    The changes in the amino and fatty acid profiles in the semifinished foodstuffs based on broiler meat and components of chicken eggs after different types of thermal treatment

    The changes in the amino and fatty acid profiles in the semifinished foodstuffs (SFFs) based on broiler meat and coagulated chicken egg melange after different types of thermal treatment (water or steam boiling, braising, baking, frying) were studied. The amino acid profiles were determined on Knauer analyzer; tryptophan by standard method. The biological value of the treated products was assessed using amino acid balance coefficients calculated by the method of N. N. Lipatov. It was found that the changes in the initial amino acid profiles of the SFFs were the least after water and steam boiling; braising and baking were found to increase the contents of the essential amino acids. The amino acid profiles in the treated SFFs were close to the reference values. The best criteria of their biological value (coefficient of rationality of amino acid composition, comparable redundance) were found after water and steam boiling. It was found that all types of thermal treatments insignificantly affected the parameters of fatty acid balance within the SFFs; the changes found were primarily related to slight increase in total content of saturated fatty acids and increase in total content of polyunsaturated fatty acids (PUFAs) in compare to initial profiles, by 2.64–3.88% depending on the treatment type.     Chicken egg white — characteristics of its properties and the prospects for functional foods development

    The overview presents the literature data and the results of our own research on prospects of using the chicken eggs as the basis of functional foods. The composition of chicken eggs and their components, characteristics of egg white proteins properties are presented thereto. The biologically active compounds included into egg composition are analyzed. The data on the biological value of egg white are given. The characteristic of egg white foaming ability is presented. It has been shown that the ability of proteins to form stable intermolecular structures, especially with partially denaturated proteins, allows them forming viscoelastic superficial films that ensure foam stability. The high foaming ability of chicken egg protein macromolecules is directly related to their interphase properties, i. e. the ability to form interphase layers at the “liquid —  gas” interface. The foaming properties of the various egg proteins are not equal, and therefore they contribute to foaming properties at various extents. The model of egg white proteins gelation is considered and the factors influencing the gelation process are described. It has been shown that very important changes in proteins properties are caused by denaturation. The proteins lose their ability to hydrate; the protective aqueous shell around the globules disappears, the proteins stick together, grow larger and lose solubility. This process is called coagulation. The influence of denaturation and aggregation on variations of protein properties is described below. Data on protein fortification with functional ingredients (calcium, iodine, plant polyphenols) and creation of functional egg and meat foods are presented here

Abstract-Reduction of efficiency of oxidative phosphorylation associated with aging and the development of neurodegenerative diseases including Alzheimer's disease is thought to be linked to the accumulation of deletions in mitochondrial DNA (∆mtDNA), which are seen as a marker of oxidative damage. Recently, we have shown that mitochondria-targeted antioxidant SkQ1 (10-(6′-plastoquinonyl)decyltriphenylphosphonium) can slow the development of signs of Alzheimer's disease in senescence-accelerated OXYS rats. The purpose of this study was to explore the relationship between the development of neurodegenerative changes in the brain of OXYS rats and changes in the amount of mtDNA and the 4834-bp mitochondrial DNA deletion (∆mtDNA 4834 ) as well as the effect of SkQ1. We studied the relative amount of mtDNA and ∆mtDNA 4834 in the hippocampus of OXYS and Wistar (control) rats at ages of 1, 2, 6, 10, and 20 days and 3, 6, and 24 months. During the period crucial for manifestation of the signs of accelerated aging of OXYS rats (from 1.5 to 3 months of age), we evaluated the effects of administration of SkQ1 (250 nmol/kg) and vitamin E (670 mmol/kg, reference treatment) on the amount of mtDNA and ∆mtDNA 4834 and on the formation of the behavioral feature of accelerated senescence in OXYS rats -passive type of behavior in the open field test. In OXYS rats, the level of ∆mtDNA 4834 in the hippocampus is increased compared to the Wistar rats, especially at the stage of completion of brain development in the postnatal period. This level remains elevated not only at the stages preceding the manifestation of the signs of accelerated brain aging and the development of pathological changes linked to Alzheimer's disease, but also during their progression. However, at age of 24 months, there were no detectable differences between the two strains. SkQ1 treatment reduced the level of ∆mtDNA 4834 in the hippocampus of Wistar and OXYS rats and slowed the formation of passive behavior in OXYS rats.     Systemic proteasome inhibition triggers neurodegeneration in a transgenic mouse model expressing human α-synuclein under oligodendrocyte promoter: implications for multiple system atrophy

    Multiple system atrophy (MSA) is a progressive late onset neurodegenerative α-synucleinopathy with unclear pathogenesis. Recent genetic and pathological studies support a central role of α-synuclein (αSYN) in MSA pathogenesis. Oligodendroglial cytoplasmic inclusions of fibrillar αSYN and dysfunction of the ubiquitin–proteasome system are suggestive of proteolytic stress in this disorder. To address the possible pathogenic role of oligodendroglial αSYN accumulation and proteolytic failure in MSA we applied systemic proteasome inhibition (PSI) in transgenic mice with oligodendroglial human αSYN expression and determined the presence of MSA-like neurodegeneration in this model as compared to wild-type mice. PSI induced open field motor disability in transgenic αSYN mice but not in wild-type mice. The motor phenotype corresponded to progressive and selective neuronal loss in the striatonigral and olivopontocerebellar systems of PSI-treated transgenic αSYN mice. In contrast no neurodegeneration was detected in PSI-treated wild-type controls. PSI treatment of transgenic αSYN mice was associated with significant ultrastructural alterations including accumulation of fibrillar human αSYN in the cytoplasm of oligodendroglia, which resulted in myelin disruption and demyelination characterized by increased g-ratio. The oligodendroglial and myelin pathology was accompanied by axonal degeneration evidenced by signs of mitochondrial stress and dysfunctional axonal transport in the affected neurites. In summary, we provide new evidence supporting a primary role of proteolytic failure and suggesting a neurodegenerative pathomechanism related to disturbed oligodendroglial/myelin trophic support in the pathogenesis of MSA

    ВЛИЯНИЕ ТЕПЛОВОГО НАГРЕВА НА ЖИРНОКИСЛОТНЫЙ СОСТАВ МЯСА ИНДЕЙКИ, ОБОГАЩЕННОГО ЛЬНЯНЫМ МАСЛОМ

    Abstract The paper examines the problem of optimization of the fatty acid composition of lipids in poultry meat, which is widely used in nutrition. The omega-6 content is significantly higher than the omega-3 content in the composition of poultry meat lipids, which is not optimal for assimilation and needs a correction. The possibility of turkey meat enrichment with linseed oil was investigated with the aim of ensuring the omega-6 to omega-3 ratio in the minced meat formulations, which provides for the nutritionally adequate balance not higher than 10 units. The paper also presents the results of the investigation of the fatty acids composition and fatty acid balance of the lipid fraction of minced meat as well as the changes due to thermal heating of meat formulations in the water medium with a temperature of 95±2  °C to a product core temperature of 70±1  °C. According to the data of the investigations, the omega-6 : omega-3 ratio in the minced meat formulations before thermal treatment was 6.5 to 7.7 units compared to the control (42 units); after thermal treatment, it was 6.5 to 8.0 units for the minced meat formulations, which included vegetable oils with linseed oil. The data on the fatty acid composition of the formulations correspond to the indicators of the fatty acid balance which was RL1…3=0.47 – 0.57 and RL1…6 = 0.32 – 0.37 units for enriched minced meat before thermal treatment and 0.48 – 0.57 and 0.31 – 0.38 units after thermal treatment, respectively. The results confirm the possibility to enrich minced meat formulations with linseed oil when producing meat balls, which can be extended to other types of products.Аннотация В статье рассматривается проблема оптимизации жирно-кислотного состава липидов мяса птицы, широко используемого в питании. В составе липидов мяса птицы содержание омега-6 значительно превышает омега-3, что не оптимально для усвоения и нуждается в коррекции. Исследована возможность обогащения мяса индейки льняным маслом, что позволило обеспечить в фаршевых композициях с растительными маслами соотношение омега-6 : омега-3, обеспечивающих их нутриентно-адекватный баланс, не превышающий 10 единиц. Представлены результаты исследования жирнокислотного состава и жирнокислотной сбалансированности липидной части фаршей из мяса, а также их изменения в результате теплового нагрева мясных композиций в водной среде с температурой 95±2 °C до температуры в продукте 70±1 °C. По данным исследований омега-6:омега-3 в фаршевых композициях до тепловой обработки это отношение составило от 6,5 до 7,7 единиц, по сравнению с контролем (42 единицы); после тепловой обработки - от 6,5 до 8,0 единиц для композиций фаршей, включающих растительные масла в сочетании с льняным маслом. Данные по жирнокислотному составу композиций согласуются со значениями показателей жирнокислотной сбалансированности, составляющими RL1…3=0,47 – 0,57 и RL1…6 = 0,32 - 0,37 единиц для обогащенных фаршей до тепловой обработки и 0,48 - 0,57 и 0,31 – 0,38 единиц после тепловой обработки соответственно, и подтверждают возможность обогащения льняным маслом рецептур фаршей при производстве фрикаделек и могут быть распространены на другие виды продуктов

    FAS-dependent cell death in α-synuclein transgenic oligodendrocyte models of multiple system atrophy

    Multiple system atrophy is a parkinsonian neurodegenerative disorder. It is cytopathologically characterized by accumulation of the protein p25α in cell bodies of oligodendrocytes followed by accumulation of aggregated α-synuclein in so-called glial cytoplasmic inclusions. p25α is a stimulator of α-synuclein aggregation, and coexpression of α-synuclein and p25α in the oligodendroglial OLN-t40-AS cell line causes α-synuclein aggregate-dependent toxicity. In this study, we investigated whether the FAS system is involved in α-synuclein aggregate dependent degeneration in oligodendrocytes and may play a role in multiple system atrophy. Using rat oligodendroglial OLN-t40-AS cells we demonstrate that the cytotoxicity caused by coexpressing α-synuclein and p25α relies on stimulation of the death domain receptor FAS and caspase-8 activation. Using primary oligodendrocytes derived from PLP-α-synuclein transgenic mice we demonstrate that they exist in a sensitized state expressing pro-apoptotic FAS receptor, which makes them sensitive to FAS ligand-mediated apoptosis. Immunoblot analysis shows an increase in FAS in brain extracts from multiple system atrophy cases. Immunohistochemical analysis demonstrated enhanced FAS expression in multiple system atrophy brains notably in oligodendrocytes harboring the earliest stages of glial cytoplasmic inclusion formation. Oligodendroglial FAS expression is an early hallmark of oligodendroglial pathology in multiple system atrophy that mechanistically may be coupled to α-synuclein dependent degeneration and thus represent a potential target for protective intervention

    An environmental assessment of risk in achieving good environmental status to support regional prioritisation of management in Europe

    The Marine Strategy Framework Directive (MSFD) aims to achieve Good Environmental Status (GES) in Europe's Seas. The requirement for regional sea authorities to identify and prioritise issues for management has meant that standardized methods to assess the current level of departure from GES are needed. The methodology presented here provides a means by which existing information describing the status of ecosystem components of a regional sea can be used to determine the effort required to achieve GES. A risk assessment framework was developed to score departure from GES for 10 out of the 11 GES descriptors, based on proposed definitions of 'good' status, and current knowledge of environmental status in each of the four regional seas (North-East Atlantic, Mediterranean Sea, Baltic Sea and Black Sea). This provides an approach for regional evaluation of environmental issues and national prioritisation of conservation objectives. Departure from GES definitions is described as 'high', 'moderate' or low' and the implications for management options and national policy decisions are discussed. While the criteria used in this study were developed specifically for application toward MSFD objectives, with modification the approach could be applied to evaluate other high-level social, economic or environmental objectives.     Class II MHC Self-Antigen Presentation in Human B and T Lymphocytes

    Human CD4[superscript +] T cells process and present functional class II MHC-peptide complexes, but the endogenous peptide repertoire of these non-classical antigen presenting cells remains unknown. We eluted and sequenced HLA-DR-bound self-peptides presented by CD4[superscript +] T cells in order to compare the T cell-derived peptide repertoire to sequences derived from genetically identical B cells. We identified several novel epitopes derived from the T cell-specific proteome, including fragments of CD4 and IL-2. While these data confirm that T cells can present peptides derived from the T-cell specific proteome, the vast majority of peptides sequenced after elution from MHC were derived from the common proteome. From this pool, we identified several identical peptide epitopes in the T and B cell repertoire derived from common endogenous proteins as well as novel endogenous epitopes with promiscuous binding.
