    Very High Cycle Corrosion Fatigue Study of the Collapsed Polcevera Bridge, Italy

    A possible scenario is proposed to put into evidence how the combined effect of fatigue at very high number of cycles and corrosion could have been responsible for the failure of one of the strands and the subsequent collapse of the so-called balanced system of the Polcevera Bridge designed by Morandi. The analysis accounts for an actual estimation of the heavy-lorry traffic and load spectrum, as well as for the European Standards prescription for the fatigue damage accumulation assessment. In addition, the effective construction phases of the viaduct are considered. The structural analysis is carried out by means of analytical models, in order to simplify the structure complexity without prejudice to the description of the most relevant aspects of the structural behavior. The main purpose is to warn the scientific community and the public administrations that the combined effects of low-amplitude fatigue and corrosion can be dangerously underestimated, and that the existing assets of 20th century bridges deserve special attention in this respect

    Disease of the Year : Differential Diagnosis of Uveitic Macular Edema

    Uveitic cystoid macular edema (UME) is an important cause of visual morbidity among patients with both infectious and non-infectious uveitis. UME may be associated in more than 30% cases of active uveitis. However, even patients with minimal features of intraocular inflammation may develop recurrent or chronic UME. Therefore, the evaluation and management of UME in patients with uveitis may be challenging. A number of vitreoretinal pathologies may result in UME and accumulation of fluid in the intra- or subretinal space. These need to be carefully distinguished from each other so that appropriate management can be initiated. All types of uveitis, including anterior uveitis (where the primary site of inflammation is not in the posterior segment) can present with UME. Other conditions such as diabetes, and surgery, can present with macular edema. This index review highlights various differential diagnoses of UME and provides illustrative case examples with multimodal imaging evaluation

    PRMT6 diminishes HIV-1 Rev binding to and export of viral RNA

    BACKGROUND: The HIV-1 Rev protein mediates nuclear export of unspliced and partially spliced viral RNA through interaction with the Rev response element (RRE) by means of an arginine rich motif that is similar to the one found in Tat. Since Tat is known to be asymmetrically arginine dimethylated by protein arginine methyltransferase 6 (PRMT6) in its arginine rich motif, we investigated whether the Rev protein could act as a substrate for this enzyme. RESULTS: Here, we report the methylation of Rev due to a single arginine dimethylation in the N-terminal portion of its arginine rich motif and the association of Rev with PRMT6 in vivo. Further analysis demonstrated that the presence of increasing amounts of wild-type PRMT6, as well as a methylation-inactive mutant PRMT6, dramatically down-regulated Rev protein levels in concentration-dependent fashion, which was not dependent on the methyltransferase activity of PRMT6. Quantification of Rev mRNA revealed that attenuation of Rev protein levels was due to a posttranslational event, carried out by a not yet defined activity of PRMT6. However, no relevant protein attenuation was observed in subsequent chloramphenicol acetyltransferase (CAT) expression experiments that screened for RNA export and interaction with the RRE. Binding of the Rev arginine rich motif to the RRE was reduced in the presence of wild-type PRMT6, whereas mutant PRMT6 did not exert this negative effect. In addition, diminished interactions between viral RNA and mutant Rev proteins were observed, due to the introduction of single arginine to lysine substitutions in the Rev arginine rich motif. More importantly, wild-type PRMT6, but not mutant methyltransferase, significantly decreased Rev-mediated viral RNA export from the nucleus to the cytoplasm in a dose-dependent manner.     3D Bioprinting of Gelatin–Xanthan Gum Composite Hydrogels for Growth of Human Skin Cells

    In recent years, bioprinting has attracted much attention as a potential tool for generating complex 3D biological constructs capable of mimicking the native tissue microenvironment and promoting physiologically relevant cell–cell and cell–matrix interactions. The aim of the present study was to develop a crosslinked 3D printable hydrogel based on biocompatible natural polymers, gelatin and xanthan gum at different percentages to be used both as a scaffold for cell growth and as a wound dressing. The CellInk Inkredible 3D printer was used for the 3D printing of hydrogels, and a glutaraldehyde solution was tested for the crosslinking process. We were able to obtain two kinds of printable hydrogels with different porosity, swelling and degradation time. Subsequently, the printed hydrogels were characterized from the point of view of biocompatibility. Our results showed that gelatin/xanthan-gum bioprinted hydrogels were biocompatible materials, as they allowed both human keratinocyte and fibroblast in vitro growth for 14 days. These two bioprintable hydrogels could be also used as a helpful dressing material

    Comparison between Fractal and Statistical Approaches to Model Size Effects in VHCF

    Size effects concern the anomalous scaling of relevant mechanical properties of materials and structures over a sufficiently wide dimensional range. In the last few years, thanks to technological advances, such effects have been experimentally detected also in the very high cycle fatigue (VHCF) tests. Research groups at Politecnico di Torino are very active in this field, observing size effects on fatigue strength, fatigue life and fatigue limit up to the VHCF regime for different metal alloys. In addition, different theoretical models have been put forward to explain these effects. In the present paper, two of them are introduced, respectively based on fractal geometry and statistical concepts. Furthermore, a comparison between the models and experimental results is provided. Both models are able to predict the decrement in the fatigue life and in the conventional fatigue limit

    Sulphur Compounds: Comparison of Different Sorbent Tubes for their Detection

    Different techniques have been developed for the analysis of gaseous sulphur pollutants, to maximize the analytical signals. In a complex matrix, such as odorous emissions, the detection of sulphur compounds can be critical in GC analysis, due to the lower concentration of these pollutants and the disturbing effect of co-eluting hydrocarbons. However, their detection is fundamental because they have a non-negligible odour impact. In the field of gaseous emissions analytics, it is common to use sorbent tubes for the sampling step. This technology uses different adsorbent materials, with different selectivity depending on the nature of the gas to be analysed. This work aims to evaluate the ability of three different sorbent tubes to collect different sulphur compounds, belonging to the classes of mercaptans, thioethers and aromatic heterocyclic compounds. A standard solution of 10 sulphur compounds was prepared by diluting in methanol 50 µL of each liquid standard into a 10 mL flask. Subsequently, this solution was diluted in methanol to obtain sulphur standards at five different concentrations (approximately 5-500 ng/µL). The tubes were loaded with the standard solutions with an aliquot of each solution, using a gas chromatograph packed column injector and subsequently analysed by TD-GC-MS. By the results obtained - average Response Factor (RF) and its Relative Standard Deviation (% RSD), it is possible to conduct a comparison among these tubes and evaluate their performance. From the comparison of the tubes, discussing % RSD, it is possible to highlight a slightly better performance, in terms of the number of compounds with % RSD ≤ 30%, for tubes specific for sulphur compounds. Focusing on RF values, multi-sorbent bed tubes show slightly higher RFs for very volatile sulphur compounds, but Sulphur tubes present higher RF values for 6 compounds out of 10 compounds considered. The performance of Tenax TA tubes, instead, appears strictly correlated with the compound’s volatility and therefore they don’t appear useful for sorption of very volatile compounds

    Safety of vedolizumab in liver transplant recipients : a systematic review

    Background: The management of inflammatory bowel disease in patients who have previously undergone liver transplantation can be a clinical challenge. There are serious concerns among physicians regarding the use of biologics for treating such immuno-compromised patients. Objective: We performed a systematic review on vedolizumab therapy in transplant recipients to assess its safety. Methods: PubMed/Embase/Scopus were searched up to November 2018 to identify papers regarding liver transplant recipients and therapy with vedolizumab. Primary outcomes were adverse events. Secondary outcomes were liver transplant and inflammatory bowel disease outcomes. Results: Eight studies (31 patients) were included. Nine out of 31 patients experienced an infection within a mean follow-up time ranging from 5\u201320 months. No malignancies were reported. Inflammatory bowel disease clinical response was experienced by 20/26 patients. Abnormalities in liver tests were recorded in 2/22 patients. Conclusion: Vedolizumab may be considered safe for treating inflammatory bowel disease in liver transplant recipients. Caution is recommended for patients with an unstable liver graft function

    Microstructural Visual Pathway White Matter Alterations in Primary Open-Angle Glaucoma:A Neurite Orientation Dispersion and Density Imaging Study

    BACKGROUND AND PURPOSE: DTI studies of patients with primary open-angle glaucoma have demonstrated that glaucomatous degeneration is not confined to the retina but involves the entire visual pathway. Due to the lack of direct biologic interpretation of DTI parameters, the structural nature of this degeneration is still poorly understood. We used neurite orientation dispersion and density imaging (NODDI) to characterize the microstructural changes in the pregeniculate optic tracts and the postgeniculate optic radiations of patients with primary open-angle glaucoma, to better understand the mechanisms underlying these changes.& nbsp;MATERIALS AND METHODS: T1- and multishell diffusion-weighted scans were obtained from 23 patients with primary open-angle glaucoma and 29 controls. NODDI parametric maps were produced from the diffusion-weighted scans, and probabilistic tractography was used to track the optic tracts and optic radiations. NODDI parameters were computed for the tracked pathways, and the measures were compared between both groups. The retinal nerve fiber layer thickness and visual field loss were assessed for the patients with glaucoma.& nbsp;RESULTS: The optic tracts of the patients with glaucoma showed a higher orientation dispersion index and a lower neurite density index compared with the controls (
