    Higher order Laguerre-Gauss mode degeneracy in realistic, high finesse cavities

    Higher order Laguerre-Gauss (LG) beams have been proposed for use in future gravitational wave detectors, such as upgrades to the Advanced LIGO detectors and the Einstein Telescope, for their potential to reduce the effects of the thermal noise of the test masses. This paper details the theoretical analysis and simulation work carried out to investigate the behaviour of LG beams in realistic optical setups, in particular the coupling between different LG modes in a linear cavity. We present a new analytical approximation to compute the coupling between modes, using Zernike polynomials to describe mirror surface distortions. We apply this method in a study of the behaviour of the LG33 mode within realistic arm cavities, using measured mirror surface maps from the Advanced LIGO project. We show mode distortions that can be expected to arise due to the degeneracy of higher order spatial modes within such cavities and relate this to the theoretical analysis. Finally we identify the mirror distortions which cause significant coupling from the LG33 mode into other order 9 modes and derive requirements for the mirror surfaces.Comment: 12 pages Submitted to PRD 19/07/201

    Genome size and ploidy of Paracoccidioides brasiliensis reveals a haploid DNA content: Flow cytometry and GP43 sequence analysis

    The aim of this study was to evaluate genome size and ploidy of the dimorphic pathogenic fungus Paracoccidioides brasiliensis. The cell cycle analysis of 10 P. brasiliensis isolates by flow cytometry (FCM) revealed a genome size ranging from 26.3+/-0.1Mb (26.9+/-0.1fg) to 35.5+/-0.2Mb (36.3+/-0.2fg) per uninucleated yeast cell. The DNA content of conidia from P. brasiliensis ATCC 60855-30.2+/-0.8Mb (30.9+/-0.8fg) -showed no significant differences with the yeast form, possibly excluding the occurrence of ploidy shift during morphogenesis. The ploidy of several P. brasiliensis isolates was assessed by comparing genome sizing by FCM with the previously described average haploid size obtained from electrophoretic karyotyping. The analysis of intra-individual variability of a highly polymorphic P. brasiliensis gene, GP43, indicated that only one allele seems to be present. Overall, the results showed that all analysed isolates presented a haploid, or at least aneuploid, DNA content and no association was detected between genome size/ploidy and the clinical-epidemiological features of the studied isolates. This work provides new knowledge on P. brasiliensis genetics/genomics, important for future research in basic cellular/molecular mechanisms and for the development/design of molecular techniques in this fungus

    Incoherent dynamics in neutron-matter interaction

    Coherent and incoherent neutron-matter interaction is studied inside a recently introduced approach to subdynamics of a macrosystem. The equation describing the interaction is of the Lindblad type and using the Fermi pseudopotential we show that the commutator term is an optical potential leading to well-known relations in neutron optics. The other terms, usually ignored in optical descriptions and linked to the dynamic structure function of the medium, give an incoherent contribution to the dynamics, which keeps diffuse scattering and attenuation of the coherent beam into account, thus warranting fulfilment of the optical theorem. The relevance of this analysis to experiments in neutron interferometry is briefly discussed.Comment: 15 pages, revtex, no figures, to appear in Phys. Rev.

    Bioresorbable ureteral stents from natural origin polymers

    In this work, stents were produced from natural originpolysaccharides. Alginate, gellan gum, and a blend ofthese with gelatin were used to produce hollow tube (stents)following a combination of templated gelation and criticalpoint carbon dioxide drying. Morphological analysis of thesurface of the stents was carried out by scanning electronmicroscopy. Indwelling time, encrustation, and stability ofthe stents in artificial urine solution was carried out up to 60days of immersion. In vitro studies carried out with simulatedurine demonstrated that the tubes present a high fluid uptakeability, about 1000%. Despite this, the materials are able tomaintain their shape and do not present an extensive swellingbehavior. The bioresorption profile was observed to behighly dependent on the composition of the stent and it canbe tuned. Complete dissolution of the materials may occurbetween 14 and 60 days. Additionally, no encrustation wasobserved within the tested timeframe. The ability to resistbacterial adherence was evaluated with Gram-positive Staphylococcus aureus and two Gram-negatives Escherichia coliDH5 alpha and Klebsiella oxytoca. For K. oxytoca, no differenceswere observed in comparison with a commercialstent (BiosoftVR duo, Porges), although, for S. aureus alltested compositions had a higher inhibition of bacterialadhesion compared to the commercial stents. In case ofE. coli, the addition of gelatin to the formulations reducedthe bacterial adhesion in a highly significant manner comparedto the commercial stents.     Functionality of the paracoccidioides mating α-pheromone-receptor system

    Recent evidence suggests that Paracoccidioides species have the potential to undergo sexual reproduction, although no sexual cycle has been identified either in nature or under laboratory conditions. In the present work we detected low expression levels of the heterothallic MAT loci genes MAT1-1 and MAT1-2, the a-pheromone (PBa) gene, and the a- and apheromone receptor (PREB and PREA) genes in yeast and mycelia forms of several Paracoccidioides isolates. None of the genes were expressed in a mating type dependent manner. Stimulation of P. brasiliensis MAT1-2 strains with the synthetic a pheromone peptide failed to elicit transcriptional activation of MAT1-2, PREB or STE12, suggesting that the strains tested are insensitive to a-pheromone. In order to further evaluate the biological functionality of the pair a-pheromone and its receptor, we took advantage of the heterologous expression of these Paracoccidioides genes in the corresponding S. cerevisiae null mutants. We show that S. cerevisiae strains heterologously expressing PREB respond to Pba pheromone either isolated from Paracoccidioides culture supernatants or in its synthetic form, both by shmoo formation and by growth and cell cycle arrests. This allowed us to conclude that Paracoccidioides species secrete an active a-pheromone into the culture medium that is able to activate its cognate receptor. Moreover, expression of PREB or PBa in the corresponding null mutants of S. cerevisiae restored mating in these non-fertile strains. Taken together, our data demonstrate pheromone signaling activation by the Paracoccidioides a-pheromone through its receptor in this yeast model, which provides novel evidence for the existence of a functional mating signaling system in Paracoccidioides.MHJS and JFM were supported by Fundacão para a Ciência e Tecnologia (FCT) grants.     Extension of Yeast Chronological Lifespan by Methylamine

    Background: Chronological aging of yeast cells is commonly used as a model for aging of human post-mitotic cells. The yeast Saccharomyces cerevisiae grown on glucose in the presence of ammonium sulphate is mainly used in yeast aging research. We have analyzed chronological aging of the yeast Hansenula polymorpha grown at conditions that require primary peroxisome metabolism for growth. Methodology/Principal Findings: The chronological lifespan of H. polymorpha is strongly enhanced when cells are grown on methanol or ethanol, metabolized by peroxisome enzymes, relative to growth on glucose that does not require peroxisomes. The short lifespan of H. polymorpha on glucose is mainly due to medium acidification, whereas most likely ROS do not play an important role. Growth of cells on methanol/methylamine instead of methanol/ammonium sulphate resulted in further lifespan enhancement. This was unrelated to medium acidification. We show that oxidation of methylamine by peroxisomal amine oxidase at carbon starvation conditions is responsible for lifespan extension. The methylamine oxidation product formaldehyde is further oxidized resulting in NADH generation, which contributes to increased ATP generation and reduction of ROS levels in the stationary phase. Conclusion/Significance: We conclude that primary peroxisome metabolism enhanced chronological lifespan of H. polymorpha. Moreover, the possibility to generate NADH at carbon starvation conditions by an organic nitrogen source supports further extension of the lifespan of the cell. Consequently, the interpretation of CLS analyses in yeast should include possible effects on the energy status of the cell.

    Modeling a mobile group recommender system for tourism with intelligent agents and gamification

    To provide recommendations to groups of people is a complex task, especially due to the group’s heterogeneity and conflicting preferences and personalities. This heterogeneity is even deeper in occasional groups formed for predefined tour packages in tourism. Group Recommender Systems (GRS) are being designed for helping in situations like those. However, many limitations can still be found, either on their time-consuming configurations and excessive intrusiveness to build the tourists’ profile, or in their lack of concern for the tourists’ interests during the planning and tours, like feeling a greater liberty, diminish the sense of fear/being lost, increase their sense of companionship, and promote the social interaction among them without losing a personalized experience. In this paper, we propose a conceptual model that intends to enhance GRS for tourism by using gamification techniques, intelligent agents modeled with the tourists’ context and profile, such as psychological and socio-cultural aspects, and dialogue games between the agents for the post-recommendation process.
