    Scanning Electron Microscopic Studies on Microvascular Architecture of Human Coronary Vessels by Corrosion Casts: Normal and Focal Necrosis

    Microvascular architecture of the normal human heart and myocardial focal necrosis were studied by scanning electron microscopy of corrosion casts. Casts macroscopically identical in form to the left ventricular posterior wall were prepared. The following results were obtained in the normal human heart. (I) Most of the arterioles communicated with capillary plexuses smoothly and straight forwardly in the left ventricular posterior free wall. (2) Arterioles which branched from the arteries ran in various directions and continued into capillaries either at right angles or obliquely in the trabeculae carneae. (3) capillaries running parallel with the cardiac muscle fibers ran in different directions to cross over with each other in different layers of myocardium. Capillaries in the myocardium formed a continuous and coarse net-like architecture with many bifurcations and anastomoses. Capillaries were about 5-7 μm in diameter. (4) Some veins gathering capillaries in the epicardium ran into the myocardium and the others ran in the epicardium. Veins connecting with capillaries in the myocardium ran in the myocardial layer and communicated with larger veins. (5) An arteriovenous anastomosis and two different types of venous-venous anastomoses were observed in the left ventricular posterior wall. At the site of focal necrosis, cross sections of dilated vessels were observed in large numbers by light microscopy and scanning electron microscopy. (I) At the site of focal necrosis, dilated capillaries running with tortuosity were seen in large numbers by scanning electron microscopy of corrosion casts. (2) When compared with vessels in the normal myocardium, small arterial branches were dilated and run tortuously. (3) These dilated capillary plexuses were observed in the area which communicated with twigs branching off at the right angle from the arterial branch

    Genotypic Characterization of Non-O157 Shiga Toxin–Producing Escherichia coli in Beef Abattoirs of Argentina

    The non-O157 Shiga toxin-producing Escherichia coli (STEC) contamination in carcasses and feces of 811 bovines in nine beef abattoirs from Argentina was analyzed during a period of 17 months. The feces of 181 (22.3%) bovines were positive for non-O157 STEC, while 73 (9.0%) of the carcasses showed non-O157 STEC contamination. Non-O157 STEC strains isolated from feces (227) and carcasses (80) were characterized. The main serotypes identified were O178:H19, O8:H19, O130:H11, and O113:H21, all of which have produced sporadic cases of hemolytic-uremic syndrome in Argentina and worldwide. Twenty-two (7.2%) strains carried a fully virulent stx/eae/ehxA genotype. Among them, strains of serotypes O103:[H2], O145:NM, and O111:NM represented 4.8% of the isolates. XbaI pulsed-field gel electrophoresis pattern analysis showed 234 different patterns, with 76 strains grouped in 30 clusters. Nine of the clusters grouped strains isolated from feces and from carcasses of the same or different bovines in a lot, while three clusters were comprised of strains distributed in more than one abattoir. Patterns AREXSX01.0157, AREXBX01.0015, and AREXPX01.0013 were identified as 100% compatible with the patterns of one strain isolated from a hemolytic-uremic syndrome case and two strains previously isolated from beef medallions, included in the Argentine PulseNet Database. In this survey, 4.8% (39 of 811) of the bovine carcasses appeared to be contaminated with non- O157 STEC strains potentially capable of producing sporadic human disease, and a lower proportion (0.25%) with strains able to produce outbreaks of severe disease.Fil: Masana, Marcelo.     Random amplification of polymorphic DNA reveals clonal relationships among enteropathogenic Escherichia coli isolated from non-human primates and humans

    Enteropathogenic Escherichia coli ( EPEC) strains are important agents of infantile diarrhea all over the world, gaining even greater importance in developing countries. EPEC have also been isolated from various animal species, but most isolates belong to serotypes that differ from those recovered from humans. However, it has been demonstrated that several isolates from non- human primates belong to the serogroups and/ or serotypes related to those implicated in human disease. The objective of this study was to evaluate the genetic differences between thirteen strains isolated from non- human primates and the same number of strains isolated from human infections. Human isolates belonged to the same serogroup/ serotype as the monkey strains and the evaluation was done by analysis of random amplified polymorphic DNA. Dendrogram analysis showed that there was no clustering between human and monkey strains. Human and non- human isolates of the EPEC serotypes O127:H40 and O128:H2 shared 90 and 87% of their bands, respectively, indicating strong genomic similarity between the strains, leading to the speculation that they may have arisen from the same pathogenic clone. To our knowledge, this study is the first one comparing genomic similarity between human and non- human primate strains and the results provide further evidence that monkey EPEC strains correlate with human EPEC, as suggested in a previous investigation

    Multiple episodes of ice loss from the Wilkes Subglacial Basin during the Last Interglacial

    The Last Interglacial (LIG: 130,000-115,000 years ago) was a period of warmer global mean temperatures and higher and more variable sea levels than the Holocene (11,700-0 years ago). Therefore, a better understanding of Antarctic ice-sheet dynamics during this interval would provide valuable insights for projecting sea-level change in future warming scenarios. Here we present a high-resolution record constraining ice-sheet changes in the Wilkes Subglacial Basin (WSB) of East Antarctica during the LIG, based on analysis of sediment provenance and an ice melt proxy in a marine sediment core retrieved from the Wilkes Land margin. Our sedimentary records, together with existing ice-core records, reveal dynamic fluctuations of the ice sheet in the WSB, with thinning, melting, and potentially retreat leading to ice loss during both early and late stages of the LIG. We suggest that such changes along the East Antarctic Ice Sheet margin may have contributed to fluctuating global sea levels during the LIG

    Febre purpúrica brasileira: caracterização rápida das cepas invasoras de Haemophilus aegyptius

    Cepas de H. aegyptius isoladas em surtos de Febre Purpúrica Brasileira (FPB) no Brasil, foram caracterizadas pelo método de aglutinação em lâmina utilizando um anti-soro produzido com cepa de H. aegyptius isolada de cultura de sangue de paciente com FPB. Através desse método foi possível identificar cepas de H. aegyptius responsáveis por surtos de conjuntivite com características antigênicas iguais às cepas isoladas de FPB. A sensibilidade e especificidade da soroaglutinação em lâmina foi de 97,7% e 89,6% respectivamente, podendo ser utilizado como método de triagem em estudos de conjuntivites purulentas, para detectar cepas invasivas de H. aegyptius associadas a FPB, possibilitando assim a implantação de medidas que ampliem a eficiência na prevenção e na vigilância epidemiológica da doença.Strains of H. aegyptius isolated during outbreak of Brazilian Purpuric Fever (BPF) in Brazil were characterized antigenically by slide agglutination test utilizing antiserum produced with a H. aegyptius strain isolated from blood culture from a patient with BPF. By means of this method, it were identified H. aegyptius strains responsible for outbreaks of conjunctivitis with identical antigenic characteristics to strains isolated from BPF. The sensitivity and specificity of slide seroagglutination test was 97.7% and 89.6% respectively; therefore this assay was efficient to be used as a screening method in the studies of purulent conjunctivitis for detecting high risk populations for BPF, and to implement measures that will increase the efficiency of epidemiologic surveillance

    Surto alimentar por Salmonella Enteritidis no Noroeste do Estado de São Paulo, Brasil

    A foodborne outbreak which affected 211 persons occurred, in a School, in 1993. The epidemiological data obtained by interviewing the affected and non affected persons sampled showed as predominant symptoms: diarrhoea, fever (77.7%), abdominal cramps (67.7%), vomiting (65.8%), hot-and-cold sensations (54.5%) and headache (44.5%). The median incubation period was of 17 hours, the limits being 3 and 29 hours. The disease period was of from 3 to 4 days. The food concerned was a kind of paté, a mayonnaise mixture prepared with fresh eggs with boiled potatoes that was consumed with bread. The biological material analysis - 3 coprocultures, and leftovers of the food revealed the presence of one and the same organism: Salmonella Enteritidis. In the food, the numbers of this bacterium per gram were sufficient to account for the manifestation of the disease (104 and 105/g). The antibiogram of all the isolates showed the same sensibility pattern. The preparation related to this outbreak suggests the endogenous contamination of the eggs; the cross contamination - the outbreak affected three school periods, as the food was prepared separately for each school period; and the conditions under which the food was kept during the time from preparation to consumption. The observation of the 3 food handlers, by successive coprocultures, for one week, indicates that they were not asymptomatic carriers nor were they affected as a result of this outbreak by the causal bacteria.Em 1993 ocorreu um surto alimentar em escola, com 211 afetados. Os dados epidemiológicos levantados por entrevista de amostragem de afetados e não afetados mostraram que os sintomas predominantes foram diarréia, febre (77,7%), dor abdominal (67,7%), vômito (65,8%), calafrios (54,5%) e cefaléia (44,5%). A mediana de incubação foi de 17 horas, com limites entre 3 e 29 horas. A duração da doença foi de 3 a 4 dias. O alimento consumido foi um tipo de patê, mistura de molho de maionese preparada com ovos crus com batata cozida, passado em pão. A análise de material biológico (3 coproculturas) e de restos de alimentos revelou a presença do mesmo microrganismo, a Salmonella Enteritidis. No caso dos alimentos, o número encontrado desta bactéria por gramo de produto era compatível com a quantidade de células necessária para desencadear a doença (10(4)e 10(5)/g). O antibiograma de todas as cepas isoladas revelou o mesmo padrão de sensibilidade. As falhas no preparo do alimento relacionadas com o levantamento indicam a possibilidade de contaminação endógena dos ovos; contaminação cruzada - o surto afetou três períodos escolares, sendo que para cada um o alimento foi preparado em separado - e as condições de manutenção do alimento após preparo e até o consumo. A observação por uma semana seguida das 3 merendeiras envolvidas, através de coprocultura, não indicaram que as mesmas fossem portadoras assintomáticas desta bactéria ou que tivessem sido envolvidas no surto em questão
