    Mehanizam akutne neurotoksičnosti u Sprague-Dawley štakora izazvane trovanjem endosulfanom

    The purpose of this study was to investigate the molecular mechanism underlying oxidative and inflammatory neuronal cell death induced by endosulfan, a pesticide belonging to the chemical family of organochlorines. The cortical and hippocampal tissues derived from Sprague-Dawley (SD) rats treated with endosulfan exhibited increased intracellular accumulation of reactive oxygen species and oxidative damages to cellular macromolecules such as depletion of glutathione, lipid peroxidation, and protein carbonylation. Conversely, the expression of antioxidant enzymes including γ-glutamylcysteine ligase (GCL), superoxide dismutase (SOD), and heme oxygenase-1 (HO-1) was markedly reduced in the brain tissues exposed to endosulfan. Moreover, during endosulfan-induced neuronal cell death, mRNA expression of pro-inflammatory cytokines such as tumour necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) was elevated, which seemed to be mediated by the activation of nuclear factor-kappa B (NF-κB) by phosphorylation of p65 subunit. These results suggest a new molecular mechanism underlying the endosulfan-induced acute neurotoxicity via induction of oxidative stress and pro-inflammatory responses.Istražen je molekularni mehanizam koji dovodi do smrti neurona potaknute oksidativnim i upalnim procesima uzrokovanim organoklornim pesticidom endosulfanom. U tkivima korteksa i hipokampusa Sprague-Dawley (SD) štakora tretiranih endosulfanom uočena su oksidativna oštećenja staničnih makromolekula, poput smanjene razine glutationa, lipidne peroksidacije i karbonilacije proteina, te povećane unutarstanične akumulacije reaktivnih kisikovih spojeva. Isto tako, u moždanom tkivu nakon izlaganja endosulfanu značajno je smanjena ekspresija enzimskih antioksidansa, uključujući i γ-glutamilcistein ligazu (GCL), superoksidnu dismutazu (SOD) i hem oksigenazu-1 (HO-1). Tijekom endosulfanom izazvane smrti neurona povećala se i ekspresija mRNA pro-upalnih citokina poput čimbenika nekroze tumora-α (TNF-α) i interleukina-1β (IL-1β), što je čini se bilo posredovano aktivacijom nuklearnoga faktora kapa B (NF-κB) putem fosforilacije podjedinice p65. Navedeni rezultati upućuju na novi molekularni mehanizam koji stoji iza akutne neurotoksičnosti izazvane endosulfanom putem indukcije oksidativnoga stresa i pro-upalnih odgovora

    COMPARATIVE GENETICS OF APODEMUS AGRARIUS (RODENTIA: MAMMALIA) FROM INSULAR AND CONTINENTAL EURASIAN POPULATIONS: CYTOCHROME B SEQUENCE ANALYSES

    To reexamine genetic divergence of Apodemus agrarius in insular Taiwan and the Korean Jeju from other populations in continental Eurasia, we obtained 91 cytochrome b complete sequences of A. agrarius across Eurasia, and these sequences were compared to eight corresponding sequences of A. agrarius, obtained from GenBank. We first found that the two insular populations are two clades, and that each of them is divergent from continental Eurasian populations, clustered into another clade. Each of the two insular clades appeared to be isolated in spite of land connection to nearby continent during the last glacial period, and we considered the two insular forms as A. a. insulaemus and A. a. chejuensis and one continental Eurasian form as A. a. agrarius, although further analyses are needed to confirm our present findings. Additionally, the Taiwan clade seemed to be composed of two subclades (western and eastern), separated by Taiwan’s central mountain range

    A Study on Apparel Design for Chiropractic Training

    In severe cases of spinal misalignment due to wrong back posture during adolescence or focusing on work, modern people can have back pain. Such a symptom is commonly referred to as scoliosis and it already became one of the lifestyle diseases. Hence, the purpose of the present study was to develop a functional musculoskeletal correction wear that can help with maintaining a correct posture by relaxing tonic muscles and maximizing the tension of the relaxed muscle using the principle of sports taping. Market research for design development was conducted at department stores and specialty road shops separately. Department stores were visited on weekdays when the store is not crowded, and specialty road shops were visited at around three o’clock on weekdays after checking their opening, lunch, and closing times. Since the apparel should not interfere with exercise movements while maintaining the body shape, the survey was carried out mostly on yoga and lifestyle training apparel focusing on elasticity and percentage of blending fiber. A camera and audio recorder were prepared and took pictures of clothing tags that show the ratio of polyester and spandex while trying on and checking products at the stores. Data were collected by asking the staff at road specialty shops who has more professional knowledge about the advantages and functions of the existing products and recording their answers. Point color schemes in black color family in which body shapes can look beautiful were mostly checked. Due to the design characteristics that have to maintain the spine, the designs of all-in-one products that can maintain the upper and lower torsos were checked at underwear stores. The existing products were evaluated for their designs to be adopted including the material, integrated into the new design, and then modified and supplemented, and the following chiropractic apparel design was drawn (Table 1). An all-in-one design that wraps around the hips and upper thighs for easy counterbalancing between the upper and lower body was chosen. A design with an accent line that makes body shape looks beautiful and Used compression wear material that is comfortable for activities and presses tight against the skin. Activates the muscle activities of latissimus dorsi muscle using taping method, and provides tension to maintain correct posture of round back and shoulders. In conclusion, prevention, exercise, and stretching are important for scoliosis and sound posture for the loin should be constantly maintained. To that end, not only the proper use of chairs but also apparel that is worn all the time can be helpful

    Virtual Surface Characteristics of a Tactile Display Using Magneto-Rheological Fluids

    Virtual surface characteristics of tactile displays are investigated to characterize the feeling of human touch for a haptic interface application. In order to represent the tactile feeling, a prototype tactile display incorporating Magneto-Rheological (MR) fluid has been developed. Tactile display devices simulate the finger’s skin to feel the sensations of contact such as compliance, friction, and topography of the surface. Thus, the tactile display can provide information on the surface of an organic tissue to the surgeon in virtual reality. In order to investigate the compliance feeling of a human finger’s touch, normal force responses of a tactile display under various magnetic fields have been assessed. Also, shearing friction force responses of the tactile display are investigated to simulate the action of finger dragging on the surface. Moreover, different matrix arrays of magnetic poles are applied to form the virtual surface topography. From the results, different tactile feelings are observed according to the applied magnetic field strength as well as the arrays of magnetic poles combinations. This research presents a smart tactile display technology for virtual surfaces

    Kyungheechunggan-Tang-01, a New Herbal Medication, Suppresses LPS-Induced Inflammatory Responses through JAK/STAT Signaling Pathway in RAW 264.7 Macrophages

    Medicinal plants have been used as alternative therapeutic tools to alleviate inflammatory diseases. The objective of this study was to evaluate anti-inflammatory properties of Kyungheechunggan-tang- (KCT-) 01, KCT-02, and Injinchunggan-tang (IJCGT) as newly developed decoctions containing 3–11 herbs in LPS-induced macrophages. KCT-01 showed the most potent inhibitory effects on LPS-induced NO, PGE2, TNF-α, and IL-6 production among those three herbal formulas. In addition, KCT-01 significantly inhibited LPS-induced iNOS and COX-2 at protein levels and expression of iNOS, COX-2, TNF-α, and IL-6 at mRNA levels. Molecular data revealed that KCT-01 attenuated the activation of JAK/STAT signaling cascade without affecting NF-κB or AP-1 activation. In ear inflammation induced by croton oil, KCT-01 significantly reduced edema, MPO activity, expression levels of iNOS and COX-2, and STAT3 phosphorylation in ear tissues. Taken together, our findings suggest that KCT-01 can downregulate the expression of proinflammatory genes by inhibiting JAK/STAT signaling pathway under inflammatory conditions. This study provides useful data for further exploration and application of KCT-01 as a potential anti-inflammatory medicine

    A Single-Center, Randomized Double-Blind Placebo-Controlled Study Evaluating the Effects of Poly-Gamma-Glutamate on Human NK Cell Activity after an 8-Week Oral Administration in Healthy Volunteers

    A randomized double-blind placebo-controlled immunity study involving 99 healthy volunteers was performed to investigate the effect of poly-γ-glutamate (γ-PGA) on human natural killer (NK) cell activity in peripheral blood. The volunteers were randomly assigned to one of three groups and orally treated with solutions (25 mL) containing 0 mg (placebo), 250 mg (low dosage), or 500 mg (high dosage) of γ-PGA. Each volunteer took one dose every 12 hours for 8 weeks. Blood samples were drawn before the initial treatment and at the 4th and the 8th weeks of treatment. NK cell activity was assessed by measuring its degranulation, cytokine production, and cytotoxicity against the K562 cell line. Our results revealed that the cytotoxic activities of NK cells from the high-dosage γ-PGA group were significantly higher (P<0.05 for all comparisons) compared to the low dosage and placebo groups at weeks 4 and 8 after the initial treatment. This increase in the NK cell activity among peripheral blood mononuclear cells (PBMCs) of healthy individuals was also confirmed in vitro (as assessed by the degranulation and cytokine production). These results suggest that the oral administration of γ-PGA induces a cell-mediated immunity by increasing the NK cell activity in humans

    Lack of Mitochondrial DNA Sequence Divergence between Two Subspecies of the Siberian Weasel from Korea: Mustela sibirica coreanus from the Korean Peninsula and M. s. quelpartis from Jeju Island

    The objective of this study was to determine the degree of mitochondrial DNA (mtDNA) divergence between two subspecies of Mustela sibirica from Korea (M. s. coreanus on the Korean Peninsula and M. s. quelpartis on Jeju Island) and to examine the taxonomic status of M. s. quelpartis. Thus, we obtained complete sequences of mtDNA cytochrome b gene (1,140 bp) from the two subspecies, and these sequences were compared to a corresponding haplotype of M. s. coreanus, downloaded from GenBank. From this analysis, it was observed that the sequences from monogenic M. s. quelpartis on Jeju Island were identical to the sequences of four M. s. coreanus from four locations across the Korean Peninsula, and that the two subspecies formed a single clade; the average nucleotide distance between the two subspecies was 0.26% (range, 0.00 to 0.53%). We found that the subspecies quelpartis is not genetically distinct from the subspecies coreanus, and that this cytochrome b sequencing result does not support the current classification, distinguishing these two subspecies by pelage color. Further systematic analyses using morphometric characters and other DNA markers are necessary to confirm the taxonomic status of M. s. quelpartis

    The lipoxygenase gene family: a genomic fossil of shared polyploidy between Glycine max and Medicago truncatula

    <p>Abstract</p> <p>Background</p> <p>Soybean lipoxygenases (<it>Lxs</it>) play important roles in plant resistance and in conferring the distinct bean flavor. <it>Lxs </it>comprise a multi-gene family that includes <it>GmLx1</it>, <it>GmLx2 </it>and <it>GmLx3</it>, and many of these genes have been characterized. We were interested in investigating the relationship between the soybean lipoxygenase isozymes from an evolutionary perspective, since soybean has undergone two rounds of polyploidy. Here we report the tetrad genome structure of soybean <it>Lx </it>regions produced by ancient and recent polyploidy. Also, comparative genomics with <it>Medicago truncatula </it>was performed to estimate <it>Lxs </it>in the common ancestor of soybean and <it>Medicago</it>.</p> <p>Results</p> <p>Two <it>Lx </it>regions in <it>Medicago truncatula </it>showing synteny with soybean were analyzed. Differential evolutionary rates between soybean and <it>Medicago </it>were observed and the median Ks values of Mt-Mt, Gm-Mt, and Gm-Gm paralogs were determined to be 0.75, 0.62, and 0.46, respectively. Thus the comparison of Gm-Mt paralogs (Ks = 0.62) and Gm-Mt orthologs (Ks = 0.45) supports the ancient duplication of <it>Lx </it>regions in the common ancestor prior to the <it>Medicago</it>-<it>Glycine </it>split. After speciation, no <it>Lx </it>regions generated by another polyploidy were identified in <it>Medicago</it>. Instead tandem duplication of <it>Lx </it>genes was observed. On the other hand, a lineage-specific duplication occurred in soybean resulting in two pairs of <it>Lx </it>regions. Each pair of soybean regions was co-orthologous to one <it>Lx </it>region in <it>Medicago</it>. A total of 34 <it>Lx </it>genes (15 <it>MtLxs </it>and 19 <it>GmLxs) </it>were divided into two groups by phylogenetic analysis. Our study shows that the <it>Lx </it>gene family evolved from two distinct <it>Lx </it>genes in the most recent common ancestor.</p> <p>Conclusion</p> <p>This study analyzed two pairs of <it>Lx </it>regions generated by two rounds of polyploidy in soybean. Each pair of soybean homeologous regions is co-orthologous to one region of <it>Medicago</it>, demonstrating the quartet structure of the soybean genome. Differential evolutionary rates between soybean and <it>Medicago </it>were observed; thus optimized rates of Ks per year should be applied for accurate estimation of coalescence times to each case of comparison: soybean-soybean, soybean-<it>Medicago</it>, or <it>Medicago</it>-<it>Medicago</it>. In conclusion, the soybean <it>Lx </it>gene family expanded by ancient polyploidy prior to taxon divergence, followed by a soybean- specific duplication and tandem duplications, respectively.</p
