    Differences in the epidemiology of theileriosis on smallholder dairy farms in contrasting agro-ecological and grazing strata of highland Kenya

    A prospective cohort study was conducted in five purposively-sampled agro-ecological zone (AEZ)-grazing system strata in Murang’a District, Kenya, between March 1995 and June 1996. The study strata were selected based on a preliminary characterization study to represent the widest range of risks to East Coast fever (ECF) in the District and included zero-grazing and open-grazing farms. In total, 225 calves from 188 smallholder farms were examined from birth to 6 months of age and visited within the first 2 weeks of life and thereafter at bi-weekly intervals for up to 14 visits. The purpose of the study was to characterize the differences in epidemiology (risks of infection, morbidity and mortality) and potential control of ECF between the selected strata. Evidence of Theileria parva infection was assessed by increased antibody levels as measured in an indirect ELISA assay by the percent positivity (PP) of serum samples relative to a strong positive reference serum. Sero-conversion risks of T. parva were highest in the open-grazing strata. Antibody prevalence in adult cattle and ECF morbidity and mortality risks were also highest in open-grazing strata. While different, all five AEZ-grazing strata were considered to be endemically unstable for ECF. East Coast fever challenge was low in all zero-grazing strata and this challenge is likely to remain low due to continuing intensification of smallholder farming in the central highlands.     The Effects of Freezing on Faecal Microbiota as Determined Using MiSeq Sequencing and Culture-Based Investigations

    peer-reviewedBackground High-throughput sequencing has enabled detailed insights into complex microbial environments, including the human gut microbiota. The accuracy of the sequencing data however, is reliant upon appropriate storage of the samples prior to DNA extraction. The aim of this study was to conduct the first MiSeq sequencing investigation into the effects of faecal storage on the microbiota, compared to fresh samples. Culture-based analysis was also completed. Methods Seven faecal samples were collected from healthy adults. Samples were separated into fresh (DNA extracted immediately), snap frozen on dry ice and frozen for 7 days at -80°C prior to DNA extraction or samples frozen at -80°C for 7 days before DNA extraction. Sequencing was completed on the Illumina MiSeq platform. Culturing of total aerobes, anaerobes and bifidobacteria was also completed. Results No significant differences at phylum or family levels between the treatment groups occurred. At genus level only Faecalibacterium and Leuconostoc were significantly different in the fresh samples compared to the snap frozen group (p = 0.0298; p = 0.0330 respectively). Diversity analysis indicated that samples clustered based on the individual donor, rather than by storage group. No significant differences occurred in the culture-based analysis between the fresh, snap or -80°C frozen samples. Conclusions Using the MiSeq platform coupled with culture-based analysis, this study highlighted that limited significant changes in microbiota occur following rapid freezing of faecal samples prior to DNA extraction.     Stabilization of self-focusing instability in wide-aperture semiconductor lasers

    A mechanism for the stabilization of the output of filamentary broad-area edge-emitting semiconductor lasers is analyzed experimentally and theoretically. This mechanism occurs when the carrier density is profiled in the transverse direction. The laser structure consisted of a wide-aperture edge-emitting laser diode operating in pulsed mode to avoid thermal guiding effects. The injection current profile was modified from the usual step-function case to a Lorentzian-like profile through the inclusion of a 10 mum p-type epitaxial spreading layer. The resulting nonlinear transverse mode is described and the possibility of its observation in two transverse dimensions is discussed

    Changes in structural network topology correlate with severity of hallucinatory behavior in Parkinson's disease

    Inefficient integration between bottom-up visual input and higher order visual processing regions is implicated in visual hallucinations in Parkinson's disease (PD). Here, we investigated white matter contributions to this perceptual imbalance hypothesis. Twenty-nine PD patients were assessed for hallucinatory behavior. Hallucination severity was correlated to connectivity strength of the network using the network-based statistic approach. The results showed that hallucination severity was associated with reduced connectivity within a subnetwork that included the majority of the diverse club. This network showed overall greater between-module scores compared with nodes not associated with hallucination severity. Reduced between-module connectivity in the lateral occipital cortex, insula, and pars orbitalis and decreased within-module connectivity in the prefrontal, somatosensory, and primary visual cortices were associated with hallucination severity. Conversely, hallucination severity was associated with increased between- and within-module connectivity in the orbitofrontal and temporal cortex, as well as regions comprising the dorsal attentional and default mode network. These results suggest that hallucination severity is associated with marked alterations in structural network topology with changes in participation along the perceptual hierarchy. This may result in the inefficient transfer of information that gives rise to hallucinations in PD.     Altered cerebrovascular response to acute exercise in patients with Huntington’s Disease

    Objective. To determine whether a single session of exercise is sufficient to induce cerebral adaptations in individuals with Huntington’s disease, and explore the time dynamics of any acute cerebrovascular response. Methods. In this case-control study we employed arterial-spin labelling magnetic resonance imaging in 19 HD gene-positive participants (32-65 years old, 13 males) and 19 controls (29-63 years old, 10 males) matched for age, gender, body mass index and self-reported activity levels, to measure global and regional perfusion in response to 20-minutes of moderate intensity cycling. Cerebral perfusion was measured at baseline and 15-, 40- and 60-minutes after exercise cessation. Results. Relative to baseline, cerebral perfusion increased in HD patients yet was unchanged in control participants in the precentral gyrus, middle frontal gyrus and hippocampus 40 minutes after exercise cessation (+15 to +32.5% change in HD participants, -7.7 to 0.8% change in controls). CAG repeat length predicted the change in the precentral gyrus, and the intensity of the exercise intervention predicted hippocampal perfusion change in HD participants. In both groups, exercise increased hippocampal blood flow 60-minutes after exercise cessation. Conclusions. Here we demonstrate the utility of acute exercise as a clinically sensitive experimental paradigm to modulate the cerebrovasculature. Twenty minutes of aerobic exercise induced transient cerebrovascular adaptations in the hippocampus and cortex selectively in HD participants and likely represents latent neuropathology not evident at rest

    High speed video capture for mobile phone cameras

    We consider an electromechanical model for the operation of a voice coil motor in a mobile phone camera, with the aim of optimizing how a lens can be moved to a desired focusing motion. Although a methodology is developed for optimizing lens shift, there is some concern about the experimentally-determined model parameters that are at our disposal. Central to the model is the value of the estimated magnetic force constant, Kf: its value determines how far it is actually possible to move lens, but it appears that, from the value given, it would not be possible to shift the lens through the displacements desired. Furthermore, earlier experiments have also estimated the value of the back EMF constant, Kg , to be roughly five times greater than Kf, even though we present two theoretical arguments that show that Kf = Kg: a conclusion supported by readily-available manufacturers’ data

    Cohort Profile: The Mater-University of Queensland Study of Pregnancy (MUSP)

    How did the study come about? At the initiative of three hospital-based obstetricians one of the authors (JMN) and another sociologist attended a 1978 meeting to explore the potential for collaborative research. No specific project was proposed by the obstetricians. Two of the obstetricians had been trained in Scotland; one in Aberdeen where he had had some familiarity with the work of obstetrician Dugald Baird and sociologist Raymond Illsely. The obstetricians argued for an agenda which was clinically oriented. The two sociologists were more focussed on understanding how some social constructs (e.g. social class) might be relevant to explaining health outcomes. The initial year of the study was characterised by frequent (at least weekly) meetings at which the aims and substance of the proposed study were debated vigorously. As an initial decision the team focused on factors associated with adverse pregnancy outcomes. The first major project was to be a 3-5 year longitudinal study of pregnant women which would include a 6-month child post-natal follow-up as the main outcome of interest

    Non-invasive measurement of hepatic venous oxygen saturation (ShvOâ‚‚) with quantitative susceptibility mapping in normal mouse liver and livers bearing colorectal metastases

    PURPOSE: The purpose of this prospective study was to investigate the potential of QSM to noninvasively measure hepatic venous oxygen saturation (ShvO2). Materials & Methods: All animal studies were performed in accordance with the UK Home Office Animals Science Procedures Act (1986) and UK National Cancer Research Institute (NCRI) guidelines. QSM data was acquired from a cohort of mice (n=10) under both normoxic (medical air, 21% O2/balance N), and hyperoxic conditions (100% O2). Susceptibility measurements were taken from large branches of the portal and hepatic vein under each condition and were used to calculate venous oxygen saturation in each vessel. Blood was extracted from the IVC of three mice under norm- and hyperoxic conditions, and oxygen saturation was measured using a blood gas analyser to act as a gold standard. QSM data was also acquired from a cohort of mice bearing colorectal liver metastases (CRLM). SvO2 was calculated from susceptibility measurements made in the portal and hepatic veins, and compared to the healthy animals. RESULTS: SvO2 calculated from QSM measurements showed a significant increase of 14.93% in the portal vein (p < 0.05), and an increase of 21.39% in the hepatic vein (p < 0.01). Calculated results showed excellent agreement with those from the blood gas analyser (26.14% increase). ShvO2 was significantly lower in the disease cohort (30.18 ± 11.6%), than the healthy animals (52.67 ± 17.8%) (p < 0.05), but differences in the portal vein were not significant. CONCLUSION: QSM is a feasible tool for non-invasively measuring hepatic venous oxygen saturation and can detect differences in oxygen consumption in livers bearing colorectal metastases
