    Statistical Analysis of Data on Linker Histones/DNA Interactions

    2000 Mathematics Subject Classification: 62P10, 92C40Linker histones (H1, H1o H5, subtypes and variants) play a pivotal role in formation of higher order chromatin structure and thus - as main regulators of the expression of genetic information kept in DNA. That is why the knowledge of the nature of linker histones/DNA interactions is of a greatest interest in understanding of such important issues as transcription regulation, cell division, and cancerogenesis. As DNA is a main "target" of most anticancer antibiotics, the analysis of competitive reactions between that drugs (in our case actinomycin D and netropsin) and linker histones for binding to certain sites in DNA gives hopeful information concerning the mode of such interactions. In this work we present statistical analysis of some experimental data concerning the influence of some anticancer antibiotics on linker histones/DNA interactions. First, it was investigated the formulated hypothesis of the dependence of H1/DNA interaction on actinomycin D concentration. Such a relation was expected knowing the different mode for binding of the both drugs to DNA double helix. The applied statistical analysis using chi-square test for independence showed that the concentration of Actinomycin D in reaction mixture had no essential effect on linker histone/DNA binding. On the contrary, the same analysis with the second antibiotic - netropsin showed that we could not reject the hypothesis of dependence.     Materialising architecture for social care: brick walls and compromises in design for later life.

    This article reports on an ethnography of architectural projects for later life social care in the UK. Informed by recent debates in material studies and ‘materialities of care’ we offer an analysis of a care home project that is sensitive to architectural materials that are not normally associated with care and wellbeing. Although the care home design project we focus on in this article was never built, we found that design discussions relating to both a curved brick wall and bricks more generally were significant to its architectural ‘making’. The curved wall and the bricks were used by the architects to encode quality and values of care into their design. This was explicit in the design narrative that was core to a successful tender submitted by a consortium comprising architects, developers, contractors, and a care provider to a local authority who commissioned the care home. However, as the project developed, initial consensus for the design features fractured. Using a materialised analysis, we document the tussles generated by the curved wall and the bricks and argue that mundane building materials can be important to, and yet marginalised within, the relations inherent within an ‘architectural care assemblage.’ During the design process we saw how decisions about materials are contentious and they act as a catalyst of negotiations that compromise ‘materialities of care.

    Star Architecture as Socio-Material Assemblage

    Taking inspiration from new materialism and assemblage, the chapter deals with star architects and iconic buildings as socio-material network effects that do not pre-exist action, but are enacted in practice, in the materiality of design crafting and city building. Star architects are here conceptualized as part of broader assemblages of actors and practices ‘making star architecture’ a reality, and the buildings they design are considered not just as unique and iconic objects, but dis-articulated as complex crafts mobilizing skills, technologies, materials, and forms of knowledge not necessarily ascribable to architecture. Overcoming narrow criticism focusing on the symbolic order of icons as unique creations and alienated repetitions of capitalist development, the chapter’s main aim is to widen the scope of critique by bridging culture and economy, symbolism and practicality, making star architecture available to a broad, fragmented arena of (potential) critics, unevenly equipped with critical tools and differentiated experiences

    On the Usefulness of Modern Animism: Co-Creating Architecture with Soils as Ontopolitical Practice

    In opposition to the general understanding of animism as an irrational religious set of beliefs, the—secular—modern animism embodied in this practice of built and grown architecture is operative. It conceives of places—ecosystems—as beings with agency that we garden with to nurture and express their resilience. It is a useful ontology for ecological practice; this architectural animism is ontopolitical; it co-creates a common world. At a recent symposium, a number of projects were presented as architectural soils and to give a voice to these earthly beings, the method was to write a letter from them to me. This first letter with the response in this article forms the beginning of an animistic correspondence. The medium places the reader in a pluriverse, in between the multiple voices of various “actants.” The text is isomorphic to the embodied dialogue of this earthy practice. It is useful both to nurture societal awareness—empathy and care—towards these fragmentary ecosystemic beings, and as research method to conceive them, and our relationship

    Anti-Bacterial Effects of Poly-N-Acetyl-Glucosamine Nanofibers in Cutaneous Wound Healing: Requirement for Akt1

    Treatment of cutaneous wounds with poly-N-acetyl-glucosamine nanofibers (sNAG) results in increased kinetics of wound closure in diabetic animal models, which is due in part to increased expression of several cytokines, growth factors, and innate immune activation. Defensins are also important for wound healing and anti-microbial activities. Therefore, we tested whether sNAG nanofibers induce defensin expression resulting in bacterial clearance.The role of sNAG in defensin expression was examined using immunofluoresence microscopy, pharmacological inhibition, and shRNA knockdown in vitro. The ability of sNAG treatment to induce defensin expression and bacterial clearance in WT and AKT1-/- mice was carried out using immunofluoresent microscopy and tissue gram staining. Neutralization, using an antibody directed against β-defensin 3, was utilized to determine if the antimicrobial properties of sNAG are dependent on the induction of defensin expression.sNAG treatment causes increased expression of both α- and β-type defensins in endothelial cells and β-type defensins in keratinocytes. Pharmacological inhibition and shRNA knockdown implicates Akt1 in sNAG-dependent defensin expression in vitro, an activity also shown in an in vivo wound healing model. Importantly, sNAG treatment results in increased kinetics of wound closure in wild type animals. sNAG treatment decreases bacterial infection of cutaneous wounds infected with Staphylococcus aureus in wild type control animals but not in similarly treated Akt1 null animals. Furthermore, sNAG treatment of S. aureus infected wounds show an increased expression of β-defensin 3 which is required for sNAG-dependent bacterial clearance. Our findings suggest that Akt1 is involved in the regulation of defensin expression and the innate immune response important for bacterial clearance. Moreover, these findings support the use of sNAG nanofibers as a novel method for enhancing wound closure while simultaneously decreasing wound infection

    Networks, interfaces and computer-generated images: Learning from digital visualisations of urban redevelopment projects

    Over the past five years, computer-generated images (CGIs) have become commonplace as a means to market urban redevelopments. To date, however, they have been given relatively little attention as a new form of visualising the urban. In this paper we argue that these CGIs deserve more attention, and attention of a particular kind. We argue that, instead of approaching them as images situated in urban space, their digitality invites us to understand them as interfaces circulating through a software-supported network space. We use an actor-network theory understanding of ‘network’ and argue that the action done on and with CGIs as they are created takes place at a series of interfaces. These interfaces—between and among humans, software, and hardware—are where work is done both to create the CGI and to create the conditions for their circulation. These claims are explored in relation to the CGIs made for a large urban redevelopment project in Doha, Qatar. We conclude by suggesting that geographers need to reconsider their understanding of digital images and be as attentive to the interfaces embedded in the image as to the CGI’s visual content.The project was funded by a grant from the Economic and Social Research Council RES-062-23-3305

    Broken seniority symmetry in the semimagic proton mid-shell nucleus <sup>95</sup>Rh

    Lifetime measurements of low-lying excited states in the semimagic ( N = 50 ) nucleus 95Rh have been performed by means of the fast-timing technique. The experiment was carried out using γ -ray detector arrays consisting of LaBr3(Ce) scintillators and germanium detectors integrated into the DESPEC experimental setup commissioned for the Facility for Antiproton and Ion Research (FAIR) Phase-0, Darmstadt, Germany. The excited states in 95Rh were populated primarily via the β decays of 95Pd nuclei, produced in the projectile fragmentation of a 850 MeV/nucleon 124Xe beam impinging on a 4 g / cm2 9Be target. The deduced electromagnetic E2 transition strengths for the γ -ray cascade within the multiplet structure depopulating from the isomeric Iπ = 21 / 2+ state are found to exhibit strong deviations from predictions of standard shell model calculations which feature approximately conserved seniority symmetry. In particular, the observation of a strongly suppressed E2 strength for the 13 / 2+ → 9 / 2+ ground state transition cannot be explained by calculations employing standard interactions. This remarkable result may require revision of the nucleon-nucleon interactions employed in state-of-the-art theoretical model calculations, and might also point to the need for including three-body forces in the Hamiltonian
