    Elevated impulsivity and impaired decision-making cognition in heavy users of MDMA ("Ecstasy”)

    Rationale: In animal studies, the common club drug 3,4-methylendioxymethamphetamine (MDMA, "Ecstasy”) consistently caused a prolonged loss of presynaptic serotonergic neurons, and evidence suggests that MDMA consumption may also affect the human serotonergic system. Serotonin (5-HT) has been implicated in the regulation of impulsivity and such executive functions as decision-making cognition. In fact, MDMA users have shown elevated impulsivity in two studies, but little is known about decision making in drug-free MDMA consumers. Objective: The aim of this study was to examine the cognitive neurotoxicity of MDMA with regard to behavioral impulsivity and decision-making cognition. Methods: Nineteen male, abstinent, heavy MDMA users; 19 male, abstinent cannabis users; and 19 male, drug-naïve controls were examined with the Matching Familiar Figures Test (MFFT) as well as with a Go/No-Go Task (GNG) for impulsivity and with a Gambling Task (GT) for executive functioning. Results: MDMA users showed significantly elevated impulsivity in the MFFT Impulsivity score (I-score), but not in commission errors of the GNG, compared with controls. Cannabis users did not yield altered impulsivity compared with controls. In the GT, MDMA users performed significantly worse than cannabis consumers and controls, whereas cannabis users exhibited the same decision-making capacity as controls. In addition, the I-score as well as the decision-making performance was correlated with measures of MDMA intake. The I-score and the decision-making performance were also correlated. Conclusion: These results suggest that heavy use of MDMA may elevate behavioral impulsivity and impair decision-making cognition possibly mediated by a selective impairment of the 5-HT syste

    Autobiographical Memory for Emotional Events in Amnesia

    This study investigated autobiographical memory for emotionally flavoured experiences in amnesia. Ten amnesic patients and 10 matched control subjects completed the Autobiographical Memory Interview and three semi-structured interviews which assessed memory for personal events associated with pain, happiness and fear. Despite retrograde amnesia for autobiographical facts and incidents, amnesics remembered a similar number of emotionally significant personal experiences as control subjects. Their recollections generally lacked elaboration and detail, but pain-related memories appeared to be more mildly impaired than memories associated with happiness and fear. The findings are discussed in relation to recent views on the relationship between affect and memory

    Altered Error Processing following Vascular Thalamic Damage: Evidence from an Antisaccade Task

    Event-related potentials (ERP) research has identified a negative deflection within about 100 to 150 ms after an erroneous response – the error-related negativity (ERN) - as a correlate of awareness-independent error processing. The short latency suggests an internal error monitoring system acting rapidly based on central information such as an efference copy signal. Studies on monkeys and humans have identified the thalamus as an important relay station for efference copy signals of ongoing saccades. The present study investigated error processing on an antisaccade task with ERPs in six patients with focal vascular damage to the thalamus and 28 control subjects. ERN amplitudes were significantly reduced in the patients, with the strongest ERN attenuation being observed in two patients with right mediodorsal and ventrolateral and bilateral ventrolateral damage, respectively. Although the number of errors was significantly higher in the thalamic lesion patients, the degree of ERN attenuation did not correlate with the error rate in the patients. The present data underline the role of the thalamus for the online monitoring of saccadic eye movements, albeit not providing unequivocal evidence in favour of an exclusive role of a particular thalamic site being involved in performance monitoring. By relaying saccade-related efference copy signals, the thalamus appears to enable fast error processing. Furthermore early error processing based on internal information may contribute to error awareness which was reduced in the patients

    Abstract. Many human lesion und functional brain imaging studies suggest involvement of the cerebellum in cognitive functions. However, negative and inconsistent findings are rarely discussed. It is still an open question as to which areas of cognition the cerebellum contributes, as well as how, and to what extent. Frequently cited earlier findings in one area of cognition have been challenged in more recent studies, that is the cerebellum may not be directly involved in attention. Furthermore, disorders in patients with acquired cerebellar disease are frequently mild and less severe compared to lesions of the corresponding areas of the cerebral cortex. Patients with cerebellar disease often perform within the normal range of neuropsychological test norms. This pattern is illustrated based on general intelligence and verbal working memory, which have been assessed by a large number of authors using comparable tests. Findings, however, appear to be more pronounced in individual cases with acute onset cerebellar disorders and in children, in particular with congenital disease. The review suggests that the inconsistencies in cognitive impairments may offer clues as to the nature of cerebellar cognitive involvement

    The Effect of Semantic Categorisation on Recall Memory in Amnesia

    Amnesic patients were compared to a healthy control group on recall of word lists containing semantically-related or unrelated words. As expected on the basis of previous literature, the amnesic group performed below the control group on all measures of recall. When total recall scores for each list were used as the index of performance, their scores were not significantly affected by the type of list, unlike those of the control group. Comparison of serial position effects for different parts of the lists revealed that the control group derived greater benefit from semantic relatedness in recall of items from the middle positions. This effect was not shown by the amnesic group, who showed similar U-shaped serial position curves for recall of all three lists, and appeared to use a more passive recall strategy than the control group. The findings are discussed in relation to our current understanding of amnesic deficits

    Narcolepsy: Pathophysiology and Neuropsychological Changes

    Narcolepsy is now recognized as a distinctive disorder with specific pathophysiology and neurochemical abnormalities. Findings on the role of the neuropeptide hypocretin are opening new avenues of research and new strategies for therapy. Recently, neuropsychological and electrophysiological studies have provided evidence for reduced memory performance on standard memory tests in addition to subjective complaints of forgetfulness which may be related to changes in attentional processing. Further studies are, however, necessary to clarify the neuropsychological profile in narcolepsy. This review focuses on the recent advances in understanding narcolepsy

    Developmental Prosopagnosia: A Review

    This article reviews the published literature on developmental prosopagnosia, a condition in which the ability to recognize other persons by facial information alone has never been acquired. Due to the very low incidence of this syndrome, case reports are sparse. We review the available data and suggest assessment strategies for patients suffering from developmental prosopagnosia. It is suggested that developmental prosopagnosia is not a unitary condition but rather consists of different subforms that can be dissociated on the grounds of functional impairments. On the basis of the available evidence, hypotheses about the aetiology of developmental prosopagnosia as well as about the selectivity of deficits related to face recognition are discussed
