    Induction of Cell Cycle and NK Cell Responses by Live-Attenuated Oral Vaccines against Typhoid Fever

    The mechanisms by which oral, live-attenuated vaccines protect against typhoid fever are poorly understood. Here, we analyze transcriptional responses after vaccination with Ty21a or vaccine candidate, M01ZH09. Alterations in response profiles were related to vaccine-induced immune responses and subsequent outcome after wild-type Salmonella Typhi challenge. Despite broad genetic similarity, we detected differences in transcriptional responses to each vaccine. Seven days after M01ZH09 vaccination, marked cell cycle activation was identified and associated with humoral immunogenicity. By contrast, vaccination with Ty21a was associated with NK cell activity and validated in peripheral blood mononuclear cell stimulation assays confirming superior induction of an NK cell response. Moreover, transcriptional signatures of amino acid metabolism in Ty21a recipients were associated with protection against infection, including increased incubation time and decreased severity. Our data provide detailed insight into molecular immune responses to typhoid vaccines, which could aid the rational design of improved oral, live-attenuated vaccines against enteric pathogens

    Hadronic Mass Moments in Inclusive Semileptonic B Meson Decays

    We have measured the first and second moments of the hadronic mass-squared distribution in B -> X_c l nu, for P(lepton) > 1.5 GeV/c. We find <M_X^2 - M_D[Bar]^2> = 0.251 +- 0.066 GeV^2, )^2 > = 0.576 +- 0.170 GeV^4, where M_D[Bar] is the spin-averaged D meson mass. From that first moment and the first moment of the photon energy spectrum in b -> s gamma, we find the HQET parameter lambda_1 (MS[Bar], to order 1/M^3 and beta_0 alpha_s^2) to be -0.24 +- 0.11 GeV^2. Using these first moments and the B semileptonic width, and assuming parton-hadron duality, we obtain |V_cb| = 0.0404 +- 0.0013.Comment: 11 pages postscript, also available through http://w4.lns.cornell.edu/public/CLNS, submitted to PR

    Observation of the Ωc0\Omega_{c}^{0} Charmed Baryon at CLEO

    The CLEO experiment at the CESR collider has used 13.7 fb−1^{-1} of data to search for the production of the Ωc0\Omega_c^0 (css-ground state) in e+e−e^{+}e^{-} collisions at s≃10.6\sqrt{s} \simeq 10.6 {\rm GeV}. The modes used to study the Ωc0\Omega_c^0 are Ω−π+\Omega^- \pi^+, Ω−π+π0\Omega^- \pi^+ \pi^0, Ξ−K−pi+π+\Xi^- K^- pi^+ \pi^+, Ξ0K−pi+\Xi^0 K^- pi^+, and Ω−π+π−π+\Omega^- \pi^+ \pi^- \pi^+. We observe a signal of 40.4±\pm9.0(stat) events at a mass of 2694.6±\pm2.6(stat)±\pm1.9(syst) {\rm MeV/c2c^2}, for all modes combined.Comment: 10 pages postscript, also available through http://w4.lns.cornell.edu/public/CLN

    Evidence of New States Decaying into Ξc′π\Xi^{\prime}_{c}\pi

    Using 13.7 fb−1fb^{-1} of data recorded by the CLEO detector at CESR, we report evidence for two new charmed baryons: one decaying into Ξc0′π+\Xi_c^{0 \prime}\pi^+ with the subsequent decay Ξc0′→Ξc0γ\Xi_c^{0 \prime} \to \Xi_c^0 \gamma, and its isospin partner decaying into Ξc+′π−\Xi_c^{+ \prime} \pi^- followed by Ξc+′→Ξc+γ\Xi_c^{+\prime} \to \Xi_c^+\gamma. We measure the following mass differences for the two states: M(Ξc0γπ+)−M(Ξc0)M(\Xi_c^0 \gamma \pi^+)-M(\Xi_c^0)=318.2+-1.3+-2.9 MeV, and M(Ξc+γπ−)−M(Ξc+)M(\Xi_c^+ \gamma \pi^-)-M(\Xi_c^+)=324.0+-1.3+-3.0 MeV. We interpret these new states as the JP=1/2−Ξc1J^P = 1/2^- \Xi_{c1} particles, the charmed-strange analogs of the Λc1+(2593)\Lambda_{c1}^+(2593).Comment: 10 pages postscript, also available through http://w4.lns.cornell.edu/public/CLN

    Measurement of the Relative Branching Fraction of Υ(4S)\Upsilon(4S) to Charged and Neutral B-Meson Pairs

    We analyze 9.7 x 10^6 B\bar{B}$ pairs recorded with the CLEO detector to determine the production ratio of charged to neutral B-meson pairs produced at the Y(4S) resonance. We measure the rates for B^0 -> J/psi K^{(*)0} and B^+ -> J/psi K^{(*)+} decays and use the world-average B-meson lifetime ratio to extract the relative widths f+-/f00 = Gamma(Y(4S) -> B+B-)/Gamma(Y(4S) -> B0\bar{B0}) = = 1.04 +/- 0.07(stat) +/- 0.04(syst). With the assumption that f+- + f00 = 1, we obtain f00 = 0.49 +/- 0.02(stat) +/- 0.01(syst) and f+- = 0.51 +/- 0.02(stat) +/- 0.01(syst).     First Observation of the Decays B0→D∗−ppˉπ+B^{0}\to D^{*-}p\bar{p}\pi^{+} and B^{0}\to D^{*-}p\bar{n}$

    We report the first observation of exclusive decays of the type B to D^* N anti-N X, where N is a nucleon. Using a sample of 9.7 times 10^{6} B-Bbar pairs collected with the CLEO detector operating at the Cornell Electron Storage Ring, we measure the branching fractions B(B^0 \to D^{*-} proton antiproton \pi^+) = ({6.5}^{+1.3}_{-1.2} +- 1.0) \times 10^{-4} and B(B^0 \to D^{*-} proton antineutron) = ({14.5}^{+3.4}_{-3.0} +- 2.7) times 10^{-4}.     A Search for B→τνB\to \tau\nu

    We report results of a search for B→τνB\to\tau\nu in a sample of 9.7 million charged BB meson decays. The search uses both πν\pi\nu and ℓννˉ\ell\nu\bar\nu decay modes of the τ\tau, and demands exclusive reconstruction of the companion Bˉ\bar B decay to suppress background. We set an upper limit on the branching fraction B(B→τν)<8.4×10−4{\cal B}(B\to \tau\nu) < 8.4\times 10^{-4} at 90% confidence level. With slight modification to the analysis we also establish B(B±→K±ννˉ)<2.4×10−4{\cal B}(B^\pm\to K^\pm\nu\bar\nu) < 2.4\times 10^{-4} at 90% confidence level.Comment: 10 ages postscript, also available through http://w4.lns.cornell.edu/public/CLN

    The exchange activities of [Fe] hydrogenase (iron–sulfur-cluster-free hydrogenase) from methanogenic archaea in comparison with the exchange activities of [FeFe] and [NiFe] hydrogenases

    [Fe] hydrogenase (iron–sulfur-cluster-free hydrogenase) catalyzes the reversible reduction of methenyltetrahydromethanopterin (methenyl-H4MPT+) with H2 to methylene-H4MPT, a reaction involved in methanogenesis from H2 and CO2 in many methanogenic archaea. The enzyme harbors an iron-containing cofactor, in which a low-spin iron is complexed by a pyridone, two CO and a cysteine sulfur. [Fe] hydrogenase is thus similar to [NiFe] and [FeFe] hydrogenases, in which a low-spin iron carbonyl complex, albeit in a dinuclear metal center, is also involved in H2 activation. Like the [NiFe] and [FeFe] hydrogenases, [Fe] hydrogenase catalyzes an active exchange of H2 with protons of water; however, this activity is dependent on the presence of the hydride-accepting methenyl-H4MPT+. In its absence the exchange activity is only 0.01% of that in its presence. The residual activity has been attributed to the presence of traces of methenyl-H4MPT+ in the enzyme preparations, but it could also reflect a weak binding of H2 to the iron in the absence of methenyl-H4MPT+. To test this we reinvestigated the exchange activity with [Fe] hydrogenase reconstituted from apoprotein heterologously produced in Escherichia coli and highly purified iron-containing cofactor and found that in the absence of added methenyl-H4MPT+ the exchange activity was below the detection limit of the tritium method employed (0.1 nmol min−1 mg−1). The finding reiterates that for H2 activation by [Fe] hydrogenase the presence of the hydride-accepting methenyl-H4MPT+ is essentially required. This differentiates [Fe] hydrogenase from [FeFe] and [NiFe] hydrogenases, which actively catalyze H2/H2O exchange in the absence of exogenous electron acceptors

    Highly Frequent Mutations in Negative Regulators of Multiple Virulence Genes in Group A Streptococcal Toxic Shock Syndrome Isolates

    Streptococcal toxic shock syndrome (STSS) is a severe invasive infection characterized by the sudden onset of shock and multiorgan failure; it has a high mortality rate. Although a number of studies have attempted to determine the crucial factors behind the onset of STSS, the responsible genes in group A Streptococcus have not been clarified. We previously reported that mutations of csrS/csrR genes, a two-component negative regulator system for multiple virulence genes of Streptococcus pyogenes, are found among the isolates from STSS patients. In the present study, mutations of another negative regulator, rgg, were also found in clinical isolates of STSS patients. The rgg mutants from STSS clinical isolates enhanced lethality and impaired various organs in the mouse models, similar to the csrS mutants, and precluded their being killed by human neutrophils, mainly due to an overproduction of SLO. When we assessed the mutation frequency of csrS, csrR, and rgg genes among S. pyogenes isolates from STSS (164 isolates) and non-invasive infections (59 isolates), 57.3% of the STSS isolates had mutations of one or more genes among three genes, while isolates from patients with non-invasive disease had significantly fewer mutations in these genes (1.7%). The results of the present study suggest that mutations in the negative regulators csrS/csrR and rgg of S. pyogenes are crucial factors in the pathogenesis of STSS, as they lead to the overproduction of multiple virulence factors

    Two-body B Meson Decays to eta and eta': Observation of B->eta K*

    In a sample of 19 million produced B mesons, we have observed the decays B -> eta K* and improved our previous measurements of B -> eta'K. The branching fractions we measure for these decay modes are BR(B+ -> eta K*+) = (26.4 +9.6-8.2 +- 3.3) x 10−610^{-6}, BR(B0 -> eta K*0) = (13.8 +5.5-4.6 +- 1.6) x 10−610^{-6}, BR(B+ -> eta' K+) = (80 +10-9 +- 7) x 10−610^{-6} and BR(B0 -> eta' K0) = (89 +18-16 +- 9) x 10−610^{-6}. We have searched with comparable sensitivity for related decays and report upper limits for these branching fractions.Comment: 12 page postscript file, 2 figure
