    Sequencing studies for the identification and characterization of new and old Rubus viruses

    In Europe, raspberry plants are commonly infected with a complex of aphid-transmitted viruses that together cause raspberry mosaic disease (RMD). During the previous 30 years, by grafting and vector transmission to a range of red and black raspberry cultivars, these viruses have been loosely characterized and identified as Raspberry leaf spot virus (RLSV), Raspberry leaf mottle Virus (RLMV), Black raspberry necrosis virus (BRNV) and Rubus yellow net virus. An additional, very widespread virus, Raspberry vein chlorosis virus (RVCV), is spread by a different aphid vector. Recently some sequence data have been obtained for RYNV, BRNV and Raspberry mottle virus (RMoV), a virus found in plants showing RMD symptoms. We have carried out sequencing studies using random amplification and mass analysis approaches and will present information on the relationship between RMoV, RLSV and RLMV, as well as the first data for RVCV and a novel, possibly segmented minus-strand RNA virus infecting raspberry.     Impact of Facility Location on the Financial Performance of Integrated and Distributed LVL Production in Subtropical Eastern Australia

    In subtropical eastern Australia, the declining availability of traditional, large hardwood native forest logs has motivated hardwood sawmills to explore potentially utilising small logs in the manufacture of veneer-based engineered wood products (EWPs), such as laminated veneer lumber (LVL). An aspatial mathematical model that maximises net present value (NPV) over a 30-year project life has been applied to estimate the financial performance of LVL manufacture in this region. Of particular interest was how facility location affected financial performance, and whether distributed production of veneer (close to the log resource) and LVL (distant from the log resource) may be more profitable than integrated production under some circumstances. While integrated production of veneer and LVL near the resource maximised NPV, distributed production was found to be more profitable than integrated production in situations where the LVL manufacturing facility had to be located relatively far from the resource. Nevertheless, the level of value-adding and processing scale had a greater impact on financial performance than facility location. The analysis also highlighted that log procurement strategy substantially affected financial performance. Encouragingly for forest growers and wood processors, utilising large volumes of small diameter logs, was important for maximisation of NPV of larger-scale LVL facilities

    Towards reducing the capital cost of manufacturing Laminated Veneer Lumbers: Investigating finger jointing solutions

    The capital cost of setting up a Laminated Veneer Lumber (LVL) plant which produces continuous LVL billet products, through a continuous veneer assembly and hot-pressing processes, is significant. However, the utilisation of batch-type presses, similar to those employed in the plywood industry, could significantly reduce this initial cost and may provide new opportunities for small to medium scale operations. This process would produce shorter billet lengths which would need to be joined together to produce lengths viable for structural products. Scarf joints have been used commercially to join some veneer-based engineered wood products but have limitations, while finger joints are a common method for jointing sawn timber products and offer some key advantages but is not a common method to join veneer-based products. Consequently, this paper focusses on investigating the influence of key manufacturing parameters on the performance of finger jointed LVL. The effect of the joint orientation (horizontal or vertical), the finger length, the gluing pressure and the adhesive type on the joint strength and stiffness were investigated. The finger jointed LVL were tested in edge bending, flat bending and tension, and the results were compared to reference unjointed LVL. The bending performance of the finger jointed LVL was also compared to scarfed jointed LVL. In total 304 tests were performed. The results indicated that the average strength values of finger jointed LVL can reach up to 99% of the average strength of unjointed LVL and compares to scarf jointed LVL on flat bending. Horizontal joints, being more practical to produce for deep beams, performed similarly to vertical joints. The 25 mm joints were found to have no mechanical advantages over the 20 mm investigated finger joints. A gluing pressure lower than the Eurocode's recommended level for solid timber achieved sufficient bonding for the products to be utilised. The gluing pressure was also found not to influence the performance of the joint, for the range of pressures investigated. Both polyurethane and resorcinol-formaldehyde adhesives produced high performing products, with the latter displaying superior adhesive bond durability. The paper concludes that finger jointing LVL represents a viable solution to manufacture usable LVL lengths from short LVL billets, but have lower edge bending efficiency than scarf jointed LVL

    Impact of accessory gene regulator (agr) dysfunction on vancomycin pharmacodynamics among Canadian community and health-care associated methicillin-resistant Staphylococcus aureus

    <p>Abstract</p> <p>Background</p> <p>The accessory gene regulator (<it>agr</it>) is a quorum sensing cluster of genes which control colonization and virulence in <it>Staphylococcus aureus</it>. We evaluated <it>agr </it>function in community- (CA) and healthcare-associated (HA) MRSA, to compare the pharmacodynamics and bactericidal activity of vancomycin against <it>agr </it>functional and dysfunctional HA-MRSA and CA-MRSA.</p> <p>Methods</p> <p>40 clinical isolates of MRSA from the Canadian Nosocomial Infection Surveillance Program were evaluated for delta-haemolysin production, as a surrogate marker of <it>agr </it>function. Time kill experiments were performed for vancomycin at 0 to 64 times the MIC against an initial inoculum of 10<sup>6 </sup>and 10<sup>8 </sup>cfu/ml of <it>agr </it>functional and dysfunctional CA-MRSA and HA-MRSA and these data were fit to a hill-type pharmacodynamic model.</p> <p>Results</p> <p>15% isolates were <it>agr </it>dysfunctional, which was higher among HA-MRSA (26.3%) versus CA-MRSA (4.76%). Against a low initial inoculum of 10<sup>6 </sup>cfu/ml of CA-MRSA, vancomycin pharmacodynamics were similar among <it>agr </it>functional and dysfunctional strains. However, against a high initial inoculum of 10<sup>8 </sup>cfu/ml, killing activity was notably attenuated against <it>agr </it>dysfunctional CA-MRSA (USA400) and HA-MRSA (USA100). CA-MRSA displayed a 20.0 fold decrease in the maximal reduction in bacterial counts (Emax) which was 3.71 log<sub>10 </sub>CFU/ml for <it>agr </it>functional vs. 2.41 log<sub>10 </sub>CFU/ml for <it>agr </it>dysfunctional MRSA (p = 0.0007).</p> <p>Conclusions</p> <p>Dysfunction in <it>agr </it>was less common among CA-MRSA vs. HA-MRSA. <it>agr </it>dysfunction demonstrated an impact on vancomycin bactericidal activity and pharmacodynamics against a high initial inoculum of CA-MRSA and HA-MRSA, which may have implications for optimal antimicrobial therapy against persistent, difficult to treat MRSA infections.</p

    Proteinase-activated receptor 2 modulates OA-related pain, cartilage and bone pathology

    Objective Proteinase-activated receptor 2 (PAR2) deficiency protects against cartilage degradation in experimental osteoarthritis (OA). The wider impact of this pathway upon OA-associated pathologies such as osteophyte formation and pain is unknown. Herein, we investigated early temporal bone and cartilage changes in experimental OA in order to further elucidate the role of PAR2 in OA pathogenesis. Methods OA was induced in wild-type (WT) and PAR2-deficient (PAR2−/−) mice by destabilisation of the medial meniscus (DMM). Inflammation, cartilage degradation and bone changes were monitored using histology and microCT. In gene rescue experiments, PAR2−/− mice were intra-articularly injected with human PAR2 (hPAR2)-expressing adenovirus. Dynamic weight bearing was used as a surrogate of OA-related pain. Results Osteophytes formed within 7 days post-DMM in WT mice but osteosclerosis was only evident from 14 days post induction. Importantly, PAR2 was expressed in the proliferative/hypertrophic chondrocytes present within osteophytes. In PAR2−/− mice, osteophytes developed significantly less frequently but, when present, were smaller and of greater density; no osteosclerosis was observed in these mice up to day 28. The pattern of weight bearing was altered in PAR2−/− mice, suggesting reduced pain perception. The expression of hPAR2 in PAR2−/− mice recapitulated osteophyte formation and cartilage damage similar to that observed in WT mice. However, osteosclerosis was absent, consistent with lack of hPAR2 expression in subchondral bone. Conclusions This study clearly demonstrates PAR2 plays a critical role, via chondrocytes, in osteophyte development and subchondral bone changes, which occur prior to PAR2-mediated cartilage damage. The latter likely occurs independently of OA-related bone changes

    Cloning and expression of two different genes from Streptococcus dysgalactiae encoding fibronectin receptors.

    Binding of bacteria to fibronectin has been implicated as a mechanism of bacterial adhesion to the host tissue. In this report we have analyzed the binding of a strain of Streptococcus dysgalactiae to fibronectin. The cells bind to a site in the NH2-terminal domain of the protein via trypsin-sensitive cell surface components. Furthermore, a lysate prepared by sonication of streptococcal cells contained fibronectin-binding proteins that inhibit the binding of the ligand to intact bacteria. When the proteins were separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, blotted to an Immobilon-P filter, and probed with 125I-labeled fibronectin, a 140-kDa fibronectin-binding protein was identified along with a number of smaller binding proteins. A genomic DNA library was constructed and screened for the expression of fibronectin-binding proteins. Two clones were isolated and shown to contain unrelated inserts by restriction mapping and cross-hybridization experiments. The two encoded proteins were also immunologically distinct although both bound to the same region of the fibronectin molecule, and both effectively inhibited the binding of 125I-fibronectin to bacterial cells. Immunological analyses showed that only one of the two proteins tentatively identified as fibronectin receptors was expressed in detectable quantities in the Streptococcus dysgalactiae strain under the culture conditions employed
