    Healthful choices depend on the latency and rate of information accumulation

    The drift diffusion model provides a parsimonious explanation of decisions across neurobiological, psychological and behavioural levels of analysis. Although most drift diffusion model implementations assume that only a single value guides decisions, choices often involve multiple attributes that could make separable contributions to choice. Here we fit incentive-compatible dietary choices to a multi-attribute, time-dependent drift diffusion model, in which taste and health could differentially influence the evidence accumulation process. We find that these attributes shaped both the relative value signal and the latency of evidence accumulation in a manner consistent with participants’ idiosyncratic preferences. Moreover, by using a dietary prime, we showed how a healthy choice intervention alters multi-attribute, time-dependent drift diffusion model parameters that in turn predict prime-dependent choices. Our results reveal that different decision attributes make separable contributions to the strength and timing of evidence accumulation, providing new insights into the construction of interventions to alter the processes of choice

    Peer presence increases the prosocial behavior of adolescents by speeding the evaluation of outcomes for others

    Peer presence can elicit maladaptive adolescent decision-making, potentially by increasing sensitivity to the rewards one receives. It remains unknown whether peer presence also increases adolescents’ sensitivity to others’ outcomes, which could have an adaptive effect in contexts allowing pro-social behaviors. Here, we combine social utility modeling and real-time decision process modeling to characterize how peer presence alters adolescents’ processing of self and other outcomes. We found that adolescents behaved selfishly when privately allocating monetary rewards for themselves and a peer in an incentive-compatible task. In peer presence, however, adolescents became more altruistic. Real-time decision process estimates collected using computer mouse tracking showed that altruistic behavior was associated with relatively earlier influence of peer-outcomes relative to self-outcomes, and that peer presence sped the influence of peer-outcomes without altering the time at which self-outcomes began to influence the decision process. Our results indicate a mechanism through which peer presence prompts greater prosocial behavior by altering how adolescents process prosocial outcomes

    Relationships with God among Young Adults: Validating a Measurement Model with Four Dimensions

    Experiencing a relationship with God is widely acknowledged as an important aspect of personal religiosity for both affiliated and unaffiliated young adults, but surprisingly few attempts have been made to develop measures appropriate to its latent, multidimensional quality. This paper presents a new model for measuring relationships with God based on religious role theory, attachment to God theory, and insights from interview-based studies, which allows for a wider array of dimensions than have been considered in prior work: anger, anxiety, intimacy, and consistency. To test our model's internal validity, we use confirmatory factor analysis with nationally representative data. To test its external validity, we (1) use difference-in-means tests across gender, race/ethnicity, geographical region, and religious affiliation; and (2) analyze correlations between our four new dimensions and four other commonly used measures of religiosity, thereby demonstrating both our model's validity and value for future studies of personal religiosity

    Enzyme-catalysed polymer cross-linking:Biocatalytic tools for chemical biology, materials science and beyond

    Intermolecular cross-linking is one of the most important techniques that can be used to fundamentally alter the material properties of a polymer. The introduction of covalent bonds between individual polymer chains creates 3D macromolecular assemblies with enhanced mechanical properties and greater chemical or thermal tolerances. In contrast to many chemical cross-linking reactions, which are the basis of thermoset plastics, enzyme catalysed processes offer a complimentary paradigm for the assembly of cross-linked polymer networks through their predictability and high levels of control. Additionally, enzyme catalysed reactions offer an inherently ‘greener’ and more biocompatible approach to covalent bond formation, which could include the use of aqueous solvents, ambient temperatures, and heavy metal-free reagents. Here, we review recent progress in the development of biocatalytic methods for polymer cross-linking, with a specific focus on the most promising candidate enzyme classes and their underlying catalytic mechanisms. We also provide exemplars of the use of enzyme catalysed cross-linking reactions in industrially relevant applications, noting the limitations of these approaches and outlining strategies to mitigate reported deficiencies

    Distraction in Verbal Short-Term Memory: Insights from Developmental Differences

    The contribution of two mechanisms of auditory distraction in verbal serial short-term memory—interference with the serial rehearsal processes used to support short-term recall and general attentional diversion—was investigated by exploiting differences in auditory distraction in children and adults. Experiment 1 showed that serial rehearsal plays a role in children’s as well as adults’ distractibility: Auditory distraction from irrelevant speech was greater for both children and adults as the burden on rehearsal increased. This pattern was particularly pronounced in children, suggesting that underdeveloped rehearsal skill in this population may increase their distractibility. Experiment 2 showed that both groups were more susceptible to changing- than steady-state speech when the task involved serial rehearsal—indicating that both groups suffer interference-by-process—but that children, but not adults, were also susceptible to any sort of sound (steady or changing) in a task thought to be devoid of serial rehearsal. The overall pattern of results suggests that children’s increased susceptibility to auditory distraction during verbal short-term memory performance is due to a greater susceptibility to attentional diversion; in this view, under-developed rehearsal-skill increases children’s distractibility by exacerbating their under-developed attentional control rather than by increasing interference-by-process

    Body Mass Index and Mortality in Coronavirus Disease 2019 and Other Diseases:A Cohort Study in 35,506 ICU Patients

    OBJECTIVES: Obesity is a risk factor for severe coronavirus disease 2019 and might play a role in its pathophysiology. It is unknown whether body mass index is related to clinical outcome following ICU admission, as observed in various other categories of critically ill patients. We investigated the relationship between body mass index and inhospital mortality in critically ill coronavirus disease 2019 patients and in cohorts of ICU patients with non-severe acute respiratory syndrome coronavirus 2 viral pneumonia, bacterial pneumonia, and multiple trauma. DESIGN: Multicenter observational cohort study. SETTING: Eighty-two Dutch ICUs participating in the Dutch National Intensive Care Evaluation quality registry. PATIENTS: Thirty-five-thousand five-hundred six critically ill patients. INTERVENTIONS: None. MEASUREMENTS AND MAIN RESULTS: Patient characteristics and clinical outcomes were compared between four cohorts (coronavirus disease 2019, nonsevere acute respiratory syndrome coronavirus 2 viral pneumonia, bacterial pneumonia, and multiple trauma patients) and between body mass index categories within cohorts. Adjusted analyses of the relationship between body mass index and inhospital mortality within each cohort were performed using multivariable logistic regression. Coronavirus disease 2019 patients were more likely male, had a higher body mass index, lower Pao2/Fio2 ratio, and were more likely mechanically ventilated during the first 24 hours in the ICU compared with the other cohorts. Coronavirus disease 2019 patients had longer ICU and hospital length of stay, and higher inhospital mortality. Odds ratios for inhospital mortality for patients with body mass index greater than or equal to 35 kg/m2 compared with normal weight in the coronavirus disease 2019, nonsevere acute respiratory syndrome coronavirus 2 viral pneumonia, bacterial pneumonia, and trauma cohorts were 1.15 (0.79- 1.67), 0.64 (0.43-0.95), 0.73 (0.61-0.87), and 0.81 (0.57-1.15), respectively. CONCLUSIONS: The obesity paradox, which is the inverse association between body mass index and mortality in critically ill patients, is not present in ICU patients with coronavirus disease 2019-related respiratory failure, in contrast to nonsevere acute respiratory syndrome coronavirus 2 viral and bacterial respiratory infections

    Indulgent foods can paradoxically promote disciplined dietary choices

    As obesity rates continue to rise, interventions promoting healthful choices will become increasingly important. Here, participants (N = 79) made binary choices between familiar foods; some trials contained a common consequence that had a constant probability of receipt regardless of the participant's choice. We theorized-on the basis of simulations using a value-normalization model-that indulgent common consequences potentiated disciplined choices by shaping other options' perceived healthfulness and tastiness. Our experimental results confirmed these predictions: An indulgent common consequence more than doubled the rate of disciplined choices. We used eye-gaze data to provide insights into the underlying mechanisms, finding that an indulgent common consequence biased eye gaze toward healthful foods. Furthermore, attention toward the common consequence predicted individual differences in behavioral bias. Results were replicated across two independent samples receiving distinct goal primes. These results demonstrate that introducing an irrelevant indulgent food can alter processing of healthier items-and thus promote disciplined choices

    Biphasic Elimination of Tenofovir Diphosphate and Nonlinear Pharmacokinetics of Zidovudine Triphosphate in a Microdosing Study

    Objective: Phase 0 studies can provide initial pharmacokinetics (PKs) data in humans and help to facilitate early drug development, but their predictive value for standard dosing is controversial. To evaluate the prediction of microdosing for active intracellular drug metabolites, we compared the PK profile of 2 antiretroviral drugs, zidovudine (ZDV) and tenofovir (TFV), in microdose and standard dosing regimens. Study Design: We administered a microdose (100 μg) of [superscript 14]C-labeled drug (ZDV or tenofovir disoproxil fumarate) with or without a standard unlabelled dose (300 mg) to healthy volunteers. Both the parent drug in plasma and the active metabolite, ZDV-triphosphate (ZDV-TP) or TFV-diphosphate (TFV-DP) in peripheral blood mononuclear cells (PBMCs) and CD4[superscript +] cells were measured by accelerator mass spectrometry. Results: The intracellular ZDV-TP concentration increased less than proportionally over the dose range studied (100 μg–300 mg), whereas the intracellular TFV-DP PKs were linear over the same dose range. ZDV-TP concentrations were lower in CD4[superscript +] cells versus total PBMCs, whereas TFV-DP concentrations were not different in CD4[superscript +] cells and PBMCs. Conclusions: Our data were consistent with a rate-limiting step in the intracellular phosphorylation of ZDV but not TFV. Accelerator mass spectrometry shows promise for predicting the PK of active intracellular metabolites of nucleosides, but nonlinearity of PK may be seen with some drugs.Johns Hopkins University (Institute for Clinical and Translational Research CTSA Grant UL1-RR025005

    A comparison of transgenic rodent mutation and in vivo comet assay responses for 91 chemicals.

    A database of 91 chemicals with published data from both transgenic rodent mutation (TGR) and rodent comet assays has been compiled. The objective was to compare the sensitivity of the two assays for detecting genotoxicity. Critical aspects of study design and results were tabulated for each dataset. There were fewer datasets from rats than mice, particularly for the TGR assay, and therefore, results from both species were combined for further analysis. TGR and comet responses were compared in liver and bone marrow (the most commonly studied tissues), and in stomach and colon evaluated either separately or in combination with other GI tract segments. Overall positive, negative, or equivocal test results were assessed for each chemical across the tissues examined in the TGR and comet assays using two approaches: 1) overall calls based on weight of evidence (WoE) and expert judgement, and 2) curation of the data based on a priori acceptability criteria prior to deriving final tissue specific calls. Since the database contains a high prevalence of positive results, overall agreement between the assays was determined using statistics adjusted for prevalence (using AC1 and PABAK). These coefficients showed fair or moderate to good agreement for liver and the GI tract (predominantly stomach and colon data) using WoE, reduced agreement for stomach and colon evaluated separately using data curation, and poor or no agreement for bone marrow using both the WoE and data curation approaches. Confidence in these results is higher for liver than for the other tissues, for which there were less data. Our analysis finds that comet and TGR generally identify the same compounds (mainly potent mutagens) as genotoxic in liver, stomach and colon, but not in bone marrow. However, the current database content precluded drawing assay concordance conclusions for weak mutagens and non-DNA reactive chemicals

    Care for children with atopic dermatitis in the Netherlands during the COVID-19 pandemic:Lessons from the first wave and implications for the future

    The first wave of the coronavirus disease 2019 (COVID-19) pandemic had an enormous impact on health-care services, including on care provision for children with atopic dermatitis (AD). We investigated the impact of COVID-19 on the care for children with moderate to severe AD at our tertiary outpatient clinic and examined satisfaction with care. We reviewed outpatient records, comparing total number and types of consultations during the first COVID-19 wave (March until July 2020) with the corresponding months of 2019 and 2018. In addition, we conducted a questionnaire-based study investigating the impact of COVID-19 on clinical and psychological symptoms, and satisfaction with care. A total number of 913 consultations (466 individual children) were conducted during the first COVID-19 wave in 2020, while 698 (391 individual children) and 591 consultations (356 individual children) were conducted in 2019 and 2018. The proportion of remote consultations was higher (56.2%) compared to 14.0% in 2019 and 12.7% in 2018. Worsening of AD was reported by 9.7% of caretakers. Overall satisfaction with provided care was high (8.6; interquartile range [IQR] = 7.3–10.0). Caretakers receiving face-to-face consultation were significantly (p = 0.026) more satisfied (9.0; IQR = 8.0–10.0) than caretakers receiving remote consultation (7.9; IQR = 7.0–9.5). The COVID-19 pandemic had an unprecedented impact on care provision for children with AD, particularly on the number of remote consultations. Overall satisfaction with care was high. The impact of COVID-19 on disease severity remained limited. Remote consultations seem to be a useful tool that can be put into practice during the COVID-19 pandemic
