    Study of antimicrobial activity and technology optimization of Calendulae flos galenicals

    The article presents materials on optimization of manufacture technology for Calendulae flos galenicals with medium level of antibacterial activity. For antibacterial study of extracts, we used agar well diffusion method. In our research, we utilized six test-strain microorganisms: E. coli ATCC 25922, S. aureus ATCC 25923, P. vulgaris ATCC 4636, P. aeruginosa ATCC 27853, C. albicans ATCC 885/653, and B. subtilis ATCC 6633. We have found that the maximum level of extracts’ antimicrobial activity is achieved in the range of ethanol content in the extractant from 70 to 97 % v/

    A Study on the Dynamics of the Development of Red Steppe Cows and Their Ayrshire and Holstein Crossbreeds

    When purchasing animals from abroad, agricultural enterprises often underestimate their demand for feed quality and feeding standards for ensuring genetically determined milk productivity. Therefore, increasing attention should be paid to raising local breeds. During the creation of new production relations, it is very important to preserve achievements of scientists and breeders. The widespread use of red steppe cows is due to good feed payment, unpretentiousness, and their adaptability to the steppe zone with its arid climate. There was a real threat of extinction of purebred red steppe cattle: with the intensification of dairy cattle breeding, as well as low milk productivity, these animals are inferior to the best dairy breeds by productivity, but they are better by endurance, resistance to diseases, and adaptability to extreme environmental conditions. Therefore, scientific research is needed on improving the productive and technological qualities of this breed using the best global gene pool of dairy cattle. This article presents the results of a study on the development of heifers of purebred red steppe cows and their Ayrshire and red-motley Holstein crossbreeds. Differences in the live weight indicators of the purebred and crossbred heifers were due to the effect of crossbreeding and intensive growing technology, which made it possible to maximize the genetic potential of the crossbred animals.     Using medical science as an example, there are analysed issues and questions regarding an evolution of the Programme for fundamental scientific research of state science academies. It is demonstrated that the current system for managing fundamental sciences is incredibly conservative. It is concluded that problems with shaping key performance indicators for the Programme of fundamental scientific research 2016 and forthcoming years, including its structure, are the result of a separation of scientific resource management from the management of scientific research. Authors of the article confirm the necessity to integrate scientific institutions into one unique system of scientific research and implementation of scientific inventions into healthcare practice. На примере медицинской науки рассмотрены вопросы и проблемы эволюции Программы фундаментальных научных исследований государственных академий наук. Показано, что существующая система управления фундаментальной наукой чрезвычайно консервативна. Делается вывод, что проблемы формирования плановых показателей Программы фундаментальных научных исследований на2016 г. и последующие периоды, включая её структуру, являются результатом отделения управления научными ресурсами от управления научными исследованиями.     New approaches to the study of cell vital activity cultivated in different growing conditions with analysis of oxygen in the medium

    Objectives to develop new approaches to the study of morphofunctional state of chondroblasts, cultured at 37C on a 3D carrier in different environments: in a CO2incubator with 5% of CO2and in a thermostat in an air-proof tube. Material and methods.The study cell culture chondroblasts, isolated from the cartilage of the articular surfaces of the extra-fingers' phalanges. 3D carrier for cells the demineralized lyophilized human spongiosa Lioplast. The resulting tissue-engineered structures were grown in a complete cell culture medium at 37С under different conditions: in a closed system in thermostat and in an open system in CO2incubator (5% CO2). To assess the morphofunctional state of the cells on the surface of the 3D carrier, the picrosirius red staining, a LIVE/DEADfluorescent dye kit, and scanning electron microscopy were used. The oxygen concentration in the culture medium was evaluated by the modified Winkler titration method. Results.The complex of morphological methods revealed the presence of living cells on the surface of human spongiosa within the 7-day period of cultivation. The cells either are fusiform or have a polygonal form and have a capacity to grow in 2 or more layers. The titrimetric analysis has demonstrated a decline in the concentration of dissolved oxygen in the medium with cellular tissue material in 7 days of cultivation. The concentration declined by 72.4% in a thermostat and by 63.5% in a CO2incubator. In the tests tubes which contained only the medium and no cells, there was a similar decline in oxygen concentration by 47.3% in a thermostat and by 66.1% in a CO2incubator. Conclusion.1. A method of measuring the amount of dissolved oxygen in a culture medium, during the adhesive cell cultivation on a 3D carrier, was developed, based on the Winkler titration method. 2. A comparative analysis of the amount of dissolved oxygen in the medium in the process of chondroblast cultivation on a 3D human spongiosa carrier, both in a CO2incubator and in a closed test tube, revealed an overall tendency to a decrease in the concentration of oxygen within 7 days of cultivation. 3. A decrease in oxygen concentration in the test tubes with human spongiosa samples (without cells), within the 7 days of cultivation, was registered. 4. An efficient and cost-saving method of graft manufacturing for the purposes of chondroplasty is the transfer of juvenile joint cartilage chondroblasts to 3D human spongiosa carriers and their further cultivation in air-proof test tubes copletely filled with medium within a period of 7 days

    With the help of enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) in 332 patients with acute feverish illness after a tick bite, treated at the Perm Regional Hospital of Infectious Diseases in the spring-summer period of 2010, the diagnostics of human granulocytic anaplasmosis was performed. By Omnix’s testsystems HGA-ELISA-IgM and HGA-ELISA-IgG 845 serum, as well as by «nested» PCR, using primers ge3a1-ge10r2 and ge9f3-ge2r4 for A. phagocytophilum genetic material amplification, 952 samples of dried blood spots were studied in the dynamics of the disease. The optimal period for the use of ELISA and PCR was determined. Anaplasmosis was confirmed by both methods in 79 patients (ELISA – in 68, PCR –in 25). In general, serodiagnosis, particularly ELISA, and PCR should be regarded not as alternative but as complementary to each other’s methods of laboratory diagnosis of anaplasmosis.Проведена диагностика гранулоцитарного анаплазмоза человека с помощью иммуноферментного анализа (ИФА) и полимеразной цепной реакции (ПЦР) у 332 пациентов, находившихся на лечении в краевой клинической инфекционной больнице г. Перми в весенне-летний период 2010 г. с острыми лихорадочными заболеваниями после присасывания клещей. В динамике болезни исследовано 845 сывороток крови тест-системами ООО «Омникс» ГАЧ-ИФА-IgM и ГАЧ-ИФА-IgG, а также 952 пробы сухих пятен крови методом «nested» ПЦР с использованием праймеров ge3a1-ge10r2 и ge9f3-ge2r4 для амплификации генетического материала A. phagocytophilum. Определены оптимальные сроки использования ИФА и ПЦР. Обоими методами анаплазмоз подтвержден у 79 пациентов (ИФА – у 68, ПЦР – у 25). В целом, серодиагностику, в частности ИФА, и ПЦР следует рассматривать не как альтернативные, а как взаимодополняющие друг друга методы лабораторной диагностики анаплазмоза
