    SIRT1 in astrocytes regulates glucose metabolism and reproductive function

    Sirtuin 1 (Sirt1) is an NAD-dependent class III deacetylase that functions as a cellular energy sensor. In addition to its well-characterized effects in peripheral tissues, evidence suggests that SIRT1 in neurons plays a role in the central regulation of energy balance and reproduction, but no studies have addressed the contribution of astrocytes. We show here that overexpression of SIRT1 in astrocytes causes markedly increased food intake, body weight gain, and glucose intolerance, but expression of a deacetylase-deficient SIRT1 mutant decreases food intake and body weight and improves glucose tolerance, particularly in female mice. Paradoxically, the effect of these SIRT1 mutants on insulin tolerance was reversed, with overexpression showing greater insulin sensitivity. The mice overexpressing SIRT1 were more active, generated more heat, and had elevated oxygen consumption, possibly in compensation for the increased food intake. The female overexpressing mice were also more sensitive to diet-induced obesity. Reproductively, the mice expressing the deacetylase-deficient SIRT1 mutant had impaired estrous cycles, decreased LH surges, and fewer corpora lutea, indicating decreased ovulation. The GnRH neurons were responsive to kisspeptin stimulation, but hypothalamic expression of Kiss1 was reduced in the mutant mice. Our results showed that SIRT1 signaling in astrocytes can contribute to metabolic and reproductive regulation independent of SIRT1 effects in neurons.Fil: Choi, Irene.     Helicobacter pylori-derived extracellular vesicles increased in the gastric juices of gastric adenocarcinoma patients and induced inflammation mainly via specific targeting of gastric epithelial cells

    Evidence indicates that Helicobacter pylori is the causative agent of chronic gastritis and perhaps gastric malignancy. Extracellular vesicles (EVs) play an important role in the evolutional process of malignancy due to their genetic material cargo. We aimed to evaluate the clinical significance and biological mechanism of H. pylori EVs on the pathogenesis of gastric malignancy. We performed 16S rDNA-based metagenomic analysis of gastric juices either from endoscopic or surgical patients. From each sample of gastric juices, the bacteria and EVs were isolated. We evaluated the role of H. pylori EVs on the development of gastric inflammation in vitro and in vivo. IVIS spectrum and confocal microscopy were used to examine the distribution of EVs. The metagenomic analyses of the bacteria and EVs showed that Helicobacter and Streptococcus are the two major bacterial genera, and they were significantly increased in abundance in gastric cancer (GC) patients. H. pylori EVs are spherical and contain CagA and VacA. They can induce the production of tumor necrosis factor-��, interleukin (IL)-6 and IL-1�� by macrophages, and IL-8 by gastric epithelial cells. Also, EVs induce the expression of interferon gamma, IL-17 and EV-specific immunoglobulin Gs in vivo in mice. EVs were shown to infiltrate and remain in the mouse stomach for an extended time. H. pylori EVs, which are abundant in the gastric juices of GC patients, can induce inflammation and possibly cancer in the stomach, mainly via the production of inflammatory mediators from gastric epithelial cells after selective uptake by the cells. ? 2017 KSBMB.     Scanning Tunneling Spectroscopy in MgB 2

    We present extensive Scanning Tunneling Spectroscopy (STM/S) measurements at low temperatures in the multiband superconductor MgB2_2. We find a similar behavior in single crystalline samples and in single grains, which clearly shows the partial superconducting density of states of both the π\pi and σ\sigma bands of this material. The superconducting gaps corresponding to both bands are not single valued. Instead, we find a distribution of superconducting gaps centered around 1.9mV and 7.5mV, corresponding respectively to each set of bands. Interband scattering effects, leading to a single gap structure at 4mV and a smaller critical temperature can be observed in some locations on the surface. S-S junctions formed by pieces of MgB2_2 attached to the tip clearly show the subharmonic gap structure associated with this type of junctions. We discuss future developments and possible new effects associated with the multiband nature of superconductivity in this compound.Comment: 11 pages, 6 figures, submitted to Physica

    Toroidal mode number estimation of the edge-localized modes using the KSTAR 3-D electron cyclotron emission imaging system

    A new and more accurate technique is presented for determining the toroidal mode number n of edge-localized modes (ELMs) using two independent electron cyclotron emission imaging (ECEI) systems in the Korea Superconducting Tokamak Advanced Research (KSTAR) device. The technique involves the measurement of the poloidal spacing between adjacent ELM filaments, and of the pitch angle ?? O of filaments at the plasma outboard midplane. Equilibrium reconstruction verifies that ?? O is nearly constant and thus well-defined at the midplane edge. Estimates of n obtained using two ECEI systems agree well with n measured by the conventional technique employing an array of Mirnov coils.open3

    A Multilevel Product Model for Simulation-Based Design of Mechanical Systems

    This paper presents a multilevel product model that supports Simulation-Based Design (SBD) of mechanical systems, from pre liminary to detailed design stages The pnmary goal of the SBD is to achieve product designs featuring better performance and greater du rability and reliability through computer-based modeling, engineering analysis, and design trade-off. A Computer-Aided Design (CAD) model combined with engineering parameters and mathematical equations that simulate physical behavior of the mechanical system constitute its product model for SBD. For preliminary design, improvement of system performance, including dynamics and human factors, is usually the primary focus A CAD model with reasonably accurate physical parameters, such as mass properties of major components or assemblies, is defined as the base definition of the product model for SBD. A number of simulation models are derived from the base definition to sup port simulation of the mechanical system performance A parametric study can be conducted to search for design alternatives using dimen sion parameters created in the parameterized CAD model. The CAD model and base definition are then refined from the preliminary design stage to support intermediate designs. Intermediate designs will primarily focus on product subsystem performance. A product model is evolved by refining geometric representation of mechanical components in CAD, and expanding product assembly into parts and sub assemblies for further engineering analysis Component designs for performance, such as fatigue, mechanical reliability, and structural per formance, as well as maintainability are the primary focus in the detailed design stage. A detailed product model evolved from that of the previous design is needed In the detailed design stage, a systematic design trade-off method supports design improvement.     A design theory for e-service environments: The interoperability challenge

    The delivery of e-services across organizational boundaries poses a number of issues in terms of design of inter-organizational systems that support service delivery effectively. In this context interoperability emerges as a mandatory requirement for the design of Information Technology (IT) platforms supporting collaborative e-service environments. In this paper we address this issue by presenting a design theory for IT platforms supporting e-services based on both a deep understanding of the interoperability concept and a design research approach.     Harmonic maps from degenerating Riemann surfaces

    We study harmonic maps from degenerating Riemann surfaces with uniformly bounded energy and show the so-called generalized energy identity. We find conditions that are both necessary and sufficient for the compactness in W1,2W^{1,2} and C0C^{0} modulo bubbles of sequences of such maps.Comment: 27 page

    Extreme Electron-Phonon Coupling in Boron-based Layered Superconductors

    The phonon-mode decomposition of the electron-phonon coupling in the MgB2-like system Li_{1-x}BC is explored using first principles calculations. It is found that the high temperature superconductivity of such systems results from extremely strong coupling to only ~2% of the phonon modes. Novel characteristics of E_2g branches include (1) ``mode lambda'' values of 25 and greater compared to a mean of 0.4\sim 0.4 for other modes, (2) a precipitous Kohn anomaly, and (3) E_2g phonon linewidths within a factor of ~2 of the frequency itself, indicating impending breakdown of linear electron-phonon theory.     Quintessential Maldacena-Maoz Cosmologies

    Maldacena and Maoz have proposed a new approach to holographic cosmology based on Euclidean manifolds with disconnected boundaries. This approach appears, however, to be in conflict with the known geometric results [the Witten-Yau theorem and its extensions] on spaces with boundaries of non-negative scalar curvature. We show precisely how the Maldacena-Maoz approach evades these theorems. We also exhibit Maldacena-Maoz cosmologies with [cosmologically] more natural matter content, namely quintessence instead of Yang-Mills fields, thereby demonstrating that these cosmologies do not depend on a special choice of matter to split the Euclidean boundary.     Influence of uncorrelated overlayers on the magnetism in thin itinerant-electron films

    The influence of uncorrelated (nonmagnetic) overlayers on the magnetic properties of thin itinerant-electron films is investigated within the single-band Hubbard model. The Coulomb correlation between the electrons in the ferromagnetic layers is treated by using the spectral density approach (SDA). It is found that the presence of nonmagnetic layers has a strong effect on the magnetic properties of thin films. The Curie temperatures of very thin films are modified by the uncorrelated overlayers. The quasiparticle density of states is used to analyze the results. In addition, the coupling between the ferromagnetic layers and the nonmagnetic layers is discussed in detail. The coupling depends on the band occupation of the nonmagnetic layers, while it is almost independent of the number of the nonmagnetic layers. The induced polarization in the nonmagnetic layers shows a long-range decreasing oscillatory behavior and it depends on the coupling between ferromagnetic and nonmagnetic layers.Comment: 9 pages, RevTex, 6 figures, for related work see: http://orion.physik.hu-berlin.d
