    A comparison of serum and plasma levels of vascular endothelial growth factor during the menstrual cycle in healthy female volunteers

    Angiogenesis is the formation of new blood vessels from the existing vasculature, and is essential for the growth and metastasis of most solid tumours. One of the most important growth factors involved in the angiogenesis process is vascular endothelial growth factor. Vascular endothelial growth factor expression has been shown to be regulated by female hormones in breast cancer cell lines, and two previous authors have reported on cyclical variations in serum vascular endothelial growth factor concentrations with conflicting results. No work has been performed on variations in plasma levels of vascular endothelial growth factor during the menstrual cycle. We therefore conducted the first prospective trial to compare serum and plasma levels of vascular endothelial growth factor in healthy pre-menopausal volunteers. Twenty healthy pre-menopausal women were recruited and had blood samples taken over one menstrual cycle with an average of eight samples taken per patient. Plasma and serum samples were then analysed for sex hormones and vascular endothelial growth factor 165. Serum vascular endothelial growth factor levels were found to be significantly higher than plasma vascular endothelial growth factor levels (P<0.005). We found no significant difference between serum and plasma vascular endothelial growth factor in the luteal and follicular phases of the cycle. The majority of the measurements for plasma levels of vascular endothelial growth factor at all phases of the cycle were under the limit of detection of the vascular endothelial growth factor ELISA kit. We found no significant correlation between plasma or serum levels of vascular endothelial growth factor and either FSH, LH, Oestradiol or Progesterone levels. This study has demonstrated no difference in serum concentrations of vascular endothelial growth factor during the different phases of the menstrual cycle in a group of healthy volunteers. We also demonstrated no obvious difference in plasma concentrations of vascular endothelial growth factor between the phases of the cycle, but most of the measurements were below the level of accuracy reported by the ELISA kit manufacturer. With the sensitivity of this ELISA test, therefore, we must still regard the question of whether there is a variation in plasma concentrations of vascular endothelial growth factor throughout the menstrual cycle as unanswered

    Land use influences on acidification and recovery of freshwaters in Galloway, south-west Scotland

    International audienceThe long term response of surface waters to changes in sulphur deposition and afforestation is investigated for three upland river systems in the Galloway region of south-west Scotland. From 1984-1999, these rivers exhibited a statistically significant decline in non-marine sulphate concentrations in response to reduced acid deposition. This reduction in non-marine sulphate was, however, insufficient to induce a pH recovery over the period. A statistically significant increase in river pH was observed between 1956-1970 (0.05 yr-1) when subsidised agricultural lime payments were at a maximum. In 1976, this subsidy ceased and surface waters have progressively acidified. In addition, climatic change is found to influence long-term trends in pH. Mean annual pH was greatest during a dry period between 1969-1973 when total annual discharge was low. Thereafter, pH declined gradually in response to higher rainfall and increased total annual discharge. Overall, surface waters draining the afforested catchments of the Rivers Cree and Bladnoch are more acid than those draining the moorland catchment of the Luce. These results indicate that in afforested catchments, current reductions in sulphur emissions have not led to an observed improvement in the acid status of surface waters. Forestry, therefore, represents a confounding factor with regard to chemical recovery from acidification in this region.     Validation of northern latitude Tropospheric Emission Spectrometer stare ozone profiles with ARC-IONS sondes during ARCTAS: sensitivity, bias and error analysis

    We compare Tropospheric Emission Spectrometer (TES) versions 3 and 4, V003 and V004, respectively, nadir-stare ozone profiles with ozonesonde profiles from the Arctic Intensive Ozonesonde Network Study (ARCIONS, http://croc.gsfc.nasa.gov/arcions/ during the Arctic Research on the Composition of the Troposphere from Aircraft and Satellites (ARCTAS) field mission. The ozonesonde data are from launches timed to match Aura's overpass, where 11 coincidences spanned 44° N to 71° N from April to July 2008. Using the TES "stare" observation mode, 32 observations are taken over each coincidental ozonesonde launch. By effectively sampling the same air mass 32 times, comparisons are made between the empirically-calculated random errors to the expected random errors from measurement noise, temperature and interfering species, such as water. This study represents the first validation of high latitude (>70°) TES ozone. We find that the calculated errors are consistent with the actual errors with a similar vertical distribution that varies between 5% and 20% for V003 and V004 TES data. In general, TES ozone profiles are positively biased (by less than 15%) from the surface to the upper-troposphere (~1000 to 100 hPa) and negatively biased (by less than 20%) from the upper-troposphere to the lower-stratosphere (100 to 30 hPa) when compared to the ozonesonde data. Lastly, for V003 and V004 TES data between 44° N and 71° N there is variability in the mean biases (from −14 to +15%), mean theoretical errors (from 6 to 13%), and mean random errors (from 9 to 19%)

    Efficient injection from large telescopes into single-mode fibres: Enabling the era of ultra-precision astronomy

    Photonic technologies offer numerous advantages for astronomical instruments such as spectrographs and interferometers owing to their small footprints and diverse range of functionalities. Operating at the diffraction-limit, it is notoriously difficult to efficiently couple such devices directly with large telescopes. We demonstrate that with careful control of both the non-ideal pupil geometry of a telescope and residual wavefront errors, efficient coupling with single-mode devices can indeed be realised. A fibre injection was built within the Subaru Coronagraphic Extreme Adaptive Optics (SCExAO) instrument. Light was coupled into a single-mode fibre operating in the near-IR (J-H bands) which was downstream of the extreme adaptive optics system and the pupil apodising optics. A coupling efficiency of 86% of the theoretical maximum limit was achieved at 1550 nm for a diffraction-limited beam in the laboratory, and was linearly correlated with Strehl ratio. The coupling efficiency was constant to within <30% in the range 1250-1600 nm. Preliminary on-sky data with a Strehl ratio of 60% in the H-band produced a coupling efficiency into a single-mode fibre of ~50%, consistent with expectations. The coupling was >40% for 84% of the time and >50% for 41% of the time. The laboratory results allow us to forecast that extreme adaptive optics levels of correction (Strehl ratio >90% in H-band) would allow coupling of >67% (of the order of coupling to multimode fibres currently). For Strehl ratios <20%, few-port photonic lanterns become a superior choice but the signal-to-noise must be considered. These results illustrate a clear path to efficient on-sky coupling into a single-mode fibre, which could be used to realise modal-noise-free radial velocity machines, very-long-baseline optical/near-IR interferometers and/or simply exploit photonic technologies in future instrument design.Comment: 15 pages, 16 figures, 1 table, published in A&

    Modeling Single Well Injection-Withdrawal (SWIW) Tests for Characterization of Complex Fracture-Matrix Systems

    The ability to reliably predict flow and transport in fractured porous rock is an essential condition for performance evaluation of geologic (underground) nuclear waste repositories. In this report, a suite of programs (TRIPOLY code) for calculating and analyzing flow and transport in two-dimensional fracture-matrix systems is used to model single-well injection-withdrawal (SWIW) tracer tests. The SWIW test, a tracer test using one well, is proposed as a useful means of collecting data for site characterization, as well as estimating parameters relevant to tracer diffusion and sorption. After some specific code adaptations, we numerically generated a complex fracture-matrix system for computation of steady-state flow and tracer advection and dispersion in the fracture network, along with solute exchange processes between the fractures and the porous matrix. We then conducted simulations for a hypothetical but workable SWIW test design and completed parameter sensitivity studies on three physical parameters of the rock matrix - namely porosity, diffusion coefficient, and retardation coefficient - in order to investigate their impact on the fracture-matrix solute exchange process. Hydraulic fracturing, or hydrofracking, is also modeled in this study, in two different ways: (1) by increasing the hydraulic aperture for flow in existing fractures and (2) by adding a new set of fractures to the field. The results of all these different tests are analyzed by studying the population of matrix blocks, the tracer spatial distribution, and the breakthrough curves (BTCs) obtained, while performing mass-balance checks and being careful to avoid some numerical mistakes that could occur. This study clearly demonstrates the importance of matrix effects in the solute transport process, with the sensitivity studies illustrating the increased importance of the matrix in providing a retardation mechanism for radionuclides as matrix porosity, diffusion coefficient, or retardation coefficient increase. Interestingly, model results before and after hydrofracking are insensitive to adding more fractures, while slightly more sensitive to aperture increase, making SWIW tests a possible means of discriminating between these two potential hydrofracking effects. Finally, we investigate the possibility of inferring relevant information regarding the fracture-matrix system physical parameters from the BTCs obtained during SWIW testing

    Polyploidy breaks speciation barriers in Australian burrowing frogs Neobatrachus

    Polyploidy has played an important role in evolution across the tree of life but it is still unclear how polyploid lineages may persist after their initial formation. While both common and well-studied in plants, polyploidy is rare in animals and generally less understood. The Australian burrowing frog genus Neobatrachus is comprised of six diploid and three polyploid species and offers a powerful animal polyploid model system. We generated exome-capture sequence data from 87 individuals representing all nine species of Neobatrachus to investigate species-level relationships, the origin and inheritance mode of polyploid species, and the population genomic effects of polyploidy on genus-wide demography. We describe rapid speciation of diploid Neobatrachus species and show that the three independently originated polyploid species have tetrasomic or mixed inheritance. We document higher genetic diversity in tetraploids, resulting from widespread gene flow between the tetraploids, asymmetric inter-ploidy gene flow directed from sympatric diploids to tetraploids, and isolation of diploid species from each other. We also constructed models of ecologically suitable areas for each species to investigate the impact of climate on differing ploidy levels. These models suggest substantial change in suitable areas compared to past climate, which correspond to population genomic estimates of demographic histories. We propose that Neobatrachus diploids may be suffering the early genomic impacts of climate-induced habitat loss, while tetraploids appear to be avoiding this fate, possibly due to widespread gene flow. Finally, we demonstrate that Neobatrachus is an attractive model to study the effects of ploidy on the evolution of adaptation in animals

Investigarla forma del control de estímulos que se desarrolla bajo condiciones de castigoes un tópico que necesita más atención por razones teóricas y prácticas.En preparaciones en las cuales el control de estímulos se puede desarrollar(e.g., programas múltiples), al menos dos estímulos pueden llegar a ejercerun control discriminativo sobre la supresión de la respuesta: un estímulo discriminativoantecedente (e.g., estímulos en programas múltiples) y la entregamisma del castigador.     Focus on form: A critical review

    ‘Focus on form’ (FonF) is a central construct in task-based language teaching. The term was first introduced by Michael Long to refer to an approach where learners’ attention is attracted to linguistic forms as they engage in the performance of tasks. It contrasts with a structure-based approach – ‘focus on forms’ (FonFs) – where specific linguistic forms are taught directly and explicitly. However, there is perhaps no construct in second language acquisition (SLA) that has proved so malleable and shifted in meaning so much. This review article begins by considering how Long’s original definition of it has stretched over time and then offers an updated definition of the construct based on the view that the term is best used to refer to specific kinds of ‘activities’ or ‘procedures’ rather than to an ‘approach’. A classification of different types of focus-on-form activities/procedures is then presented. There follows a discussion of focus on form from a psycholinguistic and discoursal perspective along with a review of research relevant to these perspectives. The article addresses a number of criticisms that have been levelled against focus on form, with special consideration paid to how focus on form can be utilized in instructional contexts where more traditional (i.e. FonFs) approaches have been the norm
