NRF2 regulates viability, proliferation, resistance to oxidative stress, and differentiation of murine myoblasts and muscle satellite cells
Increased oxidative stress can slow down the regeneration of skeletal muscle and affect the activity of muscle satellite cells (mSCs). Therefore, we evaluated the role of the NRF2 transcription factor (encoded by the Nfe2l2 gene), the main regulator of the antioxidant response, in muscle cell biology. We used (i) an immortalized murine myoblast cell line (C2C12) with stable overexpression of NRF2 and (ii) primary mSCs isolated from wild-type and Nfe2l2 (transcriptionally)-deficient mice (Nfe2l2). NRF2 promoted myoblast proliferation and viability under oxidative stress conditions and decreased the production of reactive oxygen species. Furthermore, NRF2 overexpression inhibited C2C12 cell differentiation by down-regulating the expression of myogenic regulatory factors (MRFs) and muscle-specific microRNAs. We also showed that NRF2 is indispensable for the viability of mSCs since the lack of its transcriptional activity caused high mortality of cells cultured in vitro under normoxic conditions. Concomitantly, Nfe2l2 mSCs grown and differentiated under hypoxic conditions were viable and much more differentiated compared to cells isolated from wild-type mice. Taken together, NRF2 significantly influences the properties of myoblasts and muscle satellite cells. This effect might be modulated by the muscle microenvironment
The aromatase expression in myomas and myometriums of women in reproduction and perimenopausal age.
Uterine myomas represent one of the most common female pathologies. Uterine smooth muscle myomas or fibromas are benign tumours which respond to hormones and their etiology induces wide interest. The myomas were found to contain aromatase and, in addition, cells of the myomas were found to synthesize estrogen. This study was conducted on patients with the myomas, in either generative age or in the perimenopausal period. Expression of aromatase was detected in patients of various age, with large or small uterine myomas, using an immunohistochemical technique. Magnetic properties of Fe76X2Si8B14 (X= Al, Cr, Mo) amorphous alloys
Purpose: The idea of the paper is to study the influence of thermal annealing and alloying additions on magnetic properties, optimization and crystallization processes in Fe76X2Si8B14 (X=Al, Cr, Mo) amorphous alloys.
Design/methodology/approach: For annealed samples (1 h, Ta ranging from 300 K to 800 K) at room
temperature magnetic permeability was measured by applying Maxwell-Wien bridge (frequency about 1030 Hz
and magnetic field H=0.5 A/m). Magnetostriction coefficients – parallel and perpendicular were determined by
applying infra-red magneto-dilatometer. Magnetization in saturation versus temperature was measured by making use of magnetic balance (field 0.5 T).
Findings: It was shown that alloying additions in the examined alloys cause a decrease of the Curie temperature, an increase of magnetic permeability and magnetization in saturation. The observed ESMP (enhancement of soft magnetic properties) effect in the examined alloys can be attributed to the so-called relaxed amorphous phase free iron nanograins. It was shown that parallel and perpendicular magnetostriction coefficients depend on annealing temperatures which means that these quantities are sensitive on free volume content.
Research limitations/implications: The obtained results are a part of a broad area of examinations devoted
to establishing of the influence of different alloying additions and thermal annealing on soft magnetic properties
of amorphous alloys obtained by melt spinning technique.
Practical implications: The examined alloys belong to a modern group of soft magnetic materials, which can
be used as core transformers, magnetic sensors, shields of magnetic etc. The obtained results may be used for
preparing soft magnetic ribbons for specific applications.
Originality/value: The originality of the paper lies in examination of the influence of free volume content on
magnetostriction coefficients
Comparative two time-point proteome analysis of the plasma from preterm infants with and without bronchopulmonary dysplasia
Background: In this study, we aimed to analyze differences in plasma protein abundances between infants with
and without bronchopulmonary dysplasia (BPD), to add new insights into a better understanding of the pathogenesis
of this disease.
Methods: Cord and peripheral blood of neonates (≤ 30 weeks gestational age) was drawn at birth and at the 36th
postmenstrual week (36 PMA), respectively. Blood samples were retrospectively subdivided into BPD(+) and BPD(−)
groups, according to the development of BPD.
Results: Children with BPD were characterized by decreased afamin, gelsolin and carboxypeptidase N subunit 2 levels
in cord blood, and decreased galectin-3 binding protein and hemoglobin subunit gamma-1 levels, as well as an
increased serotransferrin abundance in plasma at the 36 PMA.
Conclusions: BPD development is associated with the plasma proteome changes in preterm infants, adding further
evidence for the possible involvement of disturbances in vitamin E availability and impaired immunological processes
in the progression of prematurity pulmonary complications. Moreover, it also points to the differences in proteins
related to infection resistance and maintaining an adequate level of hematocrit in infants diagnosed with BPD
Surgical treatment of rectal cancer in Poland — a report from a prospective, multi-centre observational study PSSO_01 conducted under the auspices of the Polish Society of Surgical Oncology
Introduction. Since 2016, as part of the PSSO_01 multi-centre research project conducted under the auspices of the Polish Society of Surgical Oncology, clinical data on rectal cancer treatment have been collected. The objective of the study was to illustrate the state of early results of surgical treatment. Material and methods. The research project is multi-centre in nature. Data shall be collected electronically. The study protocol does not impose or suggest any course of procedure. It only systematizes the way data are collected for scientific purposes. The analysis of early results of surgical treatment was compared with the results of population studies from other European countries (Netherlands, Belgium). Results. By the end of June 2018, 736 patients were registered in the study. In 399 (54.2%) an anterior resection was performed. More than half of patients undergoing subsequent surgical treatment (54.2%) receive neoadjuvant treatment, with the percentage of patients undergoing radiotherapy or radiochemical treatment for lower rectal cancer being about 70%. Most patients (96%) are operated in elective procedure. The percentage of laparoscopic surgeries is low (8.6%). Postoperative complications are observed in 21.1% of patients. Severe complications (grades III–V according to Clavien-Dindo classification) occur in 7.6% of patients undergoing surgery. Postoperative mortality is 1.1%. Discussion. Although the project does not have the character of a registry and does not allow for drawing wider conclusions concerning the compliance with the standards of qualification for neoadjuvant treatment, the important information is that more than half of rectal cancer patients receive preoperative treatment, and the percentage of severe postoperative complications does not exceed 10%. Conclusions. The results of the PSSO_01 project are representative and reflect the actual situation concerning surgical treatment of rectal cancer patients in Poland
Leczenie chirurgiczne raka odbytnicy w Polsce — raport z prospektywnego, wieloośrodkowego badania obserwacyjnego PSSO_01 prowadzonego pod auspicjami Polskiego Towarzystwa Chirurgii Onkologicznej
Wstęp. Od 2016 roku w ramach wieloośrodkowego projektu badawczego PSSO_01 prowadzonego pod auspicjami Polskiego Towarzystwa Chirurgii Onkologicznej, gromadzone są dane kliniczne dotyczące leczenia raka odbytnicy. Celem pracy było zobrazowanie stanu dotyczącego wczesnych wyników leczenia chirurgicznego.
Materiał i metody. Projekt badawczy ma charakter wieloośrodkowy. Dane gromadzone są elektronicznie. Protokół badania nie narzuca ani nie sugeruje żadnego sposobu postępowania, systematyzuje jedynie sposób zbierania danych w celach naukowych. Analiza dotycząca wczesnych wyników leczenia chirurgicznego została porównana z rezultatami populacyjnych badań z innych krajów europejskich (Holandia, Belgia).
Wyniki. Do końca czerwca 2018 roku w badaniu zarejestrowano 736 chorych. U 399 (54,2%) wykonano resekcję przednią. Leczenie neoadiuwantowe otrzymała ponad połowa chorych, poddanych następnie leczeniu operacyjnemu (54,2%), przy czym odsetek pacjentów poddanych radio- bądź radiochemioterapii z powodu raka dolnej części odbytnicy wyniósł około 70%. Większość chorych (96%) operowana była w trybie planowym. Odsetek operacji wykonanych techniką laparoskopową jest niski (8,6%). Powikłania pooperacyjne zaobserwowano u 21,1% chorych. Ciężkie powikłania (III–V st. wg klasyfikacji Claviena-Dindo) sięgały 7,6% operowanych chorych. Śmiertelność pooperacyjna wyniosła 1,1%.
Dyskusja. Chociaż projekt nie ma charakteru rejestru i nie pozwala na wyciągniecie szerszych wniosków dotyczących przestrzegania standardów kwalifikacji do leczenia neoadiuwantowego, istotną informacją jest to, że ponad połowa chorych na raka odbytnicy otrzymuje leczenie przedoperacyjne, a odsetek ciężkich powikłań pooperacyjnych nie przekracza 10%.
Wnioski. Wyniki projektu PSSO_01 są reprezentatywne i odzwierciedlają faktyczną sytuację dotyczącą leczenia chirurgicznego chorych na raka odbytnicy w Polsce
