    Foliar Architecture and caulinar morpho-anatomy of Zuccagnia Punctata (Fabaceae). Histolocalization of bioactive compounds

    Zuccagnia punctata Cav. (Fabaceae, Caesalpinioideae), comúnmente denominada jarilla macho, es una especie endémica de Argentina, característica de la flora xerófila de la Provincia Biogeográfica del Monte. Resulta una especie de interés medicinal ya que sus partes aéreas, usadas en medicina popular, presentan actividad antioxi- dante, antiulcerosa, antifúngica y antimicrobiana frente a bacterias patógenas humanas mul- tiresistentes. Dado que se registran escasos estudios morfológicos y anatómicos de esta promisoria especie, el objetivo del presente trabajo es realizar la descripción de sus partes aéreas apuntando a una histolocalización de los tejidos encargados de la síntesis de compues- tos bioactivos. Se realizaron técnicas de anatomía vegetal y pruebas histoquímicas. Z. punc- tata presenta hojas pseudo-paripinnadas, folíolos nanófilos subopuestos, acuminados, de base redondeada y márgenes enteros. Venación de tipo pinnada camptódroma broquidódroma con vena primaria masiva, estructura anatómica claramente adaptada a ambientes xerófitos, fo- líolo isolateral, anfistomático, con aparato estomático ciclocítico, células epidérmicas de pa- redes rectas, cutícula gruesa, tricomas glandulares capitados ubicados en criptas de ambas superficies epidérmicas y tricomas eglandulares unicelulares o bicelulares uniseriados articu- lados en el margen foliar. El nervio medio está formado por un único haz colateral cerrado con casquete de esclerénquima a nivel de floema. El tallo presenta una eustela con fuerte de- sarrollo de fibras durante el crecimiento secundario. Tanto en tallo como en hoja se observan idioblastos con drusas a nivel del mesofilo y floema. La tinción específica con vainillín sulfúrico revela que tanto el mesofilo como los tricomas glandulares se hallan comprometidos en la producción de compuestos fenólicos y terpénicos.Fil: Mercado, Maria Ines.     Effects of stimulus duration on audio-visual synchrony perception

    The integration of visual and auditory inputs in the human brain occurs only if the components are perceived in temporal proximity, that is, when the intermodal time difference falls within the so-called subjective synchrony range. We used the midpoint of this range to estimate the point of subjective simultaneity (PSS). We measured the PSS for audio-visual (AV) stimuli in a synchrony judgment task, in which subjects had to judge a given AV stimulus using three response categories (audio first, synchronous, video first). The relevant stimulus manipulation was the duration of the auditory and visual components. Results for unimodal auditory and visual stimuli have shown that the perceived onset shifts to relatively later positions with increasing stimulus duration. These unimodal shifts should be reflected in changing PSS values, when AV stimuli with different durations of the auditory and visual components are used. The results for 17 subjects showed indeed a significant shift of the PSS for different duration combinations of the stimulus components. Because the shifts were approximately equal for duration changes in either of the components, no net shift of the PSS was observed as long as the durations of the two components were equal. This result indicates the need to appropriately account for unimodal timing effects when quantifying intermodal synchrony perceptio

    Evaluación de parámetros nutricionales y sensoriales durante la conservación de pulpa de "chilto"

    Desde hace unos años y dentro de un programa de Bioprospección de plantas nativas alimenticias y medicinales del NOA, nuestro instituto (INBIOFIV) estudia las propiedades nutricionales y funcionales de frutos de “chilto” (Solanum betaceum). Este es un fruto carnoso, comestible y nativo de las Yungas del NOA del cual se encuentran 3 variedades principales que se diferencian por el color del epicarpio (naranja o bola de chivo, roja y sangre de buey). En estudios previos hemos descripto las propiedades nutricionales y algunas potencialidades funcionales de los frutos bola de chivo y sangre de buey. En el marco de un proyecto premiado por ARCOR comenzamos a analizar productos derivados del mismo y su conservación en el tiempo. El objetivo de este trabajo fue analizar la conservación de la pulpa con y sin semilla de las variedades bola de chivo y sangre de buey en el tiempo, para lo que evaluamos parámetros nutricionales, físicos (color, pH) y sensoriales durante tres meses de conservación en frio (-20°C). El procesamiento de los frutos frescos permitió obtener pulpa libre de semillas o pulpa con semillas. Los resultados obtenidos demuestran la estabilidad nutricional (lípidos, proteínas y fibras), y en propiedades organolépticas como el color (luminosidad, parámetros de color) de la pulpa refrigerada. La estabilidad de la pulpa de “chilto” permitiría su utilización en la industria alimenticia para la fabricación de jugos, cocteles, dulces, entre otros.Fil: Orqueda, Maria Eugenia.     Multiple sulfatase deficiency with neonatal manifestation.

    Multiple Sulfatase Deficiency (MSD; OMIM 272200) is a rare autosomal recessive inborn error of metabolism caused by mutations in the sulfatase modifying factor 1 gene, encoding the formylglycine-generating enzyme (FGE), and resulting in tissue accumulation of sulfatides, sulphated glycosaminoglycans, sphingolipids and steroid sulfates. Less than 50 cases have been published so far. We report a new case of MSD presenting in the newborn period with hypotonia, apnoea, cyanosis and rolling eyes, hepato-splenomegaly and deafness. This patient was compound heterozygous for two so far undescribed SUMF1 mutations (c.191C > A; p.S64X and c.818A > G; p.D273G)

    The lncRNA CASC15 regulates SOX4 expression in RUNX1-rearranged acute leukemia

    Abstract Background Long non-coding RNAs (lncRNAs) play a variety of cellular roles, including regulation of transcription and translation, leading to alterations in gene expression. Some lncRNAs modulate the expression of chromosomally adjacent genes. Here, we assess the roles of the lncRNA CASC15 in regulation of a chromosomally nearby gene, SOX4, and its function in RUNX1/AML translocated leukemia. Results CASC15 is a conserved lncRNA that was upregulated in pediatric B-acute lymphoblastic leukemia (B-ALL) with t (12; 21) as well as pediatric acute myeloid leukemia (AML) with t (8; 21), both of which are associated with relatively better prognosis. Enforced expression of CASC15 led to a myeloid bias in development, and overall, decreased engraftment and colony formation. At the cellular level, CASC15 regulated cellular survival, proliferation, and the expression of its chromosomally adjacent gene, SOX4. Differentially regulated genes following CASC15 knockdown were enriched for predicted transcriptional targets of the Yin and Yang-1 (YY1) transcription factor. Interestingly, we found that CASC15 enhances YY1-mediated regulation of the SOX4 promoter. Conclusions Our findings represent the first characterization of this CASC15 in RUNX1-translocated leukemia, and point towards a mechanistic basis for its action

    Evidence and morality in harm-reduction debates: can we use value-neutral arguments to achieve value-driven goals?

    It is common to argue that politicians make selective use of evidence to tacitly reinforce their moral positions, but all stakeholders combine facts and values to produce and use research for policy. The drug policy debate has largely been framed in terms of an opposition between evidence and politics. Focusing on harm reduction provides useful ground to discuss a further opposition proposed by evidence advocates, that between evidence and morality. Can evidence sway individuals from their existing moral positions, so as to “neutralise” morality? And if not, then should evidence advocates change the way in which they frame their arguments? To address these questions, analysis of N=27 interviews with stakeholders involved in drug policy and harm reduction research, advocacy, lobbying, implementation and decision-making in England, UK and New South Wales, Australia, was conducted. Participants’ accounts suggest that although evidence can help focus discussions away from values and principles, exposure to evidence does not necessarily change deeply held views. Whether stakeholders decide to go with the evidence or not seems contingent on whether they embrace a view of evidence as secular faith; a view that is shaped by experience, politics, training, and role. And yet, morality, values, and emotions underpin all stakeholders’ views, motivating their commitment to drug policy and harm reduction. Evidence advocates might thus benefit from morally and emotionally engaging audiences. This paper aims to develop better tools for analysing the role of morality in decision-making, starting with moral foundations theory. Using tools from disciplines such as moral psychology is relevant to the study of the politics of evidence-based policymaking

    Developmental Acquisition of a Rapid Calcium-Regulated Vesicle Supply Allows Sustained High Rates of Exocytosis in Auditory Hair Cells

    Auditory hair cells (HCs) have the remarkable property to indefinitely sustain high rates of synaptic vesicle release during ongoing sound stimulation. The mechanisms of vesicle supply that allow such indefatigable exocytosis at the ribbon active zone remain largely unknown. To address this issue, we characterized the kinetics of vesicle recruitment and release in developing chick auditory HCs. Experiments were done using the intact chick basilar papilla from E10 (embryonic day 10) to P2 (two days post-hatch) by monitoring changes in membrane capacitance and Ca2+ currents during various voltage stimulations. Compared to immature pre-hearing HCs (E10-E12), mature post-hearing HCs (E18-P2) can steadily mobilize a larger readily releasable pool (RRP) of vesicles with faster kinetics and higher Ca2+ efficiency. As assessed by varying the inter-pulse interval of a 100 ms paired-pulse depolarization protocol, the kinetics of RRP replenishment were found much faster in mature HCs. Unlike mature HCs, exocytosis in immature HCs showed large depression during repetitive stimulations. Remarkably, when the intracellular concentration of EGTA was raised from 0.5 to 2 mM, the paired-pulse depression level remained unchanged in immature HCs but was drastically increased in mature HCs, indicating that the Ca2+ sensitivity of the vesicle replenishment process increases during maturation. Concomitantly, the immunoreactivity of the calcium sensor otoferlin and the number of ribbons at the HC plasma membrane largely increased, reaching a maximum level at E18-P2. Our results suggest that the efficient Ca2+-dependent vesicle release and supply in mature HCs essentially rely on the concomitant engagement of synaptic ribbons and otoferlin at the plasma membrane

    Effects of Fusion between Tactile and Proprioceptive Inputs on Tactile Perception

    Tactile perception is typically considered the result of cortical interpretation of afferent signals from a network of mechanical sensors underneath the skin. Yet, tactile illusion studies suggest that tactile perception can be elicited without afferent signals from mechanoceptors. Therefore, the extent that tactile perception arises from isomorphic mapping of tactile afferents onto the somatosensory cortex remains controversial. We tested whether isomorphic mapping of tactile afferent fibers onto the cortex leads directly to tactile perception by examining whether it is independent from proprioceptive input by evaluating the impact of different hand postures on the perception of a tactile illusion across fingertips. Using the Cutaneous Rabbit Effect, a well studied illusion evoking the perception that a stimulus occurs at a location where none has been delivered, we found that hand posture has a significant effect on the perception of the illusion across the fingertips. This finding emphasizes that tactile perception arises from integration of perceived mechanical and proprioceptive input and not purely from tactile interaction with the external environment

    The empirical analysis of non-problematic video gaming and cognitive skills: a systematic review

    Videogames have become one of the most popular leisure activities worldwide, including multiple game genres with different characteristics and levels of involvement required. Although a small minority of excessive players suffer detrimental consequences including impairment of several cognitive skills (e.g., inhibition, decision-making), it has also been demonstrated that playing videogames can improve different cognitive skills. Therefore, the current paper systematically reviewed the empirical studies experimentally investigating the positive impact of videogames on cognitive skills. Following a number of inclusion and exclusion criteria, a total of 32 papers were identified as empirically investigating three specific skills: taskswitching (eight studies), attentional control (22 studies), and sub-second time perception (two studies). Results demonstrated that compared to control groups, non-problematic use of videogames can lead to improved task-switching, more effective top-down attentional control and processing speed and increased sub-second time perception. Two studies highlighted the impact of gaming on cognitive skills differs depends upon game genre. The studies reviewed suggest that videogame play can have a positive impact on cognitive processes for players
