The MGDO software library for data analysis in Ge neutrinoless double-beta decay experiments
The GERDA and Majorana experiments will search for neutrinoless double-beta
decay of germanium-76 using isotopically enriched high-purity germanium
detectors. Although the experiments differ in conceptual design, they share
many aspects in common, and in particular will employ similar data analysis
techniques. The collaborations are jointly developing a C++ software library,
MGDO, which contains a set of data objects and interfaces to encapsulate, store
and manage physical quantities of interest, such as waveforms and high-purity
germanium detector geometries. These data objects define a common format for
persistent data, whether it is generated by Monte Carlo simulations or an
experimental apparatus, to reduce code duplication and to ease the exchange of
information between detector systems. MGDO also includes general-purpose
analysis tools that can be used for the processing of measured or simulated
digital signals. The MGDO design is based on the Object-Oriented programming
paradigm and is very flexible, allowing for easy extension and customization of
the components. The tools provided by the MGDO libraries are used by both GERDA

Non-invasive measurement of hepatic venous oxygen saturation (ShvO₂) with quantitative susceptibility mapping in normal mouse liver and livers bearing colorectal metastases
PURPOSE: The purpose of this prospective study was to investigate the potential of QSM to noninvasively measure hepatic venous oxygen saturation (ShvO2). Materials & Methods: All animal studies were performed in accordance with the UK Home Office Animals Science Procedures Act (1986) and UK National Cancer Research Institute (NCRI) guidelines. QSM data was acquired from a cohort of mice (n=10) under both normoxic (medical air, 21% O2/balance N), and hyperoxic conditions (100% O2). Susceptibility measurements were taken from large branches of the portal and hepatic vein under each condition and were used to calculate venous oxygen saturation in each vessel. Blood was extracted from the IVC of three mice under norm- and hyperoxic conditions, and oxygen saturation was measured using a blood gas analyser to act as a gold standard. QSM data was also acquired from a cohort of mice bearing colorectal liver metastases (CRLM). SvO2 was calculated from susceptibility measurements made in the portal and hepatic veins, and compared to the healthy animals. RESULTS: SvO2 calculated from QSM measurements showed a significant increase of 14.93% in the portal vein (p < 0.05), and an increase of 21.39% in the hepatic vein (p < 0.01). Calculated results showed excellent agreement with those from the blood gas analyser (26.14% increase). ShvO2 was significantly lower in the disease cohort (30.18 ± 11.6%), than the healthy animals (52.67 ± 17.8%) (p < 0.05), but differences in the portal vein were not significant. CONCLUSION: QSM is a feasible tool for non-invasively measuring hepatic venous oxygen saturation and can detect differences in oxygen consumption in livers bearing colorectal metastases
Changing Hydrozoan Bauplans by Silencing Hox-Like Genes
Regulatory genes of the Antp class have been a major factor for the invention and radiation of animal bauplans. One of the most diverse animal phyla are the Cnidaria, which are close to the root of metazoan life and which often appear in two distinct generations and a remarkable variety of body forms. Hox-like genes have been known to be involved in axial patterning in the Cnidaria and have been suspected to play roles in the genetic control of many of the observed bauplan changes. Unfortunately RNAi mediated gene silencing studies have not been satisfactory for marine invertebrate organisms thus far. No direct evidence supporting Hox-like gene induced bauplan changes in cnidarians have been documented as of yet. Herein, we report a protocol for RNAi transfection of marine invertebrates and demonstrate that knock downs of Hox-like genes in Cnidaria create substantial bauplan alterations, including the formation of multiple oral poles (“heads”) by Cnox-2 and Cnox-3 inhibition, deformation of the main body axis by Cnox-5 inhibition and duplication of tentacles by Cnox-1 inhibition. All phenotypes observed in the course of the RNAi studies were identical to those obtained by morpholino antisense oligo experiments and are reminiscent of macroevolutionary bauplan changes. An engaged approach to exploring issues around poverty and mental health: A reflective evaluation of the research process from researchers and community partners involved in the DeStress study
Background
Involving patients, service users, carers and members of the public in research has been part of health policy and practice in the UK for the last 15 years. However, low‐income communities tend to remain marginalized from the co‐design and delivery of mental health research, perpetuating the potential for health inequalities. Greater understanding is therefore needed on how to meaningfully engage low‐income communities in mental health research.
Objectives
To explore and articulate whether and how an engaged research approach facilitated knowledge coproduction relating to poverty and mental distress.
Setting
A reflective evaluation of community and researcher engagement in the DeStress study that took place in two low‐income areas of South‐west England.
Design
Reflective evaluation by the authors through on‐going feedback, a focus group and first‐person writing and discussion on experiences of working with the DeStress project, and how knowledge coproduction was influenced by an engaged research approach.
Results
An engaged research approach influenced the process and delivery of the DeStress project, creating a space where community partners felt empowered to coproduce knowledge relating to poverty‐related mental distress, treatment and the training of health professionals that would otherwise have been missed. We examine motivations for involvement, factors sustaining engagement, how coproduction influenced research analysis, findings and dissemination of outputs, and what involvement meant for different stakeholders.
Conclusion
Engaged research supported the coproduction of knowledge in mental health research with low‐income communities which led to multiple impacts
Search for Pauli Exclusion Principle Violating Atomic Transitions and Electron Decay with a P-type Point Contact Germanium Detector
A search for Pauli-exclusion-principle-violating K-alpha electron transitions
was performed using 89.5 kg-d of data collected with a p-type point contact
high-purity germanium detector operated at the Kimballton Underground Research
Facility. A lower limit on the transition lifetime of 5.8x10^30 seconds at 90%
C.L. was set by looking for a peak at 10.6 keV resulting from the x-ray and
Auger electrons present following the transition. A similar analysis was done
to look for the decay of atomic K-shell electrons into neutrinos, resulting in
a lower limit of 6.8x10^30 seconds at 90 C.L. It is estimated that the MAJORANA
DEMONSTRATOR, a 44 kg array of p-type point contact detectors that will search
for the neutrinoless double-beta decay of 76-Ge, could improve upon these
exclusion limits by an order of magnitude after three years of operation
