    Low-level seaweed supplementation improves iodine status in iodine-insufficient women

    Iodine insufficiency is now a prominent issue in the UK and other European countries due to low intakes of dairy products and seafood (especially where iodine fortification is not in place). In the present study, we tested a commercially available encapsulated edible seaweed (Napiers Hebridean Seagreens® Ascophyllum nodosumspecies) for its acceptability to consumers and iodine bioavailability and investigated the impact of a 2-week daily seaweed supplementation on iodine concentrations and thyroid function. Healthy non-pregnant women of childbearing age, self-reporting low dairy product and seafood consumption, with no history of thyroid or gastrointestinal disease were recruited. Seaweed iodine (712 μg, in 1 g seaweed) was modestly bioavailable at 33 (interquartile range (IQR) 28–46) % of the ingested iodine dose compared with 59 (IQR 46–74) % of iodine from the KI supplement (n 22). After supplement ingestion (2 weeks, 0•5 g seaweed daily, <i>n</i> 42), urinary iodine excretion increased from 78 (IQR 39–114) to 140 (IQR 103–195) μg/l (<i>P</i>< 0•001). The concentrations of thyroid-stimulating hormone increased from 1•5 (IQR 1•2–2•2) to 2•1 (IQR 1•3–2•9) mIU/l (<i>P</i>< 0•001), with two participants having concentrations exceeding the normal range after supplement ingestion (but normal free thyroxine concentrations). There was no change in the concentrations of other thyroid hormones after supplement ingestion. The seaweed was palatable and acceptable to consumers as a whole food or as a food ingredient and effective as a source of iodine in an iodine-insufficient population. In conclusion, seaweed inclusion in staple foods would serve as an alternative to fortification of salt or other foods with KI

    Decaying dark matter: a stacking analysis of galaxy clusters to improve on current limits

    We show that a stacking approach to galaxy clusters can improve current limits on decaying dark matter by a factor ≳5−100\gtrsim 5-100, with respect to a single source analysis, for all-sky instruments such as Fermi-LAT. Based on the largest sample of X-ray-selected galaxy clusters available to date (the MCXC meta-catalogue), we provide all the astrophysical information, in particular the astrophysical term for decaying dark matter, required to perform an analysis with current instruments.Comment: 6 pages, 3 figures, supplementary file available on demand, accepted for publication in PR

    Changing guards: time to move beyond Body Mass Index for population monitoring of excess adiposity

    With the obesity epidemic, and the effects of aging populations, human phenotypes have changed over two generations, possibly more dramatically than in other species previously. As obesity is an important and growing hazard for population health, we recommend a systematic evaluation of the optimal measure(s) for population-level excess body fat. Ideal measure(s) for monitoring body composition and obesity should be simple, as accurate and sensitive as possible, and provide good categorisation of related health risks. Combinations of anthropometric markers or predictive equations may facilitate better use of anthropometric data than single measures to estimate body composition for populations. Here we provide new evidence that increasing proportions of aging populations are at high health-risk according to waist circumference, but not body mass index (BMI), so continued use of BMI as the principal population-level measure substantially underestimates the health-burden from excess adiposity

    Fat transforms ascorbic acid from inhibiting to promoting acid-catalysed N-nitrosation

    <b>Background</b>: The major potential site of acid nitrosation is the proximal stomach, an anatomical site prone to a rising incidence of metaplasia and adenocarcinoma. Nitrite, a pre-carcinogen present in saliva, can be converted to nitrosating species and N-nitroso compounds by acidification at low gastric pH in the presence of thiocyanate. <b>Aims</b>: To assess the effect of lipid and ascorbic acid on the nitrosative chemistry under conditions simulating the human proximal stomach. <b>Methods</b>: The nitrosative chemistry was modelled in vitro by measuring the nitrosation of four secondary amines under conditions simulating the proximal stomach. The N-nitrosamines formed were measured by gas chromatography–ion-trap tandem mass spectrometry, while nitric oxide and oxygen levels were measured amperometrically. <b>Results</b>: In absence of lipid, nitrosative stress was inhibited by ascorbic acid through conversion of nitrosating species to nitric oxide. Addition of ascorbic acid reduced the amount of N-nitrosodimethylamine formed by fivefold, N-nitrosomorpholine by .1000-fold, and totally prevented the formation of N-nitrosodiethylamine and N-nitrosopiperidine. In contrast, when 10% lipid was present, ascorbic acid increased the amount of Nnitrosodimethylamine, N-nitrosodiethylamine and N-nitrosopiperidine formed by approximately 8-, 60- and 140-fold, respectively, compared with absence of ascorbic acid. <b>Conclusion</b>: The presence of lipid converts ascorbic acid from inhibiting to promoting acid nitrosation. This may be explained by nitric oxide, formed by ascorbic acid in the aqueous phase, being able to regenerate nitrosating species by reacting with oxygen in the lipid phase

    The anti-adhesive effect of curcumin on Candida albicans biofilms on denture materials

    The use of natural compounds as an alternative source of antimicrobials has become a necessity given the growing concern over global antimicrobial resistance. Polyphenols, found in various edible plants, offers one potential solution to this. We aimed to investigate the possibility of using curcumin within the context of oral health as a way of inhibiting and preventing the harmful development of Candida albicans biofilms. We undertook a series of adsorption experiments with varying concentrations of curcumin, showing that 50 ug/ml could prevent adhesion. This effect could be further synergised by the curcumin pretreatment of yeast cells to obtain significantly greater inhibition (>90, p<0.001). Investigation of the biological impact of curcumin showed that it preferentially affected immature morphological forms (yeast and germlings), and actively promoted aggregation of the cells. Transcriptional analyses showed that key adhesins were down-regulated (ALS1 and ALS3), whereas aggregation related genes (ALS5 and AAF1) were up-regulated. Collectively, these data demonstrated that curcumin elicits anti-adhesive effects and that induces transcription of genes integrally involved in the processes related to biofilm formation. Curcumin and associated polyphenols therefore have the capacity to be developed for use in oral healthcare to augment existing preventative strategies for candidal biofilms on the denture surface

    Dark matter annihilation and decay profiles for the Reticulum II dwarf spheroidal galaxy

    The dwarf spheroidal galaxies (dSph) of the Milky Way are among the most attractive targets for indirect searches of dark matter. In this work, we reconstruct the dark matter annihilation (J-factor) and decay profiles for the newly discovered dSph Reticulum II. Using an optimized spherical Jeans analysis of kinematic data obtained from the Michigan/Magellan Fiber System (M2FS), we find Reticulum II's J-factor to be among the largest of any Milky Way dSph. We have checked the robustness of this result against several ingredients of the analysis. Unless it suffers from tidal disruption or significant inflation of its velocity dispersion from binary stars, Reticulum II may provide a unique window on dark matter particle properties.Comment: 5 pages, 4 figures.     Oxidative stress, protein glycation and nutrition – interactions relevant to health and disease throughout the lifecycle

    Protein glycation has been studied for over a century now and plays an important role in disease pathogenesis throughout the lifecycle. Strongly related to diabetic complications, glycation of Hb has become the gold standard method for diabetes diagnosis and monitoring. It is however attracting attention in normoglycaemia as well lately. Longitudinal studies increasingly suggest a positive relationship between glycation and the risk of chronic diseases in normoglycaemic individuals, but the mechanisms behind this association remain unclear. The interaction between glycation and oxidative stress may be particularly relevant in the normoglycaemic context, as suggested by recent epidemiological and in vitro evidence. In that context nutritional and lifestyle factors with an influence on redox status, such as smoking, fruit and vegetable and antioxidants consumption, may have the capacity to promote or inhibit glycation. However, experimental data from controlled trials are lacking the quality and rigour needed to reach firm conclusions. In the present review, we discuss the importance of glycation for health through the lifecycle and focus on the importance of oxidative stress as a driver for glycation. The importance of nutrition to modulate glycation is discussed, based on the evidence available and recommendations towards higher quality future research are made

    Direct evidence for the magnetic ordering of Nd ions in NdMn2_2Si2_2 and NdMn2_2Ge2_2 by high resolution inelastic neutron scattering

    We have investigated the low energy nuclear spin excitations in NdMn2_2Si2_2 and NdMn2_2Ge2_2 by high resolution inelastic neutron scattering. Previous neutron diffraction investigations gave ambiguous results about Nd magnetic ordering at low temperatures. The present element-specific technique gave direct evidence for the magnetic ordering of Nd ions. We found considerable difference in the process of the Nd magnetic ordering at low temperature in NdMn2_2Si2_2 and NdMn2_2Ge2_2. Our results are consistent with those of magnetization and recent neutron diffraction measurements
