk modulus is 130 (86) GPa for EuN (EuP). Exchange (spin-spin) coupling calculated from correlated band theory is small and ferromagnetic in sign for EuN, increasing in magnitude with pressure. Conversely, the exchange coupling is antiferromagnetic in sign for EuP and is larger in magnitude, but decreases with compression. Study of a two-site model with S_1*S_2 coupling within the J=0,1 spaces of each ion illustrates the dependence of the magnetic correlation functions on the model parameters, and indicates that the spin coupling is sufficient to alter the Van Vleck susceptibility.     Quantitative genetic manipulation for nauplii size reduction of Artemia franciscana Kellogg, 1906 from Indian salinas and correlated changes in the polyunsaturated fatty acids (PUFA) profile

    Thirteen generations of mass selection was carried out in a strain of Artemia franciscana collected from an Indian salina. The primary trait under selection was nauplii length, and the criteria of selection was small size. While 12.4% reduction (from 517.0±59.8 μm to 452.2±25.0 μm) was realised in the trait under selection from 13 generations, substantial increase in the polyunsaturated fatty acids (PUFA) content was realised as correlated response. The polyunsaturated fatty acid (PUFA) content showed a steady increase during the selection. The PUFA percentage in G2, G4, G6, G9, G11 and G13 generations were 21.43, 27.96, 27.19, 33.27, 36.98 and 37.25 respectively compared to 18.04 in the base generation. The content of essential polyunsaturated fatty acids such as 20:5n-3 and 22:6n-3 were also high in the selected generations, compared to the base generation indicating their nutritional superiority. The smaller nauplii with an enhanced level of PUFA, especially the essential polyunsaturated fatty acids, developed through selective breeding in the present work make it a promising strain as live feed for larviculture of marine species

    Evaluation of different media for cell proliferation in mantle tissue culture of the green mussel, Perna viridis (Linnaeus, 1758)

    The aim of the present study was to establish a suitable culture system for tissue explants from the mantle of the green mussel, Perna viridis. The experiments were conducted using healthy, live green mussels in the size range of 75 to 110 g collected from Pulicat Lake, Tamil Nadu. Three different culture media namely M199, Leibovitz L-15 and sterile seawater were used to assess the most suitable medium for growth, proliferation and viability of mantle epithelial cells. The effect of the addition of two supplements viz., 10% foetal calf serum (FCS) and 0.1% yeast extract to the culture media was also evaluated. After carefully isolating the pallial layer from the mantle tissue, 1-2 mm2 size explants were successfully cultured in 12-well plates at 25°C for up to 14 days. Cultures were monitored under light and phase contrast objectives in an inverted microscope. Cell counts were made and cell size was measured for each treatment. Cells were observed to migrate from the periphery of the explant within 24 h after initiation of cultures. The liberated cells were mostly round and were either granulocytes or hyalinocytes. Fibroblast-like cells were also observed. Our results showed that proliferation of epithelial cells from mantle tissue was maximum in seawater medium (7.4x104 cells ml-1), followed by L-15 medium (2.55x104 cells ml-1). Average cell size in seawater medium was 10.72 μm and that in L-15 and M199 media was 8.56 and 6.39 μm, respectively. Adherent cells were also more prominent and higher in number in seawater medium. Supplementation of culture media with 10% FCS and 0.1% yeast extract improved both cell proliferation and cell size in all the three culture media. Four concentrations of 0.1% yeast extract (@ 50 μl, 75 μl, 100 μl, 150 μl ml-1 medium) were tested in the present study and best results were obtained with 100 μl ml-1, with respect to both cell counts and size

    Pond based grow out system of gracilaria verrucosa

    Gracilaria verrucosa was successfully cultivated in a pond based aquaculture system at Narakkal. Kerala. India. Daily growth rate of seaweed was found to be maximum (19.22%) during 60 days of introduction. The growth. pigment constituents and the yield of agar were influenced by the environmental parameters. Agar yield and photosynthetic pigments declined in the initial period up to 50 days due to acclimatization of the plants lo new environment which was collected from Chennai, India and introduced in the pond at 'Narakkal. After 60 days, the algae started establishing in the bottom of the pond and an initial harvest of 230.25 kg was harvested after 90 days of culture period. Further regular harvest was made at every 10 days interval yielding 1015 kg of Gverrucosa during the culture period. The agar content ranged between 7-14.8% and bottom sample exhibited better yield of. agar than the plants cultured in the floating raft
