    NITRIC OXIDE INHIBITORY ACTIVITY OF STRYCHNOS SPINOSA (LOGANIACEAE) LEAF EXTRACTS AND FRACTIONS

    Background: The study was aimed at determining the anti-inflammatory activity of fractions and extracts obtained from Strychnos spinosa leaves on a mediator of inflammation nitric oxide (NO). Materials and Methods: Leaves were extracted with acetone and separated into fractions with different polarities by solventsolvent fractionation. The Griess assay was used to determine the nitric oxide (NO) inhibitory activity. Cellular toxicity was determined by "using the MTT reduction assay". Results: With the exception of the ethyl acetate fraction which had an IC50 >750 μg/mL, all extracts and fractions had significant nitric oxide-inhibitory activity. The most active being the water fraction, chloroform fraction and the dichloromethane/methanol extracts with IC50 values of 88.43 μg/mL, 96.72 μg/mL and 115.62 μg/mL, respectively. The extracts and fractions had low cytotoxicity on macrophage U937 cell lines. Conclusion: Extracts and fractions of Strychnos spinosa leaves may be promising sources of natural anti-inflammatory agents. Findings obtained from this study showed that Strychnos spinosa leaves possess promising anti-inflammatory action and could be used in the treatment of inflammation-related conditions

    In vitro antibacterial activity and acute toxicity studies of aqueous-methanol extract of Sida rhombifolia Linn. (Malvaceae)

    <p>Abstract</p> <p>Background</p> <p>Many bacteria among the Enterobacteria family are involved in infectious diseases and diarrhoea. Most of these bacteria become resistant to the most commonly used synthetic drugs in Cameroon. Natural substances seem to be an alternative to this problem. Thus the aim of this research was to investigate the <it>in vitro </it>antibacterial activity of the methanol and aqueous-methanol extracts of <it>Sida rhombifolia </it>Linn (Malvaceae) against seven pathogenic bacteria involved in diarrhoea. Acute toxicity of the most active extract was determined and major bioactive components were screened.</p> <p>Methods</p> <p>The agar disc diffusion and the agar dilution method were used for the determination of inhibition diameters and the Minimum Inhibitory Concentration (MICs) respectively. The acute toxicity study was performed according WHO protocol.</p> <p>Results</p> <p>The aqueous-methanol extract (1v:4v) was the most active with diameters of inhibition zones ranging from 8.7 - 23.6 mm, however at 200 μg/dic this activity was relatively weak compared to gentamycin. The MICs of the aqueous-methanol extract (1v:4v) varied from 49.40 to 78.30 μg/ml. <it>Salmonella dysenteriae </it>was the most sensitive (49.40 μg/ml). For the acute toxicity study, no deaths of rats were recorded. However, significant increase of some biochemical parameters such as aspartate amino-transferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) and creatinine (CRT) were found. The phytochemical analysis of the aqueous methanol extract indicated the presence of tannins, polyphenols, alkaloids, glycosides, flavonoids and saponins</p> <p>Conclusion</p> <p>The results showed that the aqueous-methanol extract of <it>S. rhombifolia </it>exhibited moderate antibacterial activity.     Amelioration of Experimental Autoimmune Encephalomyelitis by Plumbagin through Down-Regulation of JAK-STAT and NF-κB Signaling Pathways

    Plumbagin(PL), a herbal compound derived from roots of the medicinal plant Plumbago zeylanica, has been shown to have immunosuppressive properties. Present report describes that PL is a potent novel agent in control of encephalitogenic T cell responses and amelioration of mouse experimental autoimmune encephalomyelitis (EAE), through down-regulation of JAK-STAT pathway. PL was found to selectively inhibit IFN-γ and IL-17 production by CD4+ T cells, which was mediated through abrogated phosphorylation of JAK1 and JAK2. Consistent with IFN-γ and IL-17 reduction was suppressed STAT1/STAT4/T-bet pathway which is critical for Th1 differentiation, as well as STAT3/ROR pathway which is essential for Th17 differentiation. In addition, PL suppressed pro-inflammatory molecules such as iNOS, IFN-γ and IL-6, accompanied by inhibition of IκB degradation as well as NF-κB phosphorylation. These data give new insight into the novel immune regulatory mechanism of PL and highlight the great value of this kind of herb compounds in probing the complex cytokine signaling network and novel therapeutic targets for autoimmune diseases

    Antimycobacterial, antioxidant and cytotoxic activities of essential oil of gall of Pistacia atlantica Desf. From Algeria

    Background: The aim of this study was to assess the antimycobacterial, antioxidant and the cytotoxic activities of the essential oil from the gall part of Pistacia atlantica Desf from Algeria.Materials and Methods: The antimycobacterial activity was evaluated by the broth microdilution method against three species of mycobacteria: Mycobacterium smegmatis, Mycobacterium aurum and Mycobacterium fortuitum. Antioxidant activity was determined using free-radical scavenging assays. The safety of essential oil was assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay on C3A and Vero monkey kidney cells.Results: The minimal inhibitory concentration (MIC) values ranged from 0.16 to 2.5 mg/mL with minimal bactericidal concentration (MBC) values ranged from 0.62 to &gt;2.5 mg/mL. The antioxidant activity showed IC50 values ranged between 417.61 - &gt;2000μg/mL and 495.6 - &gt;2000μg/mL for DPPH and ABTS assay respectively. The cytotoxicity assay showed LC50 ranged between 26.47 to 93.64 μg/mL against Vero cells and 74.29 to 225.40 μg/mL against C3A. The results of this study show that the essential oils from the gall of P. atlantica have low toxicity and moderate activity against fast growing M. smegmatis and M. aurum.Conclusion: These results substantiate their potential as used in the treatment of non-tuberculous mycobacterial infections. This finding is predictive of their activity against M. tuberculosis, therefore, further study might be considered to investigate the activity against pathogenic Mycobacterium strains.Key words: Pistacia atlantica, Essential oil, Antimycobacterial, Antioxidant, Cytotoxicity
