    Effect of Cypermethrin on some hematological parameters and prediction of their recovery in a freshwater Teleost, Cyprinus carpio

    The present investigation was conducted to evaluate the lethality of cypermethrin “pyrethroid” (25% EC with 100% purity) on common carp, Cyprinus carpio (fingerlings). The effect was assessed on the basis of impact of short term exposures of its below safe concentrations (1/2 (0.05 μl/L) and 1/10 (0.01 μl/L) parts of safe concentration) on some hematological parameters. The sub-lethal exposure studies were done on the same day and after 7 days of exposure. Hematological recovery was also studied by maintaining the pesticide-exposed fish in a fresh water system for an additional 7 days. The hematological analysis showed significant reduction in red blood cells (RBCs) count and hemoglobin (Hb) value while total white blood cells (WBCs) count were significantly increased in the fish C. carpio. Complete recovery was obtained in all the parameters after a recovery period of 7 days.Key words: Cypermethrin, Cyprinus carpio, hematology, recover

    Genetic diversity analysis of Labeo gonius (Hamilton, 1822) in three different reservoirs of Uttarakhand by using allozyme marker

    The present population genetic study was carried out to identify basic genetic structure of Labeo gonius from three different sized reservoirs viz., Dhaura, Baigul and Nanak Sagar reservoir of Uttarakhand by applying allozyme markers. The allozyme analysis was conducted for 18 enzymes but only 10 enzymes showed their presence with scorable activity. All ten allozymes yielded overall 19 scorable loci, out of which 11 loci were found to be polymorphic in the population with a percentage of 57.89. Genetic diversity parameters, that is number of polymorphic loci, the mean number of alleles per locus, observed (HO) and expected heterozygosity (He), fixation index (FIS) and genic differentiation were estimated by using software GENEPOP Ver.3.4 indicating high level of genetic diversity in stocks of all three reservoirs of L. gonius being more heterozygous in Nanak sagar followed by Baigul and Dhaura reservoirs. FST for overall populations ranging from 0.01 to 0.18 indicate moderate genetic differentiation as per Wright’s criteria. The allozyme analysis in L. gonius showed that these populations are moderately differentiated from each other and likely to be associated with small gene exchange which is responsible for weak sub-structuring of stocks in the three reservoirs.Keywords: Allozyme, Labeo gonius, genetic diversity, population geneticsAfrican Journal of Biotechnology Vol. 12(19), pp. 2532-253

    Structured Multi-Label Biomedical Text Tagging via Attentive Neural Tree Decoding

    We propose a model for tagging unstructured texts with an arbitrary number of terms drawn from a tree-structured vocabulary (i.e., an ontology). We treat this as a special case of sequence-to-sequence learning in which the decoder begins at the root node of an ontological tree and recursively elects to expand child nodes as a function of the input text, the current node, and the latent decoder state. We demonstrate that this method yields state-of-the-art results on the important task of assigning MeSH terms to biomedical abstracts

    Hansenula polymorpha Pex37 is a peroxisomal membrane protein required for organelle fission and segregation

    Here, we describe a novel peroxin, Pex37, in the yeast Hansenula polymorpha. H. polymorpha Pex37 is a peroxisomal membrane protein, which belongs to a protein family that includes, among others, the Neurospora crassa Woronin body protein Wsc, the human peroxisomal membrane protein PXMP2, the Saccharomyces cerevisiae mitochondrial inner membrane protein Sym1, and its mammalian homologue MPV17. We show that deletion of H. polymorpha PEX37 does not appear to have a significant effect on peroxisome biogenesis or proliferation in cells grown at peroxisome‐inducing growth conditions (methanol). However, the absence of Pex37 results in a reduction in peroxisome numbers and a defect in peroxisome segregation in cells grown at peroxisome‐repressing conditions (glucose). Conversely, overproduction of Pex37 in glucose‐grown cells results in an increase in peroxisome numbers in conjunction with a decrease in their size. The increase in numbers in PEX37‐overexpressing cells depends on the dynamin‐related protein Dnm1. Together our data suggest that Pex37 is involved in peroxisome fission in glucose‐grown cells. Introduction of human PXMP2 in H. polymorpha pex37 cells partially restored the peroxisomal phenotype, indicating that PXMP2 represents a functional homologue of Pex37. H.polymorpha pex37 cells did not show aberrant growth on any of the tested carbon and nitrogen sources that are metabolized by peroxisomal enzymes, suggesting that Pex37 may not fulfill an essential function in transport of these substrates or compounds required for their metabolism across the peroxisomal membrane

    A Neural Candidate-Selector Architecture for Automatic Structured Clinical Text Annotation

    We consider the task of automatically annotating free texts describing clinical trials with concepts from a controlled, structured medical vocabulary. Specifically, we aim to build a model to infer distinct sets of (ontological) concepts describing complementary clinically salient aspects of the underlying trials: the populations enrolled, the interventions administered and the outcomes measured, i.e., the PICO elements. This important practical problem poses a few key challenges. One issue is that the output space is vast, because the vocabulary comprises many unique concepts. Compounding this problem, annotated data in this domain is expensive to collect and hence sparse. Furthermore, the outputs (sets of concepts for each PICO element) are correlated: specific populations (e.g., diabetics) will render certain intervention concepts likely (insulin therapy) while effectively precluding others (radiation therapy). Such correlations should be exploited. We propose a novel neural model that addresses these challenges. We introduce a Candidate-Selector architecture in which the model considers setes of candidate concepts for PICO elements, and assesses their plausibility conditioned on the input text to be annotated. This relies on a 'candidate set' generator, which may be learned or relies on heuristics. A conditional discriminative neural model then jointly selects candidate concepts, given the input text. We compare the predictive performance of our approach to strong baselines, and show that it outperforms them. Finally, we perform a qualitative evaluation of the generated annotations by asking domain experts to assess their quality

    Activation of Aryl Hydrocarbon Receptor (AhR) Leads to Reciprocal Epigenetic Regulation of FoxP3 and IL-17 Expression and Amelioration of Experimental Colitis

    Aryl hydrocarbon receptor (AhR), a transcription factor of the bHLH/PAS family, is well characterized to regulate the biochemical and toxic effects of environmental chemicals. More recently, AhR activation has been shown to regulate the differentiation of Foxp3(+) Tregs as well as Th17 cells. However, the precise mechanisms are unclear. In the current study, we investigated the effect of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), a potent AhR ligand, on epigenetic regulation leading to altered Treg/Th17 differentiation, and consequent suppression of colitis.Dextran sodium sulphate (DSS) administration induced acute colitis in C57BL/6 mice, as shown by significant weight loss, shortening of colon, mucosal ulceration, and increased presence of CXCR3(+) T cells as well as inflammatory cytokines. Interestingly, a single dose of TCDD (25 µg/kg body weight) was able to attenuate all of the clinical and inflammatory markers of colitis. Analysis of T cells in the lamina propria (LP) and mesenteric lymph nodes (MLN), during colitis, revealed decreased presence of Tregs and increased induction of Th17 cells, which was reversed following TCDD treatment. Activation of T cells from AhR(+/+) but not AhR (-/-) mice, in the presence of TCDD, promoted increased differentiation of Tregs while inhibiting Th17 cells. Analysis of MLN or LP cells during colitis revealed increased methylation of CpG islands of Foxp3 and demethylation of IL-17 promoters, which was reversed following TCDD treatment.These studies demonstrate for the first time that AhR activation promotes epigenetic regulation thereby influencing reciprocal differentiation of Tregs and Th17 cells, and amelioration of inflammation

    Psychosocial functioning and intelligence both partly explain socioeconomic inequalities in premature death. A population-based male cohort study

    The possible contributions of psychosocial functioning and intelligence differences to socioeconomic status (SES)-related inequalities in premature death were investigated. None of the previous studies focusing on inequalities in mortality has included measures of both psychosocial functioning and intelligence.The study was based on a cohort of 49 321 men born 1949-1951 from the general community in Sweden. Data on psychosocial functioning and intelligence from military conscription at ∼18 years of age were linked with register data on education, occupational class, and income at 35-39 years of age. Psychosocial functioning was rated by psychologists as a summary measure of differences in level of activity, power of initiative, independence, and emotional stability. Intelligence was measured through a multidimensional test. Causes of death between 40 and 57 years of age were followed in registers.The estimated inequalities in all-cause mortality by education and occupational class were attenuated with 32% (95% confidence interval: 20-45%) and 41% (29-52%) after adjustments for individual psychological differences; both psychosocial functioning and intelligence contributed to account for the inequalities. The inequalities in cardiovascular and injury mortality were attenuated by as much as 51% (24-76%) and 52% (35-68%) after the same adjustments, and the inequalities in alcohol-related mortality were attenuated by up to 33% (8-59%). Less of the inequalities were accounted for when those were measured by level of income, with which intelligence had a weaker correlation. The small SES-related inequalities in cancer mortality were not attenuated by adjustment for intelligence.Differences in psychosocial functioning and intelligence might both contribute to the explanation of observed SES-related inequalities in premature death, but the magnitude of their contributions likely varies with measure of socioeconomic status and cause of death. Both psychosocial functioning and intelligence should be considered in future studies
