    Proteome Regulation during Olea europaea Fruit Development

    Widespread in the Mediterranean basin, Olea europaea trees are gaining worldwide popularity for the nutritional and cancer-protective properties of the oil, mechanically extracted from ripe fruits. Fruit development is a physiological process with remarkable impact on the modulation of the biosynthesis of compounds affecting the quality of the drupes as well as the final composition of the olive oil. Proteomics offers the possibility to dig deeper into the major changes during fruit development, including the important phase of ripening, and to classify temporal patterns of protein accumulation occurring during these complex physiological processes.In this work, we started monitoring the proteome variations associated with olive fruit development by using comparative proteomics coupled to mass spectrometry. Proteins extracted from drupes at three different developmental stages were separated on 2-DE and subjected to image analysis. 247 protein spots were revealed as differentially accumulated. Proteins were identified from a total of 121 spots and discussed in relation to olive drupe metabolic changes occurring during fruit development. In order to evaluate if changes observed at the protein level were consistent with changes of mRNAs, proteomic data produced in the present work were compared with transcriptomic data elaborated during previous studies.This study identifies a number of proteins responsible for quality traits of cv. Coratina, with particular regard to proteins associated to the metabolism of fatty acids, phenolic and aroma compounds. Proteins involved in fruit photosynthesis have been also identified and their pivotal contribution in oleogenesis has been discussed. To date, this study represents the first characterization of the olive fruit proteome during development, providing new insights into fruit metabolism and oil accumulation process

    Using jasmonates and salicylates to reduce losses within the fruit supply chain

    The fresh produce industry is constantly growing, due to increasing consumer demand. The shelf-life of some fruit, however, is relatively short, limited by microbial contamination or visual, textural and nutritional quality loss. Thus, techniques for reducing undesired microbial contamination, spoilage and decay, as well as maintaining product’s visual, textural and nutritional quality are in high demand at all steps within the supply chain. The postharvest use of signalling molecules, i.e. jasmonates and salicylates seems to have unexplored potential. The focus of this review is on the effects of treatment with jasmonates and salicylates on the fresh produce quality, defined by decay incidence and severity, chilling injury, maintenance of texture, visual quality, taste and aroma, and nutritional content. Postharvest treatments with jasmonates and salicylates have the ability to reduce decay by increasing fruit resistance to diseases and reducing chilling injury in numerous products. These treatments also possess the ability to improve other quality characteristics, i.e. appearance, texture maintenance and nutritional content. Furthermore, they can easily be combined with other treatments, e.g. heat treatment, ultrasound treatment. A good understanding of all the benefits and limitations related to the postharvest use of jasmonates and salicylates is needed, and relevant information has been reviewed in this paper

    Jarosite-related chemical processes and water ecotoxicity in simplified anaerobic microcosm wetlands

    Simulation experiments were conducted to examine whether jarosite is decomposed and toxic products are produced under anaerobic microcosm wetland conditions. The results show that jarosite was stable under water inundation in the microcosm wetland for a period of at least 56 days when no organic substance was added. However, jarosite became increasingly unstable with increasing amount of added organic matter. This resulted in entry of ferrous Fe into the soil solutions. Concentration of other heavy metals in the leachates was extremely low except for Mn. This is attributed to the maintenance of a high pH in the microcosm wetlands, which might cause re-precipitation of originally jarosite-borne heavy metals, if any. No acute toxicity was observed for leachate from the control (non organic matter-added treatment). However, leachates from various organic matter-added treatments show varying degrees of toxicity to the test organism and soluble Fe was likely to be the dominant metal of potential toxicity. Atmospheric exposure of leachate led to oxidation of ferrous Fe and precipitation of iron hydroxide, which caused a drop in leachate pH. This, in turn, inhibited further oxidation of ferrous Fe
