    Amyloid Beta and Tau Proteins as Therapeutic Targets for Alzheimer's Disease Treatment: Rethinking the Current Strategy

    Alzheimer's disease (AD) is defined by the concurrence of accumulation of abnormal aggregates composed of two proteins: Amyloid beta (Aβ) and tau, and of cellular changes including neurite degeneration and loss of neurons and cognitive functions. Based on their strong association with disease, genetically and pathologically, it is not surprising that there has been a focus towards developing therapies against the aggregated structures. Unfortunately, current therapies have but mild benefit. With this in mind we will focus on the relationship of synaptic plasticity with Aβ and tau protein and their role as potential targets for the development of therapeutic drugs. Finally, we will provide perspectives in developing a multifactorial strategy for AD treatment

    Treatment of cutaneous neurofibromas with carbon dioxide laser: Technique and patient experience

    Cutaneous neurofibromas (cNF) are one of the hallmarks of neurofibromatosis 1 (NF1). The number of cNFs varies between individuals from a few to hundreds or even thousands and increases throughout adult life. cNFs cause a significant disease burden to adult patients and constitute an unmet need for therapy, since they may cause itch and pain and, being conspicuous and unsightly, stigmatize the patient. There is a lack of reports on how the outcome of various treatment options are perceived by the patients. Here we describe a technique for cNF removal using CO2 laser, and report how patients experience the procedure. Questionnaires were sent to patients who had had CO2 laser surgery in the French Referral Center for Neurofibromatoses, and in the Turku University Hospital, Finland, to retrospectively evaluate the patients’ global satisfaction of the procedure, treatment indications, and reasons for withdrawal from treatment, if this was the case. The number of returned questionnaires was 233/473 in France and 23/27 in Finland. The results showed that the most important indications for cNF removal were esthetic, and pain and itch caused by the tumors. In general, the procedure was well tolerated, and the degree of satisfaction was 8–10 on a scale from 0 to 10. For those 30% who discontinued the tumor removal program, the main reasons were organizational constraints, a non-satisfactory esthetic result, too many cNFs to treat, or problems with healing. Thus, the CO2 laser method is well tolerated but does not fully answer to the needs of the patients. Since medical treatment is not yet available, we encourage the use of laser removal of cNFs as a feasible method to decrease the tumor burden of the patients.</p

    Regulatory modules mediating the complex neural expression patterns of the homeobrain gene during Drosophila brain development

    Background: The homeobox gene homeobrain (hbn) is located in the 57B region together with two other homeobox genes, Drosophila Retinal homeobox (DRx) and orthopedia (otp). All three genes encode transcription factors with important functions in brain development. Hbn mutants are embryonic lethal and characterized by a reduction in the anterior protocerebrum, including the mushroom bodies, and a loss of the supraoesophageal brain commissure. Results: In this study we conducted a detailed expression analysis of Hbn in later developmental stages. In the larval brain, Hbn is expressed in all type II lineages and the optic lobes, including the medulla and lobula plug. The gene is expressed in the cortex of the medulla and the lobula rim in the adult brain. We generated a new hbnKOGal4 enhancer trap strain by reintegrating Gal4 in the hbn locus through gene targeting, which refects the complete hbn expression during development. Eight diferent enhancer-Gal4 strains covering 12 kb upstream of hbn, the two large introns and 5 kb downstream of the gene, were established and hbn expression was investigated. We characterized several enhancers that drive expression in specifc areas of the brain throughout development, from embryo to the adulthood. Finally, we generated deletions of four of these enhancer regions through gene targeting and analysed their efects on the expression and function of hbn. Conclusion: The complex expression of Hbn in the developing brain is regulated by several specifc enhancers within the hbn locus. Each enhancer fragment drives hbn expression in several specifc cell lineages, and with largely overlapping patterns, suggesting the presence of shadow enhancers and enhancer redundancy. Specifc enhancer deletion strains generated by gene targeting display developmental defects in the brain. This analysis opens an avenue for a deeper analysis of hbn regulatory elements in the future

    Die Nachhaltigkeitswirkung von neuen Gründungen : die Messung und das Management von unternehmerischen Beiträgen zur nachhaltigen Entwicklung

    The four articles of this dissertation assess how much entrepreneurs contribute to sustainable development and how this contribution can be increased. By doing so, this dissertation wants to help academics and practitioners to better measure and manage the sustainability contribution of entrepreneurship. Article 1 and 2 focus on measurement in individual firms. Article 1 develops a practical approach to capture the actual impact of individual venture and article 2 tests this approach in practice with 87 individuals participants from 58 entrepreneurial organizations. Then, article 3 explores more scalable ways to measure the sustainability contribution of multiple ventures and tests this approach with 588 ventures from Germany. Finally, article 4 explores the mechanisms through which entrepreneurial contributions to sustainable development occur and create the largest impact.Die vier Artikel der Dissertation untersuchen wie stark Gründer zur nachhaltigen Entwicklung beitragen und wie ihre Beitrag erhöht werden können. Die Forschung zielt darauf ab Wissenschaftlern und Praktikern dabei zu helfen die Nachhaltigkeitswirkung von jungen Gründern besser zu messen und zu managen. Dabei konzentrieren sich Artikel 1 und 2 auf die Messung bei einzelnen Unternehmen. Artikel 1 befasst sich mit der Entwicklung eines praktikablen Ansatzes zur Wirkungsmessung, während Artikel 2 diesen Ansatz in der Praxis testet mit 87 Teilnehmern von 58 unternehmerischen Organisationen. Artikel 3 untersucht skalierbarere Ansätze um die Beiträge von mehreren Gründungen zur nachhaltigen Entwicklung zu untersuchen und testet diesen Ansatz an 588 jungen Unternehmen in Deutschland. Abschließend befasst sich Artikel 4 mit den Mechanismen durch die junge Gründer zur nachhaltigen Entwicklung beitragen und wie der Beitrag junger Gründer erhöht werden kann
