    Dynamic clustering of time series with Echo State Networks

    In this paper we introduce a novel methodology for unsupervised analysis of time series, based upon the iterative implementation of a clustering algorithm embedded into the evolution of a recurrent Echo State Network. The main features of the temporal data are captured by the dynamical evolution of the network states, which are then subject to a clustering procedure. We apply the proposed algorithm to time series coming from records of eye movements, called saccades, which are recorded for diagnosis of a neurodegenerative form of ataxia. This is a hard classification problem, since saccades from patients at an early stage of the disease are practically indistinguishable from those coming from healthy subjects. The unsupervised clustering algorithm implanted within the recurrent network produces more compact clusters, compared to conventional clustering of static data, and provides a source of information that could aid diagnosis and assessment of the disease.Universidad de Málaga.     Predicting the safety and efficacy of butter therapy to raise tumour pHe: an integrative modelling study

    Background: Clinical positron emission tomography imaging has demonstrated the vast majority of human cancers exhibit significantly increased glucose metabolism when compared with adjacent normal tissue, resulting in an acidic tumour microenvironment. Recent studies demonstrated reducing this acidity through systemic buffers significantly inhibits development and growth of metastases in mouse xenografts.\ud \ud Methods: We apply and extend a previously developed mathematical model of blood and tumour buffering to examine the impact of oral administration of bicarbonate buffer in mice, and the potential impact in humans. We recapitulate the experimentally observed tumour pHe effect of buffer therapy, testing a model prediction in vivo in mice. We parameterise the model to humans to determine the translational safety and efficacy, and predict patient subgroups who could have enhanced treatment response, and the most promising combination or alternative buffer therapies.\ud \ud Results: The model predicts a previously unseen potentially dangerous elevation in blood pHe resulting from bicarbonate therapy in mice, which is confirmed by our in vivo experiments. Simulations predict limited efficacy of bicarbonate, especially in humans with more aggressive cancers. We predict buffer therapy would be most effectual: in elderly patients or individuals with renal impairments; in combination with proton production inhibitors (such as dichloroacetate), renal glomular filtration rate inhibitors (such as non-steroidal anti-inflammatory drugs and angiotensin-converting enzyme inhibitors), or with an alternative buffer reagent possessing an optimal pK of 7.1–7.2.\ud \ud Conclusion: Our mathematical model confirms bicarbonate acts as an effective agent to raise tumour pHe, but potentially induces metabolic alkalosis at the high doses necessary for tumour pHe normalisation. We predict use in elderly patients or in combination with proton production inhibitors or buffers with a pK of 7.1–7.2 is most promising

    Induction of CYP2E1 activity in liver transplant patients as measured by chlorzoxazone 6-hydroxylation

    Objective: To examine the phenotypic expression of CYP2E1 in liver transplant patients, as measured by the in vivo probe chlorzoxazone, and to evaluate CYP2E1 activity over me after transplantation. Methods: Thirty-three stable liver transplant: patients were given 250 mg chlorzoxazone within 1 year after transplantation as part of a multiprobe CYP cocktail; urine and blood were collected for 8 hours. Chlorzoxazone and 6-hydroxychlorzoxazone concentrations were determined by HPLC. Twenty-eight healthy control subjects, eight patients with moderate to severe liver disease, and four patients who had not received liver transplants were also studied for comparison. The chlorzoxazone metabolic ratio, calculated as the plasma concentration of 6-hydroxychlorzoxazone/chlorzoxazone at 4 hours after chlorzoxazone administration, was used as the phenotypic, index. In a subgroup of patients and control subjects, additional blood samples were obtained to allow for the calculation of chlorzoxazone pharmacokinetic parameters by noncompartmental methods. Results: The chlorzoxazone metabolic ratio for the liver transplant patients in the first month after transplantation (mean ± SD, 6.4 ± 5.1) was significantlp higher than that after 1 month after surgery (2.1 ± 2.0), when the chlorzoxazone metabolic ratio was not different from control subjects (0.8 ± 0.5). The chlorzoxazone metabolic ratios in the patients who had not received liver transplants (1.1 ± 0.7) were equivalent to those of healthy control subjects. The maximum observed 6-hydroxychlorzoxazone plasma concentration was 3046 ± 1848 ng/ml in seven liver transplant patients in the first month after surgery compared with 1618 ± 320 ng/ml in 16 healthy control subjects (p < 0.05). The maximum observed concentration of chlorzoxazone, the chlorzoxazone apparent oral clearance, and the formation clearance of 6-hydroxychlorzoxazone were also significantly different between the groups. Conclusions: We conclude that significant induction of CYP2E1, as indicated by the chlorzoxazone metabolic, ratio, occurs in the first month after surgery in liver transplant patients and that drugs that are substrates for CYP2E1 may require dosage alteration during that period. Contrary to expectations, drug metabolism is not uniformly depressed after liver transplantation

    The Ward error sum of squares hierarchical clustering method has been very widely used since its first description by Ward in a 1963 publication. It has also been generalized in various ways. However there are different interpretations in the literature and there are different implementations of the Ward agglomerative algorithm in commonly used software systems, including differing expressions of the agglomerative criterion. Our survey work and case studies will be useful for all those involved in developing software for data analysis using Ward's hierarchical clustering method.Comment: 20 pages, 21 citations, 4 figure

    Shear sum rules at finite chemical potential

    We derive sum rules which constrain the spectral density corresponding to the retarded propagator of the T_{xy} component of the stress tensor for three gravitational duals. The shear sum rule is obtained for the gravitational dual of the N=4 Yang-Mills, theory of the M2-branes and M5-branes all at finite chemical potential. We show that at finite chemical potential there are additional terms in the sum rule which involve the chemical potential. These modifications are shown to be due to the presence of scalars in the operator product expansion of the stress tensor which have non-trivial vacuum expectation values at finite chemical potential.Comment: The proof for the absence of branch cuts is corrected.Results unchange

    Testing approach to new fibre-reinforced UHTC materials in the C3HARME project

    In space applications, very high temperatures are often a driving environmental condition for certain technologies. This is especially true for the engines of rockets and for re-entry systems where thermal protection is vital. With current state-of-the-art systems of both types, there are in some cases materials used which are simply consumed because temperatures are so high, there is no choice of a material able to withstand the conditions. In order to overcome these restraints, Ultra-high-temperature ceramics (UHTC) provide an option in terms of the pure temperature capability of the material. However, bulk UHTC materials so far suffered from the problem of rather low mechanical properties and especially size limitations due to thermal shock sensitivity. If this situation could be improved by introducing a fibre reinforcement and combining the advantages of UHTC with the robustness of Ceramic Matrix Composites (CMC), the resulting UHTCMC materials could open up a whole new world of possibilities for new designs and flight regimes of the systems mentioned. The C3HARME project aims at exactly that. The goal is to develop new fibre-reinforced UHTC types and to significantly improve the maturity of them to a level of TRL 5-6. In order to do so, the thermo-mechanical characterization of samples and components is of great importance. A multitude of different tests are done to get the full data set of all the relevant properties from room temperature up to very high temperatures as they are encountered in the application scenario. The talk will give an overview about the characterization strategy and the individual tests and facilities applied with some of the results already obtained

    Sum rules and three point functions

    Sum rules constraining the R-current spectral densities are derived holographically for the case of D3-branes, M2-branes and M5-branes all at finite chemical potentials. In each of the cases the sum rule relates a certain integral of the spectral density over the frequency to terms which depend both on long distance physics, hydrodynamics and short distance physics of the theory. The terms which which depend on the short distance physics result from the presence of certain chiral primaries in the OPE of two R-currents which are turned on at finite chemical potential. Since these sum rules contain information of the OPE they provide an alternate method to obtain the structure constants of the two R-currents and the chiral primary. As a consistency check we show that the 3 point function derived from the sum rule precisely matches with that obtained using Witten diagrams.Comment: 41 page

    Does a 'direct' transfer protocol reduce time to coronary angiography for patients with non-ST-elevation acute coronary syndromes? A prospective observational study.

    OBJECTIVE: National guidelines recommend 'early' coronary angiography within 96 h of presentation for patients with non-ST elevation acute coronary syndromes (NSTE-ACS). Most patients with NSTE-ACS present to their district general hospital (DGH), and await transfer to the regional cardiac centre for angiography. This care model has inherent time delays, and delivery of timely angiography is problematic. The objective of this study was to assess a novel clinical care pathway for the management of NSTE-ACS, known locally as the Heart Attack Centre-Extension or HAC-X, designed to rapidly identify patients with NSTE-ACS while in DGH emergency departments (ED) and facilitate transfer to the regional interventional centre for 'early' coronary angiography. METHODS: This was an observational study of 702 patients divided into two groups; 391 patients treated before the instigation of the HAC-X pathway (Pre-HAC-X), and 311 patients treated via the novel pathway (Post-HAC-X). Our primary study end point was time from ED admission to coronary angiography. We also assessed the length of hospital stay. RESULTS: Median time from ED admission to coronary angiography was 7.2 (IQR 5.1-10.2) days pre-HAC-X compared to 1.0 (IQR 0.7-2.0) day post-HAC-X (p<0.001). Median length of hospital stay was 3.0 (IQR 2.0-6.0) days post-HAC-X v 9.0 (IQR 6.0-14.0) days pre-HAC-X (p<0.0005). This equates to a reduction of six hospital bed days per NSTE-ACS admission. CONCLUSIONS: The introduction of this novel care pathway was associated with significant reductions in time to angiography and in total hospital bed occupancy for patients with NSTE-ACS
