Allele-specific deposition of macroH2A1 in imprinting control regions
In the current study, we analyzed the deposition patterns of macroH2A1 at a number of different genomic loci located in X chromosome and autosomes. MacroH2A1 is preferentially deposited at methylated CpG-rich regions located close to promoters. The macroH2A1 deposition patterns at the methylated CpG islands of several imprinted domains, including the imprinting control regions (ICRs) of Xist, Peg3, H19/Igf2, Gtl2/Dlk1 and Gnas domains, show consistent allele-specificity towards inactive, methylated alleles. The macroH2A1 deposition levels at the ICRs and other differentially methylated regions of these domains are also either higher or comparable to those observed at the inactive X chromosome of female mammals. Overall, our results indicate that besides DNA methylation macroH2A1 is another epigenetic component in the chromatin of ICRs displaying differential association with two parental alleles. © The Author 2006. The Cardiac Atlas Project--An Imaging Database for Computational Modeling and Statistical Atlases of the Heart
MOTIVATION: Integrative mathematical and statistical models of cardiac anatomy and physiology can play a vital role in understanding cardiac disease phenotype and planning therapeutic strategies. However, the accuracy and predictive power of such models is dependent upon the breadth and depth of noninvasive imaging datasets. The Cardiac Atlas Project (CAP) has established a large-scale database of cardiac imaging examinations and associated clinical data in order to develop a shareable, web-accessible, structural and functional atlas of the normal and pathological heart for clinical, research and educational purposes. A goal of CAP is to facilitate collaborative statistical analysis of regional heart shape and wall motion and characterize cardiac function among and within population groups.
RESULTS: Three main open-source software components were developed: (i) a database with web-interface; (ii) a modeling client for 3D + time visualization and parametric description of shape and motion; and (iii) open data formats for semantic characterization of models and annotations. The database was implemented using a three-tier architecture utilizing MySQL, JBoss and Dcm4chee, in compliance with the DICOM standard to provide compatibility with existing clinical networks and devices. Parts of Dcm4chee were extended to access image specific attributes as search parameters. To date, approximately 3000 de-identified cardiac imaging examinations are available in the database. Anti-proliferative effects of Bifidobacterium adolescentis SPM0212 extract on human colon cancer cell lines
<p>Abstract</p> <p>Background</p> <p>Lactic acid bacteria (LAB) are beneficial probiotic organisms that contribute to improved nutrition, microbial balance, and immuno-enhancement of the intestinal tract, as well as anti-tumor activity. The aim of the present work was to study the growth inhibition of tumor cells by butanol extract of <it>Bifidobacterium adolescentis </it>isolated from healthy young Koreans.</p> <p>Methods</p> <p>The anti-proliferative activity of <it>B. adolescentis </it>isolates was assessed by XTT assays on three human colon cancer cell lines (Caco-2, HT-29, and SW480). The effects of <it>B. adolescentis </it>SPM0212 butanol extract on tumor necrosis factor-α (TNF-α) and nitric oxide (NO) production were tested using the murine macrophage RAW 264.7 cell line.</p> <p>Results</p> <p>The butanol extract of <it>B. adolescentis </it>SPM0212 dose-dependently inhibited the growth of Caco-2, HT-29, and SW480 cells by 70%, 30%, and 40%, respectively, at 200 μg/mL. Additionally, the butanol extract of <it>B. adolescentis </it>SPM0212 induced macrophage activation and significantly increased the production of TNF-α and NO, which regulate immune modulation and are cytotoxic to tumor cells.</p> <p>Conclusion</p> <p>The butanol extract of <it>B. adolescentis </it>SPM0212 increased activity of the host immune system and may improve human health by helping to prevent colon cancer as a biological response modifier.</p
A New Hardware Correlator in Korea: Performance Evaluation using KVN observations
We report results of the performance evaluation of a new hardware correlator
in Korea, the Daejeon correlator, developed by the Korea Astronomy and Space
Science Institute (KASI) and the National Astronomical Observatory of Japan
(NAOJ). We conducted Very Long Baseline Interferometry (VLBI) observations at
22~GHz with the Korean VLBI Network (KVN) in Korea and the VLBI Exploration of
Radio Astrometry (VERA) in Japan, and correlated the aquired data with the
Daejeon correlator. For evaluating the performance of the new hardware
correlator, we compared the correlation outputs from the Daejeon correlator for
KVN observations with those from a software correlator, the Distributed FX
(DiFX). We investigated the correlated flux densities and brightness
distributions of extragalactic compact radio sources. The comparison of the two
correlator outputs show that they are consistent with each other within ,
which is comparable with the amplitude calibration uncertainties of KVN
observations at 22~GHz. We also found that the 8\% difference in flux density
is caused mainly by (a) the difference in the way of fringe phase tracking
between the DiFX software correlator and the Daejeon hardware correlator, and
(b) an unusual pattern (a double-layer pattern) of the amplitude correlation
output from the Daejeon correlator. The visibility amplitude loss by the
double-layer pattern is as small as 3\%. We conclude that the new hardware
correlator produces reasonable correlation outputs for continuum observations,

Could Fractional Exhaled Nitric Oxide Test be Useful in Predicting Inhaled Corticosteroid Responsiveness in Chronic Cough? A Systematic Review
© 2016 Background Fractional exhaled nitric oxide (FENO) is a safe and convenient test for assessing T H 2 airway inflammation, which is potentially useful in the management of patients with chronic cough. Objective To summarize the current evidence on the diagnostic usefulness of FENO for predicting inhaled corticosteroid (ICS) responsiveness in patients with chronic cough. Methods A systematic literature review was conducted to identify articles published in peer-reviewed journals up to February 2015, without language restriction. We included studies that reported the usefulness of FENO (index test) for predicting ICS responsiveness (reference standard) in patients with chronic cough (target condition). The data were extracted to construct a 2 × 2 accuracy table. Study quality was assessed with Quality Assessment of Diagnostic Accuracy Studies 2. Results We identified 5 original studies (2 prospective and 3 retrospective studies). We identified considerable heterogeneities in study design and outcome definitions, and thus were unable to perform a meta-analysis. The proportion of ICS responders ranged from 44% to 59%. Sensitivity and specificity ranged from 53% to 90%, and from 63% to 97%, respectively. The reported area under the curve ranged from abou t 0.60 to 0.87; however, studies with a prospective design and a lower prevalence of asthma had lower area under the curve values. None measured placebo effects or objective cough frequency. Conclusions We did not find strong evidence to support the use of FENO tests for predicting ICS responsiveness in chronic cough. Further studies need to have a randomized, placebo-controlled design, and should use validated measurement tools for cough. Standardization would facilitate the development of clinical evidence
