    A Third Measure-Metastable State in the Dynamics of Spontaneous Shape Change in Healthy Human's White Cells

    Human polymorphonuclear leucocytes, PMN, are highly motile cells with average 12-15 µm diameters and prominent, loboid nuclei. They are produced in the bone marrow, are essential for host defense, and are the most populous of white blood cell types. PMN also participate in acute and chronic inflammatory processes, in the regulation of the immune response, in angiogenesis, and interact with tumors. To accommodate these varied functions, their behavior is adaptive, but still definable in terms of a set of behavioral states. PMN morphodynamics have generally involved a non-equilibrium stationary, spheroid Idling state that transitions to an activated, ellipsoid translocating state in response to chemical signals. These two behavioral shape-states, spheroid and ellipsoid, are generally recognized as making up the vocabulary of a healthy PMN. A third, “random” state has occasionally been reported as associated with disease states. I have observed this third, Treadmilling state, in PMN from healthy subjects, the cells demonstrating metastable dynamical behaviors known to anticipate phase transitions in mathematical, physical, and biological systems. For this study, human PMN were microscopically imaged and analyzed as single living cells. I used a microscope with a novel high aperture, cardioid annular condenser with better than 100 nanometer resolution of simultaneous, mixed dark field and intrinsic fluorescent images to record shape changes in 189 living PMNs. Relative radial roundness, R(t), served as a computable order parameter. Comparison of R(t) series of 10 cells in the Idling and 10 in the Treadmilling state reveals the robustness of the “random” appearing Treadmilling state, and the emergence of behaviors observed in the neighborhood of global state transitions, including increased correlation length and variance (divergence), sudden jumps, mixed phases, bimodality, power spectral scaling and temporal slowing.     Incidence, clinical characteristics and management of inflammatory bowel disease in Spain: large-scale epidemiological study

    (1) Aims: To assess the incidence of inflammatory bowel disease (IBD) in Spain, to describe the main epidemiological and clinical characteristics at diagnosis and the evolution of the disease, and to explore the use of drug treatments. (2) Methods: Prospective, population-based nationwide registry. Adult patients diagnosed with IBD—Crohn’s disease (CD), ulcerative colitis (UC) or IBD unclassified (IBD-U)—during 2017 in Spain were included and were followed-up for 1 year. (3) Results: We identified 3611 incident cases of IBD diagnosed during 2017 in 108 hospitals covering over 22 million inhabitants. The overall incidence (cases/100, 000 person-years) was 16 for IBD, 7.5 for CD, 8 for UC, and 0.5 for IBD-U; 53% of patients were male and median age was 43 years (interquartile range = 31–56 years). During a median 12-month follow-up, 34% of patients were treated with systemic steroids, 25% with immunomodulators, 15% with biologics and 5.6% underwent surgery. The percentage of patients under these treatments was significantly higher in CD than UC and IBD-U. Use of systemic steroids and biologics was significantly higher in hospitals with high resources. In total, 28% of patients were hospitalized (35% CD and 22% UC patients, p < 0.01). (4) Conclusion: The incidence of IBD in Spain is rather high and similar to that reported in Northern Europe. IBD patients require substantial therapeutic resources, which are greater in CD and in hospitals with high resources, and much higher than previously reported.     Explicar cómo y en qué condiciones fueron construidos algunos de los instrumentos científicos más emblemáticos del mundo moderno sigue siendo un desafío para los historiadores de la ciencia y de la tecnología. Este principio afecta también a los instrumentos náuticos. Así, el objetivo del presente artículo es considerar el “cómo” y el “en qué condiciones”, en un sentido amplio, analizando uno de estos artefactos: las cartas náuticas construidas durante la expansión marítima europea. Originalidad: La originalidad del texto no reside únicamente en llevar a cabo una reconstrucción de este instrumento en Portugal y España que aporte nuevos datos sobre su proceso de fabricación y sus fabricantes, sino también en arrojar luz sobre antiguos debates historiográficos que subyacen a la génesis de la ciencia moderna, tales como la difícil colaboración entre hombres prácticos y hombres teóricos; la aportación de comunidades de artesanos al mundo del conocimiento; el papel que los saberes prácticos tuvieron en el establecimiento de nuevas formas de estandarización en ciencia; la adopción de una metodología empírica para el dominio del mundo o la construcción de grandes imperios globales cimentados sobre la base del trabajo manual. Metodología: El análisis de estos debates requiere una aproximación metodológica que atienda a las categorías y los conceptos principales puestos en circulación tanto por la historia como por la filosofía de la ciencia más reciente. En definitiva, se trata de atender a las condiciones que permitieron el establecimiento de acuerdos (y también de controversias) epistemológicos entre diferentes comunidades de conocimiento tomando un instrumento náutico como “mediador”. This principle also affects nautical instruments. Thus, the objective of this article is to consider the “how” and the “under what conditions”, in a broad sense, by examining one of these devices: the nautical charts developed during the European maritime expansion. Originality: The originality of this article lies not only in the reconstruction of this instrument in Portugal and Spain, providing new information about its development process and its manufacturers, but also in shedding light on old historiographical debates that underlie the genesis of modern science, such as the difficult collaboration between practical men and theoretical men; the contribution of artisan communities to the world of knowledge; the role that practical knowledge played in the establishment of new forms of standardization in science; the adoption of an empirical methodology for achieving mastery of the world or the construction of large global empires founded upon manual labor. Methodology: Analyzing these debates requires a methodological approach that addresses the main categories and concepts put into circulation by the most recent history and philosophy of science. In short, unveiling the conditions that enabled the establishment of epistemological agreements (and also disputes) between different knowledge communities by taking a nautical instrument as “mediator.” Conclusions: The article concludes that examining empirical knowledge cultures and their material achievements are a sine qua non condition to understand the genesis of European scientific modernity and the construction of a global worldExplicar como e em que condições foram construídos alguns dos instrumentos científicos mais emblemáticos do mundo moderno continua sendo um desafio para os historiadores da ciência e da tecnologia.     RAPD analysis of cultivated and wild yellow nutsedge (Cyperus esculentus L.)

    Cultivated and weedy clones of yellow nutsedge were analyzed using random amplified polymorphic DNA (RAPD) markers to assess the polymorphism within the species and determine if this approach was suitable for identification of cultivar and wild populations. The RAPD markers unambiguously identified all studied clones. Nei-Li similarities were computed and used in an unweighted pair group method using arithmetic average (UPGMA) cluster analyses. Cultivated and weedy clones were clustered in two groups, but two cultivated clones were more closely related to weedy clones than to cultivated clones. The results showed a high level of genetic variability among the clones tested, particularly among the cultivated ones. Identification of yellow nutsedge cultivars and analysis of genetic diversity within and among weedy populations is possible by using only a small number of primers.     La chufa, Cyperus esculentus L. var. sativus Boeck., es una planta cultivada en la comarca de Horta Nord de la Comunidad Valenciana fundamentalmente para obtener a partir de sus tubérculos el refresco conocido como horchata. El cultivo ocupa una superficie aproximada de unas 600 has lo que supone una producción de unas 7300 t. A nivel mundial Cyperus esculentus L. está considerada como una de las veinte malas hierbas más importantes, pero es prácticamente desconocida como planta cultivada. En la primera parte de esta obra se analiza la importancia económica de esta planta en nuestro país, se describe botánicamente y se estudia su fisiología, describiéndose ampliamente todos los aspectos agronómicos que conciernen a su cultivo. En la segunda parte se presentan los estudios realizados por nuestro equipo de trabajo en aspectos nutricionales, fisiológicos (control del encamado, retardadores del crecimiento, hipotética latencia de los tubérculos) y sobre diversas tecnologías de manejo agronómico (fecha de plantación, textura del suelo, desyerbe químico, ...). La tercera parte se dedica a los trabajos de selección clonal y tipificación agronómica llevados a cabo por nuestro grupo a partir de material vegetal tanto de origen local como importado, estudiando la morfología y fisiología de las plantas, la morfología de los tubérculos, las características productivas, la composición química de los tubérculos y horchatas, análisis sensoriales de las horchatas, y las características bioquímicas de los diversos clones, analizando la heredabilidad de todos los caracteres estudiados

    Gibberellic Acid Concentrations in Seed Propagated Artichoke

    In Spain, the artichoke crop is interesting for autumn-winter production. With vegetative propagated cultivars, like ‘Blanca de Tudela’ or ‘Violeta de Provenza’, the harvest starts at the beginning of autumn and finishes at the end of spring. The currently available seed propagated cultivars are not as early as the asexually propagated ones mentioned above. For this reason, to obtain production in the autumn-winter cycle with seed propagated cultivars, it is necessary to use the earlier ones, to plant at the beginning of the summer and to spray plants with gibberellic acid (GA) to substitute the lack of vernalization requirements. The trials were conducted during three seasons. In 1999-2000, using the cultivar ‘A-106’ and ‘A-108’, two concentrations of GA (10 and 30 ppm) were compared with untreated plants. In the 2000-2001 season, using the cultivars ‘Lorca’ and ‘Nun 98465’ and in the 2001-2002 season, using the cultivars ‘A-106’ and ‘Nun 9409’, three concentrations of GA (10, 30 and 50 ppm) were also compared with untreated plants. The cultivars ‘A-106’ and ‘Lorca’ are similar to Imperial Star and earlier than the hybrids ‘Nun 98465’ and ‘Nun 9409’. In the earlier cultivars, GA applications increased early yields. No significant differences in yield or head weight were found among the GA concentrations tested.
