    Three-year outcomes of a once daily fractionation scheme for accelerated partial breast irradiation (APBI) using 3-D conformal radiotherapy (3D-CRT).

    The aim of this study was to report 3-year outcomes of toxicity, cosmesis, and local control using a once daily fractionation scheme (49.95 Gy in 3.33 Gy once daily fractions) for accelerated partial breast irradiation (APBI) using three-dimensional conformal radiotherapy (3D-CRT). Between July 2008 and August 2010, women aged ≥40 years with ductal carcinoma in situ or node-negative invasive breast cancer ≤3 cm in diameter, treated with breast-conserving surgery achieving negative margins, were accrued to a prospective study. Women were treated with APBI using 3-5 photon beams, delivering 49.95 Gy over 15 once daily fractions over 3 weeks. Patients were assessed for toxicities, cosmesis, and local control rates before APBI and at specified time points. Thirty-four patients (mean age 60 years) with Tis 0 (n = 9) and T1N0 (n = 25) breast cancer were treated and followed up for an average of 39 months. Only 3% (1/34) patients experienced a grade 3 subcutaneous fibrosis and breast edema and 97% of the patients had good/excellent cosmetic outcome at 3 years. The 3-year rate of ipsilateral breast tumor recurrence (IBTR) was 0% while the rate of contralateral breast events was 6%. The 3-year disease-free survival (DFS), overall survival (OS), and breast cancer-specific survival (BCSS) was 94%, 100%, and 100%, respectively. Our novel accelerated partial breast fractionation scheme of 15 once daily fractions of 3.33 Gy (49.95 Gy total) is a remarkably well-tolerated regimen of 3D-CRT-based APBI. A larger cohort of patients is needed to further ascertain the toxicity of this accelerated partial breast regimen

    Ecdysone receptor directly binds the promoter of the Drosophila caspase dronc, regulating its expression in specific tissues

    The steroid hormone ecdysone regulates moulting, cell death, and differentiation during insect development. Ecdysone mediates its biological effects by either direct activation of gene transcription after binding to its receptor EcR–Usp or via hierarchical transcriptional regulation of several primary transcription factors. In turn, these transcription factors regulate the expression of several downstream genes responsible for specific biological outcomes. DRONC, the Drosophila initiator caspase, is transcriptionally regulated by ecdysone during development. We demonstrate here that the dronc promoter directly binds EcR–Usp. We further show that mutation of the EcR–Usp binding element (EcRBE) reduces transcription of a reporter and abolishes transactivation by an EcR isoform. We demonstrate that EcRBE is required for temporal regulation of dronc expression in response to ecdysone in specific tissues. We also uncover the participation of a putative repressor whose function appears to be coupled with EcR–Usp. These results indicate that direct binding of EcR–Usp is crucial for controlling the timing of dronc expression in specific tissues

    Onset of Propagation of Planar Cracks in Heterogeneous Media

    The dynamics of planar crack fronts in hetergeneous media near the critical load for onset of crack motion are investigated both analytically and by numerical simulations. Elasticity of the solid leads to long range stress transfer along the crack front which is non-monotonic in time due to the elastic waves in the medium. In the quasistatic limit with instantaneous stress transfer, the crack front exhibits dynamic critical phenomenon, with a second order like transition from a pinned to a moving phase as the applied load is increased through a critical value. At criticality, the crack-front is self-affine, with a roughness exponent ζ=0.34±0.02\zeta =0.34\pm 0.02. The dynamic exponent zz is found to be equal to 0.74±0.03 0.74\pm 0.03 and the correlation length exponent ν=1.52±0.02\nu =1.52\pm 0.02. These results are in good agreement with those obtained from an epsilon expansion. Sound-travel time delays in the stress transfer do not change the static exponents but the dynamic exponent zz becomes exactly one. Real elastic waves, however, lead to overshoots in the stresses above their eventual static value when one part of the crack front moves forward. Simplified models of these stress overshoots are used to show that overshoots are relevant at the depinning transition leading to a decrease in the critical load and an apparent jump in the velocity of the crack front directly to a non-zero value. In finite systems, the velocity also shows hysteretic behaviour as a function of the loading. These results suggest a first order like transition.     Multiple sulphur and iron isotope composition of detrital pyrite in Archaean sedimentary rocks : a new tool for provenance analysis

The definitive version was published in Earth and Planetary Science Letters 286 (2009): 436-445, doi:10.1016/j.epsl.2009.07.008.Multiple S (δ34S and δ33S) and Fe (δ56Fe) isotope analyses of rounded pyrite grains from 3.1 to 2.6 Ga conglomerates of southern Africa indicate their detrital origin, which supports anoxic surface conditions in the Archaean. Rounded pyrites from Meso- to Neoarchaean gold and uranium-bearing strata of South Africa are derived from both crustal and sedimentary sources, the latter being characterised by non-mass dependent fractionation of S isotopes (Δ33S as negative as -1.35‰) and large range of Fe isotope values (δ56Fe between -1.1 and 1.2‰). Most sediment-sourced pyrite grains are likely derived from sulphide nodules in marine organic matter-rich shales, sedimentary exhalites and volcanogenic massive sulphide deposits. Some sedimentary pyrite grains may have been derived from in situ sulphidised Fe-oxides, prior to their incorporation into the conglomerates, as indicated by unusually high positive δ56Fe values. Sedimentary sulphides without significant non-mass dependent fractionation of S isotopes were also present in the source of some conglomerates. The abundance in these rocks of detrital pyrite unstable in the oxygenated atmosphere may suggest factors other than high pO2 as the cause for the absence of significant non-mass dependent fractionation processes in the 3.2 – 2.7 Ga atmosphere. Rounded pyrites from the ca. 2.6 Ga conglomerates of the Belingwe greenstone belt in Zimbabwe have strongly fractionated δ34S, Δ33S and δ56Fe values, the source of which can be traced back to black shale-hosted massive sulphides in the underlying strata.     Dynamics and Instabilities of Planar Tensile Cracks in Heterogeneous Media

    The dynamics of tensile crack fronts restricted to advance in a plane are studied. In an ideal linear elastic medium, a propagating mode along the crack front with a velocity slightly less than the Rayleigh wave velocity, is found to exist. But the dependence of the effective fracture toughness Γ(v)\Gamma(v) on the crack velocity is shown to destabilize the crack front if (dΓ)/(dv)<0(d\Gamma)/(dv)<0. Short wavelength radiation due to weak random heterogeneities leads to this instability at low velocities. The implications of these results for the crack dynamics are discussed.Comment: 12 page

    Killing the golden goose: balancing preservation and development in Wilmington, North Carolina

    There is a delicate balance between preserving objects of the past and preparing for the future. While preservationists fight a noble battle in protecting artifacts and architecture which may possess historic significance, their endeavors impede the efforts of developers in reshaping the landscape. The story of downtown Wilmington’s revitalization process is a story of a city’s changing needs, and how one organization in particular, Downtown Area Revitalization Effort, Inc., worked to remedy the city’s ills. Established in response to urban sprawl and the impending construction of an indoor shopping mall, DARE initially used preservation as a tool for economic development, but with the passage of time and changes in administration, rehabilitation was replaced by marketing and development policies. However, such occurrences were not isolated to Wilmington. The pressure to cast aside preservation in favor of renewed expansion registered in downtown areas across the country. Though organizations in cities like Charleston and Savannah were successful in preserving their historic fabric through the practice of urban husbandry, other cities such as Chattanooga and Mobile eventually succumbed to increased development through long-term project planning. By examining the economic history of downtown Wilmington in relation to urban sprawl, this thesis argues that changes in the city’s political climate brought about a shift in DARE’s policies, as the organization moved from practical preservation to industrial recruiting and development

    An unusual and rare case of burn: challenge to cause and manner of death

    In the brought dead cases where the dead body is completely charred pose difficulty to autopsy surgeon in commenting cause & manner of death. Many a times, scene of incidence, the inquest report, history narrated by relatives and postmortem findings may not go hand-in-hand. Suicides by burning especially by females are very common in India, but charred (burn) body without conflagration in which manner is suicidal is rarely occurred and reported. “Char means to burn to charcoal”. Investigation in this type of cases by police officer is again a matter of his knowledge and experience. Here we are reporting such case; a well built, 26 years old married but separated, mentally stressed female living with her mother and brother since 3 years, was found in bathroom in a charred condition on 03/01/2012 around 06:30 hours. As per inquest, manner of death is suicidal. The autopsy findings did not reveal exact cause of death but raised the suspicion of homicide. So the present case report will make everyone to think over not only importance of cause and manner of death in case of charred body but also to reinforce the investigating authority for thorough investigation in favor of justice to the victim
