    Expression analysis of transcription factors from the interaction between cacao and Moniliophthora perniciosa (Tricholomataceae)

    Cacao (Theobroma cacao) is one of the most important tropical crops; however, production is threatened by numerous pathogens, including the hemibiotrophic fungus Moniliophthora perniciosa, which causes witches' broom disease. To understand the mechanisms that lead to the development of this disease in cacao, we focused our attention on cacao transcription factors (TFs), which act as master regulators of cellular processes and are important for the fine-tuning of plant defense responses. We developed a macroarray with 88 TF cDNA from previously obtained cacao-M. perniciosa interaction libraries. Seventy-two TFs were found differentially expressed between the susceptible (Catongo) and resistant (TSH1188) genotypes and/or during the disease time course - from 24 h to 30 days after infection. Most of the differentially expressed TFs belonged to the bZIP, MYB and WRKY families and presented opposite expression patterns in susceptible and resistant cacao-M. perniciosa interactions (i.e., up-regulated in Catongo and down-regulated in TSH1188). The results of the macroarray were confirmed for bZIP and WRKY TFs by real-time PCR. These differentially expressed TFs are good candidates for subsequent functional analysis as well as for plant engineering.     A simple boiling-based DNA extraction for RAPD profiling of landfarm soil to provide representative metagenomic content

    Landfarm soils are employed in industrial and petrochemical residue bioremediation. This process induces selective pressure directed towards microorganisms capable of degrading toxic compounds. Detailed description of taxa in these environments is difficult due to a lack of knowledge of culture conditions required for unknown microorganisms. A metagenomic approach permits identification of organisms without the need for culture. However, a DNA extraction step is first required, which can bias taxonomic representativeness and interfere with cloning steps by extracting interference substances. We developed a simplified DNA extraction procedure coupled with metagenomic DNA amplification in an effort to overcome these limitations. The amplified sequences were used to generate a metagenomic data set and the taxonomic and functional representativeness were evaluated in comparison with a data set built with DNA extracted by conventional methods.     Photosynthesis, stomatal conductance and leaf water potential in crabwood (Carapa guianensis)

    Leaf water potential is one of the most important factors affecting stomatal functioning. The aim of this study was to assess the effect of variation in diurnal irradiance and vapour pressure deficit on photosynthesis (A), stomatal conductance (g s) and leaf water potential (psi) in Carapa guianensis (Aubl.). Data were collected from 07:00 to 17:00 h. Photosynthetic rates reached a maximum (2.5 µmol m-2 s-1) at 10:00 h, thereafter declined to a minimum of 1 µmol m-2 s-1 at 16:00 h. Stomatal conductance oscillated during the day, from 0.04 mol m-2 s-1 (at midday) to 0.02.mol.m-2.s-1 at the end of the afternoon. Leaf water potential was higher early in the morning (-0.3 MPa) and lower (-0.75 MPa) at mid-afternoon (14:30 -15:00 h). After reaching a minimum, psi increased up to -0.64 MPa at sunset. Photosynthetic rates increased linearly as a function of g s (P < 0.01). Also there was a positive relationship between psi and g s (P< 0.01). Photosynthetic rates declined during the day after reaching a peak early in the morning, which makes clear that environmental factors that influence psi greatly affect carbon assimilation of C. guianensis.O potencial hídrico da folha é um dos fatores mais importantes que afetam o funcionamento dos estômatos. O objetivo deste trabalho foi avaliar o efeito da variação diurna na irradiância e déficit de pressão de vapor (DPV) na fotossíntese (A), condutância estomática (g s) e potencial hídrico da folha (psi) em Carapa guianensis (Aubl.). Os dados foram coletados de 07:00 às 17:00 h. A taxa fotossintética atingiu um valor máximo (2,5 µmol m-2 s-1) às 10:00 h, depois declinou até atingir um mínimo de 1 µmolm-2 s-1 às 16:00 h. A condutância estomática oscilou durante o dia, de 0,04 molm-2s-1 (ao meio dia) para 0,02 molm-2s- 1 no final da tarde. O potencial hídrico da folha foi máximo nas primeiras horas do dia (-0,3 MPa) e mínimo (-0,75 MPa) no meio da tarde (14:30 a 15:00 h). Após ter alcançado um mínimo, o psi aumentou até -0,64 MPa no fim da tarde. A taxa fotossintética aumentou linearmente em função do g s (P < 0,01). Também houve uma relação positiva entre psi e g s (P < 0,01). A taxa fotossintética declinou durante o dia após ter alcançado um pico no início da manhã, demonstrando que os fatores ambientais que afetam o psi têm efeito significativo na assimilação do carbono de C. guianensis

    A Região do Planalto Ocidental Paulista tem se destacado como pólo produtor de borracha natural. Visando fornecer informações sobre a adaptação dos clones de seringueira [Hevea brasiliensis (Wildd. ex. Adr. de Juss) Müell. Arg.] RRIM 600, GT 1, RRIM 701, IAN 873 e PB 235, em São José do Rio Preto (SP), realizaram-se avaliações de alguns caracteres de sua biologia, pelo acompanhamento das trocas gasosas e mensuração do desenvolvimento vegetativo durante os primeiros 18 meses após o enxerto sobre Tjir 16. A altura de RRIM 600 atingiu, em média, 3,74 m e o diâmetro do caule de IAN 873 e RRIM 600, 2,50 cm. PB 235 apresentou o menor índice relativo de crescimento durante o período. No período úmido, os valores das trocas gasosas não diferiram significativamente entre os clones, com valores médios da taxa fotossintética de 9,45 mmol.m-2.s-1; para a taxa de transpiração, 3,84 mmol.m-2.s-1, e para a condutância estomática, de 0,096 mol.m-2.s-1. Diferenças entre os valores de trocas gasosas ocorreram apenas no período seco, com redução mais acentuada para PB 235. Considerando o conjunto dos caracteres analisados, o desempenho dos clones IAN 873 e RRIM 600 foi superior, e do clone PB 235, inferior.<br>The region of São Paulo Occidental Plateau is well known for its part as a major producer of natural rubber. In order to obtain information about adaptation of rubber tree clones [Hevea brasiliensis (Wildd. ex. Adr. de Juss) Müell. Arg.] RRIM 600, RRIM 701, IAN 873, PB 235 and GT 1, in the city of São José do Rio Preto some biologic characteristics were evaluated. Analysis were performed by monitoring gas exchanges and by measuring vegetative development, during the 18 months after engraft on the Tjir 16 clonal rootstocks. The height of RRIM 600 reached a mean of 3,74 m, and the diameter of the stem of IAN 873 and RRIM 600 reached the mean of 2,50 cm. Clone PB 235 presented the lowest relative growth during the period. During the wet period, the values of the gas exchange did not show significant variation among the clones; the mean value of photosynthetic rates was 9,45 mmol.m-2.s-1; of transpiration rates was 3,84 mmol.m-2 s-1, and of stomatic conductance was 0,096 mol.m-2 s-1. Differences in values of gas exchanges were observed only during the dry period, with notably large reduction for clone PB 235. The characteristics analyzed lead to the conclusion that performances of clones IAN 873 and RRIM 600 were superior, and that of clone PB 235 were inferior
