Population Variability Generated during Rescue Process and Passaging of Recombinant Mumps Viruses

Recombinant mumps viruses (MuVs) based on established vaccine strains represent attractive vector candidates as they have known track records for high efficacy and the viral genome does not integrate in the host cells. We developed a rescue system based on the consensus sequence of the L- Zagreb vaccine and generated seven different recombinant MuVs by (a) insertion of one or two additional transcription units (ATUs), (b) lengthening of a noncoding region to the extent that the longest noncoding region in MuV genome is created, or (c) replacement of original L-Zagreb sequences with sequences rich in CG and AT dinucleotides. All viruses were successfully rescued and faithfully matched sequences of input plasmids. In primary rescued stocks, low percentages of heterogeneous positions were found (maximum 0.12%) and substitutions were predominantly obtained in minor variants, with maximally four substitutions seen in consensus. ATUs did not accumulate more mutations than the natural MuV genes. Six substitutions characteristic for recombinant viruses generated in our system were defined, as they repetitively occurred during rescue processes. In subsequent passaging of primary rescue stocks in Vero cells, different inconsistencies within quasispecies structures were observed. In order to assure that unwanted mutations did not emerge and accumulate, sub-consensus variability should be closely monitored. As we show for Pro408Leu mutation in L gene and a stop codon in one of ATUs, positively selected variants can rise to frequencies over 85% in only few passages

Primjena, provedba i izvršenje zakonodavstva Europske unije zajednički je izazov Europske unije (dalje: Unija) i država članica. U radu se analizira uređenje Europske komisije kao jedne od ključnih institucija Unije koja pridaje veliku važnost osiguravanju djelotvorne primjene prava Unije. Tako se najprije, u uvodnom dijelu upozorava na važnost prava Unije odnosno njegove izvore i primjenu. U središnjem dijelu rada posebna pozornost pridaje se nadzornoj funkciji Europske komisije. Rad završava pregledom godišnjih izvješća Europske komisije o primjeni prava Unije u području poreza od 2013. do 2020. godine.The application, implementation and enforcement of European Union legislation is a common challenge of the European Union and its Member States. This paper analyses the organisation of the European Commission as one of the key institutions of the Union, which attaches great importance to ensuring the effective application of Union law. Thus, first, the introductory part emphasises the importance of Union law, i.e. its sources and application. In the central part of the paper, particular attention is paid to the supervisory function of the European Commission. The paper concludes with the overview of the European Commission's annual reports on the application of Union tax law from 2013 to 2020

Fotoesej

Fotoesej je forma vizualnog izražavanja u fotografiji i fotonovinarstvu. Fotoesej se sastoji od serije fotografija u kojoj svaka fotografija ima svoju ulogu u pričanju priče ili izlaganju osnovne ideje, a koju bi bilo nemoguće prikazati u jednoj fotografiji. Fotografije u fotoeseju su odabrane i grupirane tako da, osim što pričaju priču, jasno odražavaju osobni stav autora. Dva su osnovna tipa fotoeseja: narativni i tematski. Narativni fotoesej priča priču kroz fotografije slijeda događaja ili radnji. Može pratiti osobu ili događaj kroz određeni vremenski period i prezentirati priču kronološkim redoslijedom. Tematski fotoesej se fokusira na centralnu temu i prezentira ju kroz fotografije koje su vezane za tu temu. Fotoesej je, u stvari, specifičan vid novinske fotografije. Iako je svojom formom definiran, autoru-fotografu daje više slobode u izražavanju od srodnog vida novinske fotografije – fotopriče. Zbog toga se u diplomskom radu teoretski analiziraju vidovi novinske fotografije uz poseban naglasak na fotoesej i fotopriču koji se uspoređuju te se ukazuje na njihove razlike i sličnosti u izražavanju. U praktičnom se dijelu rada izrađuju dva fotoeseja na istu temu kojoj se pristupa na dva različita načina. Istoj temi se pristupa narativno i tematski te se metodom intervjua utvrđuje koja od dvije serije fotografija daje efikasnije rezultate u prenošenju ideja, misli, emocija i impresija
