Two Case Studies in Self-Determination: The Rock and the Bailiwick
After looking at the concept of self-determination, its history, meaning, and possible future development in Part II, this Paper will develop two case studies. Part III examines the right of self-determination for the people of Gibraltar, analyzing the relevant U.N. resolutions, agreements, treaties, and legislation that have defined the dispute between Great Britain and Spain. For example, Great Britain has ruled the Rock of Gibraltar for 280 years, primarily using it as a military base; but, today, Spain insists that it did not relinquish absolute sovereignty over Gibraltar to the British by the Treaty of Utrecht in 1713. Part IV examines the situation of the Channel Islands, specifically the Bailiwicks of Guernsey and Jersey, which are not strictly speaking, part of the United Kingdom, but are in appenage , or possessed by the British Crown. The Channel Islands came into Crown possession during the Norman Conquest of 1066 when they formed part of the Duchy of Normandy. While being culturally distinct from the United Kingdom, the Channel Islands are governed under British rule. Finally, Part V will conclude whether the case studies fall within the scope of self-determination, as currently understood by the world\u27s legal community
Usual energy and macronutrient intakes in 2-9-year-old European children
OBJECTIVE: Valid estimates of population intakes are essential for monitoring trends as well as for nutritional interventions, but such data are rare in young children. In particular, the problem of misreporting in dietary data is usually not accounted for. Therefore, this study aims to provide accurate estimates of intake distributions in European children.
DESIGN: Cross-sectional setting-based multi-centre study.
SUBJECTS: A total of 9560 children aged 2-9 years from eight European countries with at least one 24-h dietary recall (24-HDR).
METHODS: The 24-HDRs were classified in three reporting groups based on age- and sex-specific Goldberg cutoffs (underreports, plausible reports, overreports). Only plausible reports were considered in the final analysis (N=8611 children). The National Cancer Institute (NCI)-Method was applied to estimate population distributions of usual intakes correcting for the variance inflation in short-term dietary data.
RESULTS: The prevalence of underreporting (9.5%) was higher compared with overreporting (3.4%). Exclusion of misreports resulted in a shift of the energy and absolute macronutrient intake distributions to the right, and further led to the exclusion of extreme values, that is, mean values and lower percentiles increased, whereas upper percentiles decreased. The distributions of relative macronutrient intakes (% energy intake from fat/carbohydrates/proteins) remained almost unchanged when excluding misreports. Application of the NCI-Method resulted in markedly narrower intake distributions compared with estimates based on single 24-HDRs. Mean percentages of usual energy intake from fat, carbohydrates and proteins were 32.2, 52.1 and 15.7%, respectively, suggesting the majority of European children are complying with common macronutrient intake recommendations. In contrast, total water intake (mean: 1216.7 ml per day) lay below the recommended value for >90% of the children.
CONCLUSION: This study provides recent estimates of intake distributions of European children correcting for misreporting as well as for the daily variation in dietary data. These data may help to assess the adequacy of young children's diets in Europe
ARHGEF9 regulates melanoma morphogenesis in environments with diverse geometry and elasticity by promoting filopodial-driven adhesion.
Rho GTP Exchange Factors (RhoGEFs) and Rho GTPase Activating Proteins (RhoGAPs) are large families of molecules that regulate shape determination in all eukaryotes. In pathologies such as melanoma, RhoGEF and RhoGAP activity underpins the ability of cells to invade tissues of varying elasticity. To identify RhoGEFs and RhoGAPs that regulate melanoma cell shape on soft and/or stiff materials, we performed genetic screens, in tandem with single-cell quantitative morphological analysis. We show that ARHGEF9/Collybistin (Cb) is essential for cell shape determination on both soft and stiff materials, and in cells embedded in 3D soft hydrogel. ARHGEF9 is required for melanoma cells to invade 3D matrices. Depletion of ARHGEF9 results in loss of tension at focal adhesions decreased cell-wide contractility, and the inability to stabilize protrusions. Taken together we show that ARHGEF9 promotes the formation of actin-rich filopodia, which serves to establish and stabilize adhesions and determine melanoma cell shape
Snow accumulation, albedo and melt patterns following road construction on permafrost, Inuvik–Tuktoyaktuk Highway, Canada
Roads constructed on permafrost can have a significant impact on the surrounding environment, potentially inducing permafrost degradation. These impacts arise from factors such as snow accumulation near the road, which affects the soil's thermal and hydrological regime, and road dust that decreases the snow's albedo, altering the timing of snowmelt. However, our current understanding of the magnitude and the spatial extent of these effects is limited. In this study we addressed this gap by using remote sensing techniques to assess the spatial effect of the Inuvik to Tuktoyaktuk Highway (ITH) in Northwest Territories, Canada, on snow accumulation, snow albedo and snowmelt patterns. With a new, high resolution snow depth raster from airborne laser scanning, we quantified the snow accumulation at road segments in the Trail Valley Creek area using digital elevation model differencing. We found increased snow accumulation up to 36 m from the road center. The magnitude of this snow accumulation was influenced by the prevailing wind direction and the embankment height. Furthermore, by analyzing 43 Sentinel-2 satellite images between February and May 2020, we observed reduced snow albedo values within 500 m of the road, resulting in a 12-days-earlier onset of snowmelt within 100 m from the road. We examined snowmelt patterns before, during and after the road construction using the normalized difference snow index from Landsat-7 and Landsat-8 imagery. Our analysis revealed that the road affected the snowmelt pattern up to 600 m from the road, even in areas which appeared undisturbed. In summary, our study improves our understanding of the spatial impact of gravel roads on permafrost due to enhanced snow accumulation, reduced snow albedo and earlier snowmelt. Our study underscores the important contribution that remote sensing can provide to improve our understanding of the effects of infrastructure development on permafrost environments.Peer Reviewe
Depletion of RIPK3 or MLKL blocks TNF-driven necroptosis and switches towards a delayed RIPK1 kinase-dependent apoptosis
In human cells, the RIPK1-RIPK3-MLKL-PGAM5-Drp1 axis drives tumor necrosis factor (TNF)-induced necroptosis through mitochondrial fission, but whether this pathway is conserved among mammals is not known. To answer this question, we analyzed the presence and functionality of the reported necroptotic axis in mice. As in humans, knockdown of receptorinteracting kinase-3 (RIPK3) or mixed lineage kinase domain like (MLKL) blocks TNF-induced necroptosis in L929 fibrosarcoma cells. However, repression of either of these proteins did not protect the cells from death, but instead induced a switch from TNF-induced necroptosis to receptor-interacting kinase-1 (RIPK1) kinase-dependent apoptosis. In addition, although mitochondrial fission also occurs during TNF-induced necroptosis in L929 cells, we found that knockdown of phosphoglycerate mutase 5 (PGAM5) and dynamin 1 like protein (Drp1) did not markedly protect the cells from TNF-induced necroptosis. Depletion of Pink1, a reported interactor of both PGAM5 and Drp1, did not affect TNF-induced necroptosis. These results indicate that in these murine cells mitochondrial fission and Pink1 dependent processes, including Pink-Parkin dependent mitophagy, apparently do not promote necroptosis. Our data demonstrate that the core components of the necrosome (RIPK1, RIPK3 and MLKL) are crucial to induce TNF-dependent necroptosis both in human and in mouse cells, but the associated mechanisms may differ between the two species or cell types
