    Delayed kinetics of poliovirus RNA synthesis in a human cell line with reduced levels of hnRNP C proteins.

    The hnRNP C heterotetramer [(C1(3))C2] binds RNA polymerase II transcripts in the nucleus, along with other proteins of the core hnRNP complex, and plays an important role in mRNA biogenesis and transport. Infection of HeLa cells with poliovirus causes hnRNP C to re-localize from the nucleus, where it is normally retained during interphase, to the cytoplasm. We have proposed that in the cytoplasm, the protein isoforms of hnRNP C participate in the recognition of viral specific RNAs by the poliovirus replication proteins and/or in the assembly of membrane-bound RNA replication complexes. In SK-OV-3 cells, which express reduced levels of hnRNP C compared to HeLa cells or 293 cells, the kinetics of poliovirus replication are delayed. hnRNP C is also re-localized from the nucleus to the cytoplasm in SK-OV-3 cells infected with poliovirus. Increased expression of hnRNP C in SK-OV-3 cells by transient transfection increases the rate of virus production and overall yield over that seen in mock-transfected cells. We propose that hnRNP C interacts with poliovirus RNA and replication proteins to increase the efficiency of viral genomic RNA synthesis

Providing students with an opportunity to participate in discussion helps students develop an understanding of various dynamics at play in group communication and shows how group communication affects interpersonal relationships. The outcome of the activity is to improve students understanding of group decision making resulting in better decisions, less conflict, and more efficient use of time when working in groups

    Construction of a "mutagenesis cartridge" for poliovirus genome-linked viral protein: Isolation and characterization of viable and nonviable mutants

    By following a strategy of genetic analysis of poliovirus, we have constructed a synthetic "mutagenesis cartridge" spanning the genome-linked viral protein coding region and flanking cleavage sites in an infectious cDNA clone of the type 1 (Mahoney) genome. The insertion of new restriction sites within the infectious clone has allowed us to replace the wild-type sequences with short complementary pairs of synthetic oligonucleotides containing various mutations. A set of mutations have been made that create methionine codons within the genome-linked viral protein region. The resulting viruses have growth characteristics similar to wild type. Experiments that led to an alteration of the tyrosine residue responsible for the linkage to RNA have resulted in nonviable virus. In one mutant, proteolytic processing assayed in vitro appeared unimpaired by the mutation. We suggest that the position of the tyrosine residue is important for genome-linked viral protein function(s)

    Discovery of the Putative Pulsar and Wind Nebula Associated with the TeV Gamma-ray Source HESS J1813-178

    We present a Chandra X-ray observation of G12.82-0.02, a shell-like radio supernova remnant coincident with the TeV gamma-ray source HESS J1813-178. We resolve the X-ray emission from the co-located ASCA source into a point source surrounded by structured diffuse emission that fills the interior of the radio shell. The morphology of the diffuse emission strongly resembles that of a pulsar wind nebula. The spectrum of the compact source is well-characterized by a power-law with index Gamma approx 1.3, typical of young and energetic rotation-powered pulsars. For a distance of 4.5 kpc, consistent with the X-ray absorption and an association with the nearby star formation region W33, the 2-10 keV X-ray luminosities of the putative pulsar and nebula are L(PSR) = 3.2E33 ergs/s and L(PWN) = 1.4E34 ergs/s, respectively. Both the flux ratio of L(PWN)/L(PSR) = 4.3 and the total luminosity of this system predict a pulsar spin-down power of Edot > 1E37 ergs/s, placing it within the ten most energetic young pulsars in the Galaxy. A deep search for radio pulsations using the Parkes telescope sets an upper-limit of approx 0.07 mJy at 1.4 GHz for periods >~ 50 ms. We discuss the energetics of this source, and consider briefly the proximity of bright H2 regions to this and several other HESS sources, which may produce their TeV emission via inverse Compton scattering.Comment: 7 pages, 6 figure, Latex, emulateapj style.     THE EFFECTS OF FATIGUE ON SOFTBALL PITCHER FROM PITCHING MULTIPLE GAMES

    The purpose of this study was to examine the effects of fatigue on softball pitching from pitching back to back games. Players were asked to throw 15 balls in each inning for seven innings and for a total of two games. The results showed fastball’s velocity in the first inning of the first game (25.4 ± 2.5 m/s) had significantly decreased after a game and a half (24.3 ± 2.7 m/s; p = 0.00). Hence, this study suggests that the pitchers started to show signs of fatigue after this amount of pitches (165 pitches), so softball coaches may take this into consideration and make necessary pitching change. Given the established relationship between fatigue and injury in pitchers, the Amateur Softball Association may use this information to implement a pitch count to protect softball pitchers from overuse injur

    High-redshift Cool-core Galaxy Clusters Detected via the Sunyaev-Zel'dovich Effect in the South Pole Telescope Survey

    We report the first investigation of cool-core properties of galaxy clusters selected via their Sunyaev-Zel'dovich (SZ) effect. We use 13 galaxy clusters uniformly selected from 178 deg2 observed with the South Pole Telescope (SPT) and followed up by the Chandra X-ray Observatory. They form an approximately mass-limited sample (>3 × 10^(14) M_☉ h^(–1)_(70)) spanning redshifts 0.3 0.5 cool-core clusters, including two strong cool cores. This rules out the hypothesis that there are no z > 0.5 clusters that qualify as strong cool cores at the 5.4σ level. The fraction of strong cool-core clusters in the SPT sample in this redshift regime is between 7% and 56% (95% confidence). Although the SPT selection function is significantly different from the X-ray samples, the high-z c_(SB) distribution for the SPT sample is statistically consistent with that of X-ray-selected samples at both low and high redshifts. The cool-core strength is inversely correlated with the offset between the brightest cluster galaxy and the X-ray centroid, providing evidence that the dynamical state affects the cool-core strength of the cluster. Larger SZ-selected samples will be crucial in understanding the evolution of cluster cool cores over cosmic time

This manuscript provides a rationale for a program wellness model and outlines a proposed series of interventions aimed at increasing students’ understanding, ability to self-assess, and strategies related to self-care and wellness
